Supported metal catalysts, for instance, palladium, are one of the foundations of chem. reactions, especially in C-C bond formation. The present study reports preparation of a magnetically separable palladium-supported nano-biocomposite with a low cost and easy immobilization technique. Fibroin, a natural biodegradable polymer, was used through an in situ method to cover the Fe3O4 nanoparticles to make a nano-biocomposite followed by anchoring palladium on the fibroin surface. The morphol. and the structure of palladium-supported nano-biocomposite Fe3O4@fibroin-Pd were characterized by FT-IR, XRD, TGA, SEM, EDX, and TEM techniques. Consequently, the nanocatalyst activity was evaluated in the Heck coupling reactions. Only a very small amount of the nanocatalyst was employed in the reaction, and it showed excellent catalytic activity; in most cases more than 90% efficiency. The significant advantages of employing this nanocatalyst include high catalytic activity, short reaction times, easy separation of the nanocatalyst with an external magnet and great reusability. The results demonstrated that the used nanocatalysts were very active for four consecutive reaction rounds.

Detail spectroscopic characteristic of this compound were performed, and the mechanism of initiation in binary photoinitiating system together with iodine salt was discussed and confirmed by experiments Using real-time FTIR, the usefulness of the proposed meta- and para-cyanoderivatives of 1,4-di(styryl)benzene and 1,3-di(styryl)benzene as highly efficient photosensitizers for various photopolymerization processes was tested, including: free-radical photopolymerization of acrylate monomer or cationic photopolymerization of epoxy and glycidyl monomers. In addition, the formation of interpenetrating polymer networks with the use of proposed two-component initiating systems was performed. Encouraged by excellent polymerization results with the use of the developed initiating systems, addnl. application tests were carried out, including: 3D printing using vat polymerization, visible based direct ink writing, preparation of various fiber-reinforced composite or formation of multiwalled carbon nanotubes composites (MWCNTs nanocomposites), for which the manufacturing process has been analyzed in detail using real-time FTIR method.

Hepatocellular carcinoma (HCC) is one of the most common tumors affecting a large population worldwide, with the fifth and seventh greatest mortality rates among men and women, resp., and the third prime cause of mortality among cancer victims. Di-Me itaconate (DI) has been reported to be efficacious in colorectal cancer by decreasing IL-1β release from intestinal epithelial cells. In this study, diethylnitrosamine (DEN)-induced HCC in male albino Wistar rats was treated with DI as an anticancer drug. The function and mol. mechanism of DI against HCC in vivo were assessed using histopathol., ELISA (ELISA), and Western blot studies. Metabolomics using 1H-NMR was used to investigate metabolic profiles. As per mol. insights, DI has the ability to trigger mitochondrial apoptosis through iNOS- and eNOS-induced activation of the NF-κB/Bcl-2 family of proteins, CytC, caspase-3, and caspase-9 signaling cascade. Serum metabolomics investigations using 1H-NMR revealed that aberrant metabolites in DEN-induced HCC rats were restored to normal following DI therapy. Furthermore, our data revealed that the DI worked as an anti-HCC agent. The anticancer activity of DI was shown to be equivalent to that of the com. chemotherapeutic drug 5-fluorouracil.

Following up the open initiative of anti-malarial drug discovery, a GlaxoSmithKline (GSK) phenotypic screening hit was developed to generate hydroxyethylamine based plasmepsin (Plm) inhibitors exhibiting growth inhibition of the malaria parasite Plasmodium falciparum at nanomolar concentrations Lead optimization studies were performed with the aim of improving Plm inhibition selectivity vs. the related human aspartic protease cathepsin D (Cat D). Optimization studies were performed using Plm IV as a readily accessible model protein, the inhibition of which correlates with anti-malarial activity. Guided by sequence alignment of Plms and Cat D, selectivity-inducing structural motifs were modified in the S3 and S4 sub-pocket occupying substituents of the hydroxyethylamine inhibitors. This resulted in potent anti-malarials with an up to 50-fold Plm IV/Cat D selectivity factor. More detailed investigation of the mechanism of action of the selected compounds revealed that they inhibit maturation of the P. falciparum subtilisin-like protease SUB1, and also inhibit parasite egress from erythrocytes. Our results indicate that the anti-malarial activity of the compounds is linked to inhibition of the SUB1 maturase plasmepsin subtype Plm X.

