A novel “”symbiotic self-assembly”” strategy that integrates the advantages of biocompatible lipids with a structurally robust polymer to efficiently encapsulate and deliver siRNAs is reported. The assembly process is considered to be symbiotic because none of the assembling components are capable of self-assembly but can form well-defined nanostructures in the presence of others. The conditions of the self-assembly process are simple but have been chosen such that it offers the ability to arrive at a system that is noncationic for mitigating carrier-based cytotoxicity, efficiently encapsulate siRNA to minimize cargo loss, be effectively camouflaged to protect the siRNA from nuclease degradation, and efficiently escape the endosome to cause gene knockdown. The lipid-siRNA-polymer (L-siP) nanoassembly formation and its disassembly in the presence of an intracellular trigger have been extensively characterized exptl. and through computational modeling. The complexes have been evaluated for the delivery of four different siRNA mols. in six different cell lines, where an efficient gene knockdown is demonstrated. The reported generalized strategy has the potential to make an impact on the development of a safe and effective delivery agent for RNAi-mediated gene therapy that holds the promise of targeting several hard-to-cure diseases.

In previous studies, the authors investigated the change in the composition of basic fatty acids (FA) of sunflower oil during heating or long-term storage, but there are no studies of the full FA composition of sunflower oil, as well as an assessment of its dependence on the period of the oil using. The aim. Detailed study of the FA composition of refined sunflower oil from different manufacturers, assessment of the effect of the shelf life of the oil on its FA composition and its compliance with the requirements of the European Pharmacopoeia (EP). Materials and methods. The determination of the FA composition of seven samples of refined sunflower oil was carried out by gas chromatog. with a flame ionization detector using the Rt-2560 column (100 m x 0.25 mm x 0.20μm). Results. The anal. of seven samples of refined sunflower oil showed the presence of 24 FA in most of them. Polyunsaturated FA predominate among them with a percentage of 52.45-60.86%. The value of the fatty acids percentage in each groups (saturated, mono- and polyunsaturated fatty acids) are quite similar regardless of the oil shelf life at the time of the research. All test samples, except one, by the percentage content of the main FA correspond to EP requirements. The results of the quant. content of the main FA of sunflower oil determining (palmitic, stearic, oleic, and linoleic) also indicate a similar FA composition of the studied samples. Conclusions. The studied samples of sunflower oil refined in terms of the content of basic FAs meet the requirements of the EP. The obtained results indicate the high stability of the oil and the absence of a significant effect of the storage period on the FA composition

BACKGROUND : Ozone treatment can effectively inhibit fruit decay in many fruits during postharvest storage. However, little information is available for pitaya fruit. RESULTS : Ozone treatment significantly reduced the decay rate and induced the enzyme activities of peroxidase and polyphenol oxidase, and also reduced the levels of reactive oxygen species. In total, 103 metabolites were detected and changed the content after ozone treatment, including 54 primary metabolites and 49 aromatic compounds After significance and importance anal., 37 metabolites were important. Some metabolites were induced by peel senescence to respond to senescence stress, including D-fructose, D-glucose, mannose, inositol, galactonic acid, ethanedioic acid and stearic acid. Some metabolic products of peel senescence were reduced by ozone treatment, including D-arabinose, glucaric acid, galacturonic acid, 1-hexanol, 4-ethylcyclohexanol, β-linalool, palmitoleic acid and 2-hydroxy-cyclopentadecanone. Some metabolites induced by ozone treatment might play a vital role in delaying the senescence and decay, including malic acid, succinic acid, pentenoic acid, eicosanoic acid, 2-hexenal, hexanal, 2-heptenal, 4-heptenal, 2-octenal and nitro m-xylene. CONCLUSION : Ozone treatment significantly reduced decay and prolonged shelf-life without reducing fruit quality. In total, 37 metabolites might play an important role in ozone delayed fruit decay. © 2018 Society of Chem. Industry.

