Wintersweet (Chimonanthus praecox L.) is an important ornamental plant in China with a pleasant floral scent. To explore the potential mechanisms underlying differences in the fragrances among genotypes of this plant, we analyzed floral volatile organic compounds (VOCs) from two different genotypes: SW001, which has little to no fragrance, and the scented genotype H29. The major VOCs in H29 were linalool, trans-β-ocimene, benzyl acetate, Me salicylate, benzyl alc. (BAlc) and Me benzoate. The most important aroma-active compound in H29, linalool, was emitted at a low concentration in SW001, which had markedly higher levels of trans-β-ocimene than H29. Next, to investigate scent biosynthesis, we analyzed the transcriptome and proteome of fully open flowers of the two genotypes. A total of 14 443 differentially expressed unigenes and 196 differentially expressed proteins were identified. Further analyses indicated that 56 differentially expressed genes involved in the terpenoid and benzenoid biosynthesis pathways might play critical roles in regulating floral fragrance difference. Disequilibrium expression of four terpene synthase genes resulted in diverse emission of linalool and trans-β-ocimene in both genotypes. In addition, the expressions of two CpMYC2 transcription factors were both upregulated in H29, implying that they may regulate linalool production Notably, 16 of 20 genes in the benzenoid biosynthesis pathway were downregulated, corresponding to the relatively low level of benzenoid production in SW001. The lack of benzyl acetate might indicate that SW001 may lack substrate BAlc or functional acetyl-CoA:benzylalc. acetyltransferase.

Prunus mume is the only fragrant flowering species of Prunus. According to the previous studies, benzyl acetate and eugenol dominate its floral scent. However, the diversity of its floral scents remains to be elucidated. In this work, the floral volatiles emitted from eight intraspecific cultivars of P. mume with white, pink and red flowers, were collected and analyzed using headspace solid-phase microextraction combined with gas chromatograms-mass spectrometry (HS-SPME-GC-MS). In total, 31 volatile compounds were identified, in which phenylpropanoids/benzenoids accounted for over 95% of the total emission amounts Surprisingly, except for benzyl acetate and eugenol, several novel components, such as benzyl alc., cinnamyl acohol, cinnamy acetate, and benzyl benzoate were found in some cultivars. The composition of floral volatiles in cultivars with white flowers was similar, in which benzyl acetate was dominant, while within pink flowers, there were differences of floral volatile compositions Principal component anal. (PCA) showed that the emissions of benzyl alc., cinnamyl alc., benzyl acetate, eugenol, cinnamyl acetate, and benzyl benzoate could make these intraspecific cultivars distinguishable from each other. The combined utilization of waste cooking oil (WCO) and reclaimed asphalt pavement (RAP) is attractive for economic, environmental, and energy benefits. However, the quality stability of WCO is poor due to its wide sources, which limits the further development of WCO recycled RAP. In this study, the classification method of WCO was proposed according to the mol. weight difference of its chem. components, and WCO was classified into three categories including light components (LC), intermediate components (IC), and heavy components (HC). Meanwhile, the characteristics of these three WCO components and their effects on the physicochem. properties of aging asphalt were investigated comprehensively. The exptl. results show that the softening ability of different components of WCO on aging asphalt is mainly determined by their mol. weight and thermal stability. Furthermore, LC rich in Me palmitate is easy solidification and volume expansion at the low temperature, which is not conducive to the repair of low-temperature ductility of aged asphalt. The chem. property analyses of recycled asphalt indicate that there is no chem. reaction between the components of WCO and the aging asphalt. The phys. dilution ability of WCO components on carbonyl index, sulfoxide index, and high mol. weight substances of aging asphalt is pos. correlated with their mol. weight In addition, HC contains chem. substances similar in composition to asphalt, which has the most obvious effect on the colloidal structure of aging asphalt. Overall, the classification method for WCO and related findings found in this study provides a reference for ensuring the quality stability of WCO and its industrial application in the recycling of aged asphalt.

BACKGROUND : The natural 1,8-dihydroxynaphthalene (DHN) melanin biosynthesis inhibitors (MBIs) are one of the promising approaches to the integrated management of plant diseases but have received scarce attention until now. Herein, to explore the natural DHN MBIs used in the control of northern corn leaf blight (NCLB), a library of 53 essential oil compounds was used to screen the MBIs against Exserohilum turcicum, the causal pathogen of NCLB, using tricyclazole as a reference compound RESULTS : The results of morphol. change in the colony, thermogravimetric anal., UV-visible spectroscopy, and transmission electron microscopy confirmed that guaiacol could effectively inhibit the melanin production at 50 μg/mL under in vitro conditions. The in vitro bioassay results indicated that this inhibition effect was concentration-dependent and the min. inhibition concentration of guaiacol was 50 μg/mL. The in vivo exptl. results demonstrated that guaiacol significantly inhibited appressorium formation and penetration on corn leaf sheaths at the concentration of 500 μg/mL. The pot experiment results revealed that there were no differences between guaiacol (500 μg/mL) and tricyclazole (100 μg/mL) in control efficacy. The enzymic assay suggested that guaiacol might exert the activity through inhibiting DHN polymerization to form melanins, which was distinct from tricyclazole. CONCLUSIONS : Taken together, this study screened out guaiacol as a natural MBI from 53 essential oil compounds and verified its effectiveness in the control of NCLB at 500 μg/mL. Above all, this research opened an avenue for exploring natural DHN MBIs in the integrated management of plant diseases. 2022 Society of Chem. Industry.

A total of 94 volatile organic compounds (VOCs) were detected in the fresh olive pomace, with a prevalence of lipoxygenase (LOX) VOCs (78%), mainly aldehydes (19 490.9μg kg-1) despite the higher number of alcs. A decrease in LOX volatiles and a quick development of the ones linked to off-flavors (carboxylic acids, alcs., acetates) were observed, in particular after 4 days of storage at room temperature Only storage at 4 °C allowed these phenomena to be slowed down. Zaire ebolavirus (EBOV) causes a severe hemorrhagic fever in humans and non-human primates with high morbidity and mortality. EBOV infection is dependent on its structural glycoprotein (GP), but high levels of GP expression also trigger cell rounding, detachment, and downregulation of many surface mols. that is thought to contribute to its high pathogenicity. Thus, EBOV has evolved an RNA editing mechanism to reduce its GP expression and increase its fitness. We now report that the GP expression is also suppressed at the protein level in cells by protein disulfide isomerases (PDIs). Although PDIs promote oxidative protein folding by catalyzing correct disulfide formation in the endoplasmic reticulum (ER), PDIA3/ERp57 adversely triggered the GP misfolding by targeting GP cysteine residues and activated the unfolded protein response (UPR). Abnormally folded GP was targeted by ER-associated protein degradation (ERAD) machinery and, unexpectedly, was degraded via the macroautophagy/autophagylysosomal pathway, but not the proteasomal pathway. PDIA3 also decreased the GP expression from other ebolavirus species but increased the GP expression from Marburg virus (MARV), which is consistent with the observation that MARV-GP does not cause cell rounding and detachment, and MARV does not regulate its GP expression via RNA editing during infection. Furthermore, five other PDIs also had a similar inhibitory activity to EBOV-GP. Thus, PDIs neg. regulate ebolavirus glycoprotein expression, which balances the viral life cycle by maximizing their infection but minimizing their cellular effect. We suggest that ebolaviruses hijack the host protein folding and ERAD machinery to increase their fitness via reticulophagy during infection.