In the field of polymer chem., tremendous efforts have been made over the last decade to replace petrochem. monomers with building blocks from renewable resources. In this respect, itaconic acid has been used as an alternative to acrylic acid or maleic acid in unsaturated polyesters for thermal or UV-curing applications. However, examples of poly(ester amide)s from itaconic acid are scarce. Under standard polycondensation reactions, the presence of free amines leads to aza-Michael addition reactions at the α,β-unsaturated double bond of the itaconic acid and isomerization reactions to mesaconic acid. Both reactions make the resulting materials useless as UV-curing polymer resins. To avoid these undesired side reactions, we herein report the use of preformed, well-defined diols containing internal amide bonds. The resulting unsaturated poly(ester amide) resins were analyzed before and after UV-induced crosslinking. Viscosity measurements revealed a strong thixotropic behavior induced by the amide groups, which is usually not detected in structurally similar polyester resins.

Docking studies using available crystal structures have been used for structure-based optimization of this series of compounds Within this study, the authors have investigated the role of HDAC8 in the proliferation of cancer cells and optimized hits for potency and selectivity, both in vitro and in cell culture. The combination of structure-based design, synthesis, and in vitro screening to cellular testing resulted in potent and selective HDAC8 inhibitors that showed anti-neuroblastoma activity in cellular testing.

Dental resin composites are commonly used in the restorative management of teeth via adhesive bonding, which has evolved significantly over the past few decades. Although current self-etch bonding systems decrease the number of clin. steps, the acidic functional monomers employed exhibit a limited extent of demineralization of enamel in comparison to phosphoric acid etchants, and the resultant superficial ionic interactions are prone to hydrolysis. This study evaluates the etching of primers constituted with bis[2-(methacryloyloxy) ethyl] phosphate (BMEP) of dental hard tissue, interfacial characteristics, and inhibition of endogenous enzymes. We examine the incorporation of 2 concentrations of BMEP in the formulation of exptl. primers used with a hydrophobic adhesive to constitute a 2-step self-etching bonding system and compare to a com. 10-methacryloyloxydecyl dihydrogen phosphate (10-MDP)-containing system. The interaction of the primer with enamel and dentin was characterized using scanning electron, confocal laser scanning, and Raman microscopy while the polymerization reaction between the BMEP primers and hydroxyapatite was evaluated by Fourier-transform IR spectroscopy. The inhibitory effect against matrix metalloproteinase (MMP) enzymes of these primers was studied and percentage of inhibition analyzed using 1-way anal. of variance and Tukey′s post hoc test (P < 0.05). Results of the SEM micrographs demonstrated potent etching of both enamel and dentin with the formation of longer resin tags with BMEP primers compared to the 10-MDP-based system. The BMEP polymerized on interaction with pure hydroxyapatite in the dark, while the 10-MDP primer exhibited the formation of salts. Furthermore, BMEP primers were able to inhibit MMP activity in a dose-dependent manner. BMEP could be used as a self-etching primer on enamel and dentin, and the high d.p. in the presence of hydroxyapatite can contribute to an increased quality of the resin polymer network, prompting resistance to gelatinolytic and collagenolytic degradation

A novel way was adopted to graft zinc oxide (ZnO) with urethane-modified dimethacrylate (UDMA) in order to utilize them as reinforcing agents in resin-based dental composites. Exptl. novel composites were synthesized having UDMA-grafted and nongrafted ZnO, at a concentration of 0 weight%, 5 weight%, and 10 weight%. The same concentrations of ZnO were also incorporated in Filtek Z250 XT (3 M ESPE, USA). The antibacterial behavior was evaluated against Streptococcus mutans by direct-contact test at one, three, and seven days of incubation. The compressive strength and Vickers microhardness were tested as per ISO 9917 and ISO/CD6507-1, resp. For abrasive wear resistance, mass loss and roughness average after tooth-brushing cycles of 24,000 at custom-made tooth-brushing simulator were evaluated using noncontact profilometer. Data anal. was carried out using post hoc Tucky′ s test and nonparametric Kruskal-Wallis test. Direct contact test revealed that the antibacterial potential of novel and com. composites was increased with an increase in the concentration of grafted ZnO as compared with nongrafted, whereby the potential was the highest at day seven. There was a significant decrease in compressive strength and Vickers hardness of com. composites on addition of grafted ZnO while there was no significant difference in the strength of exptl. novel composite. The abrasive wear of com. and exptl. composites was within clin. limits. Low-temperature flow-synthesis method was successfully employed to synthesize grafted and nongrafted ZnO. The UDMA-grafted ZnO can be incorporated into dental composites without decreasing their strength and these composites can be used to combat secondary caries.
