Fluorinated organic mols. are of high interest for many applications across chem. and medical disciplines. Efficient methods for the synthesis of such compounds are thus needed. Within this work, application of the bench-stable cyclic hypervalent iodine(III) fluoro reagent facilitated the development of an efficient, metal-free method for the preparation of the novel class of 4-fluoro-1,3-benzoxazepines starting from readily available styrenes. The efficacy and broad applicability of this concept is demonstrated by the synthesis of 20 structurally diverse congeners in high yields, regio-, and diastereoselectivities. The presented method provides complementary chemoselectivity when compared to the common, com. available electrophilic fluorination reagents, such as selectfluor. First mechanistic investigations with isotopically labeled substrates reveal a complex reaction mechanism, proceeding via an unusual fluorination/1,2-aryl migration/cyclization cascade.

ContextChinese herb Huangqin decoction (HQD) can regulate intestinal flora in ulcerative colitis (UC) mice. Our study clarifies the mechanism of HQD in regulating the intestinal flora of UC mice. C57BL/6 mice were randomly divided into six groups: Control, Model (3% DSS), Sulfasalazine (500 mg/kg), HQD-L (250 mg/kg), HQD-M (500 mg/kg), and HQD-H (1000 mg/kg) groups. Measurement of body weight, colon length, DAI, and haematoxylin-eosin staining were conducted. FISH and 16S rDNA detected colonic bacterial infiltration and intestinal flora changes. The expression of RegIIIγ and PRRs (NOD2, TLR5, TLR4) were detected by FCM and WB, resp. In addition, WB, qPCR, or IHC were used to detect the expression of NOD2, MyD88, RIP2, and NF-κB p65 in the colon. ELISA was used to determine cytokines. Compared with the model group (DAI score, 2.38 ± 0.05; histol. score, 4.08 ± 0.54), HQD treatment significantly reduced the DAI score (L, 2.16 ± 0.09; M, 1.45 ± 0.05; H, 1.18 ± 0.05) and histol. score (L, 3.16 ± 0.82; M, 2.50 ± 0.81; H, 1.51 ± 0.76); restored the weight, the colonic length (p < 0.05). 16S rDNA identification showed HQD regulated the balance of intestinal flora. Moreover, HQD suppressed the expression of RegIIIγ (p < 0.05) and prevented colonic bacterial infiltration. Furthermore, WB results showed NOD2, and TLR4 were inhibited by HQD, especially NOD2 (p < 0.01). The data of WB, qPCR, and IHC demonstrated that the NOD2-dependent pathway was inhibited by HQD (p < 0.01). HQD (1000 mg/kg) regulates the intestinal flora of colitis mice, mainly characterized as inhibition of the NOD2-dependent pathway. These results indicate that HQD has potential. A new class of materials based on crosslinked poly (ester-urethane) containing low proportions of ionic liquid were synthesized using biobased sulfonated oligoester diol, trimethylol propane (TMP), 4,4′-methylene bis(cyclohexyle isocyanate) (HMDI) and 1-butyle-3-methylimidazolium acetate (BMIMAc). Sulfonated oligoester with a well-controlled molar mass was first obtained by melt polycondensation catalyzed by Ti(OBu)4. The resulting products were characterized by, 1HNMR, DSC and MALDI-TOF MS techniques. The systems of crosslinked poly (ester-urethane) and ionic liquid were obtained via a one-shot process. FTIR tech. was used for a more effective control of crosslinking reaction. The Thermomech. anal. of the resulting materials were performed by DSC and DMA technique. Furthermore, the hydrolytic and oxidative degradation study revealed that the degradation of systems mainly depends on the content in ionic liquid and the crosslinking degree of networks.

Paeoniflorin (PF) has been reported to be effective against several skin disorders, such as allergic contact dermatitis and psoriasis; however, it remains unclear whether PF can protect against urticarial lesions. Herein, the effects of PF on rats with urticarial lesions and the possible underlying mechanism were investigated. The effects of PF administration on a rat model of ovalbumin-induced urticarial-like lesions were evaluated via pathol. anal. using hematoxylin-eosin staining. Toluidine blue staining was performed to detect mast cells and ELISA was performed to determine serum histamine levels. PF-induced regulatory effects on autophagic activity and the potential underlying mechanism of this were also investigated using transmission electron microscopy, immunohistochem. and reverse transcription-quant. PCR. It was demonstrated that PF suppressed allergic and inflammatory responses to improve urticarial lesions, as evidenced by the attenuation of pathol. abnormalities, mast cell infiltration and histamine secretion. Mechanistically, PF treatment was found to markedly limit the production and release of inflammatory cytokine interleukin (IL)-23, while the levels of IL-17 remained unchanged. PF intervention led to an increased number of autophagosomes, along with higher levels of light chain 3B (LC3B) and Beclin-1, and lower levels of P62, indicating that PF could augment autophagic activity in urticarial lesions. PF treatment increased the expression of liver kinase B1 (LKB1) and AMP-activated protein kinase-α (AMPK-α), contributing to the PF-enhanced autophagic activity. In conclusion, PF could effectively improve urticarial lesions by inhibiting inflammatory cytokine IL-23 and increasing the autophagic activity via the LKB1/AMPK-α pathway.

The binding of the tridecapeptide yeast mating pheromone, α-factor, to its receptor represents an excellent model for the investigation of peptide hormone-receptor interactions. Preferential acylation of the α- or ε-amine in [Nle12]-α-factor was accomplished using 3-[3,5-diiodo-4-hydroxyphenyl]propanoic acid hydroxysuccinimide ester (diiodo Bolton-Hunter reagent). The product distribution depended on pH, organic cosolvent, and the ratio of organic solvent and aqueous buffer. Product distributions were followed using anal. HPLC and the products were characterized enzymically and by mass spectrometry. Citraconic anhydride preferentially α-acylated [Nle12]-α-factor and served as a temporary masking group during the synthesis of ε-Bolton-Hunter acylated pheromone. Biotin or diiodo Bolton-Hunter reagents were also directly incorporated into [Nle12]-α-factor during peptide synthesis. The biol. activity of the synthetic peptides was characterized using growth arrest assays. These studies provide insightes into the structural requirements for binding of α-factor to its receptor and form a basis for detailed investigation of the receptor using biochem. procedures.

The use of antiretroviral drugs is associated with significant safety concerns but there is still insufficient data about the toxicity profile of antiretroviral therapy (ART) drugs especially in developing countries. Hence, this study was done to describe the severity and pattern of different types of adverse drug reactions that occurs with ART. A retrospective cross-sectional study was done at Pharmacovigilance center Regional Institute of Medical Sciences utilizing data from Jan. 2016 to Dec. 2019. A total of 190 cases reported during the study period were included in this study. Incidence was higher in females (109) as compared to males (81). The most common regimen responsible was Tenofovir/Lamivudine/Efavirenz (TLE) (69.5%) followed by Zidovudine/Lamivudine/Nevirapine (ZLN) (16.3%). Involvement of dermatol. system (27.4%) was most common. The most common Adverse drug reaction (ADR) associated with TLE was skin rash (28.3%) which was less severe as compared to the most common ADR associated with ZLN, which was anemia (40.6%). On evaluation of the World Health Organization-Uppsala Monitoring Center causality of ADRs, majority were found to be possible (78.2%). TLE regimen requires special focus as it was the most common regimen causing ADR but patients on ZLN regimen need to be closely monitored as they were found to cause more serious ADRs. A more active pharmacovigilance is needed for better understanding of toxicities related to ART.