BACKGROUND : Bottle storage can affect color, aroma and phenolic composition of white wine. Very little information is reported about the bottle evolution during storage of white wines without added sulfites and/or using other antioxidants. This work is aimed at studying the evolution of the main enol. parameters, phenolic and volatile profiles of a white wine without added sulfites, during 15 mo of storage in bottle at different light and temperature conditions, compared with a control stored with sulfur dioxide. Dark storage at 12°C (D12-S, D12-NS) and 30°C (D30-S, D30-NS) were compared to investigate the temperature effect, meanwhile uncontrolled temperature and light condition (UTL-S, UTL-NS) simulated improper storage conditions. RESULTS : Volatile acidity, absorbance at 420 nm and total phenols were higher in UTL-NS and D30-NS wines. The trans forms of hydroxycinnamic esters decreased, whereas, the cis forms, as well as caffeic acid derivatives, significantly increased in samples without added sulfites. The storage without sulfites also accelerated the hydrolysis of acetate esters. However, it did not affect most of the Et esters whose content remained almost the same between sulfite added (S) and no sulfite added (NS) wines. CONCLUSION : The presence of sulfites in the bottle helped to preserve the volatile compounds of young wines; however, even more important seemed to be the optimal storage (dark and low temperature), as unsuitable conditions favored aroma degradation of bottled wine, regardless of sulfite protective action. © 2019 Society of Chem. Rift Valley fever virus (RVFV) is a zoonotic life-threatening viral infection endemic across sub-Saharan African countries and the Arabian Peninsula; however, there is a growing panic of its spread to non-endemic regions. This viral infection triggers a wide spectrum of symptoms that span from fibril illnesses to more severe symptoms such as haemorrhagic fever and encephalitis. These severe symptoms have been associated with dysregulated immune response propagated by the virulence factor, non-structural protein (NSs). Thus, this study investigates the effects of lithium on NF-κB translocation and RFVF-induced inflammation in Raw 264.7 macrophages. The supernatant from RVFV-infected Raw 264.7 cells, treated with lithium, was examined using an ELISA assay kit to measure levels of cytokines and chemokines. The H2DCF-DA and DAF-2 DA florigenic assays were used to determine the levels of ROS and RNS by measuring the cellular fluorescence intensity post RVFV-infection and lithium treatment. Western blot and immunocytochem. assays were used to measure expression levels of the inflammatory proteins and cellular location of the NF-κB, resp. Lithium was shown to stimulate interferon-gamma (IFN-γ) production as early as 3 h pi. Production of the secondary pro-inflammatory cytokine and chemokine, interleukin-6 (IL-6) and regulated on activation, normal T cell expressed and secreted (RANTES), were elevated as early as 12 h pi. Treatment with lithium stimulated increase of production of tumor necrosis factor-alpha (TNF-α) and Interleukin-10 (IL-10) in RVFV-infected and uninfected macrophages as early as 3 h pi. The RVFV-infected cells treated with lithium displayed lower ROS and RNS production as opposed to lithium-free RVFV-infected control cells. Western blot analyses demonstrated that lithium inhibited iNOS expression while stimulating expression of heme oxygenase (HO) and IκB in RVFV-infected Raw 264.7 macrophages. Results from immunocytochem. and Western blot assays revealed that lithium inhibits NF-κB nuclear translocation in RVFV-infected cells compared to lithium-free RVFV-infected cells and 5 mg/mL LPS controls. This study demonstrates that lithium inhibits NF-kB nuclear translocation and modulate inflammation profiles in RVFV-infected Raw 264.7 macrophage cells.

Herein, we developed selective coupling and polymerization systems of folded polymer micelles via phys. interaction in water. The polymer micelles serve as nanodomains to provide double core micelles, alternating necklace micelles, and micelle-connected hydrogels. For this, cation- or anion-tail unimer micelles and amine- or carboxy-tail unimer micelles were designed; the unimer micelles consist of folded amphiphilic random copolymers carrying hydrophilic poly(ethylene glycol) and hydrophobic or hydrogen-bonding pendants. Mixing a cation-tail micelle and an anion-tail micelle, and even the combination of a double cation-tail micelle and a double anion-tail micelle, selectively provided double-core micelles in water without forming large aggregates. Double core micelles afforded structural transformation into linear or cyclic polymers and dynamic exchange of the micelle domains. In contrast, mixing amine-tail micelles and carboxy-tail micelles gave an alternating necklace micelle or a hydrogel. The controlled connection of polymer micelles was achieved by designing suitable phys. interaction. This technique opened new ways to create various nanodomain self-assemblies with controlled higher-order structure.

Based on the host vinyl-ether liquid crystals, the polymer dispersed vinyl-ether liquid crystals (PDVLC), polymer dispersed crosslinked vinyl-ether liquid crystals (PDCVLC) and fluorine-containing polymer dispersed crosslinked vinyl-ether liquid crystals (F-PDCVLC) QDs films were fabricated in succession by dual-step polymerization As a result, the PDVLC film can realize fluorescence enhancement and tuning, whereas the PDCVLC film not only maintain fluorescence enhancement of PDVLC film but also improve the water-resistant performance. More importantly, the formation of crosslinked film makes it convenient to fabricate QDs film. Subsequently, the F-PDCVLC film can further improve the water-resistant properties by introducing fluorine-containing monomers to enhance the hydrophobic performance. This free-standing (F-) PDCVLC film may offer a new avenue to realize the optimization of QDs film performance.

The results from in-field oviposition preference of black fig fly, olfactory anal., and fruit volatile profiles indicate that the black fig fly gave a strong preference to unpollinated figs that showed higher emissions of volatile organic compounds Terpenes are known to be important compounds determining many insect-plant interactions, so we report a transcriptome-based identification and functional characterization of a terpene synthase (TPS) gene family in F. carica. The protein expression in Escherichia coli of eight terpene synthases (TPSs) revealed that three were monoterpene synthases belonging to the TPS-b clade, with FcTPS6 catalyzing the formation of 1,8-cineole while the other two converted GPP into linalool. Four sesquiterpene synthases from the TPS-a clade catalyze the formation of germacrene D (FcTPS1), E-β-caryophyllene (FcTPS2), cadinene (FcTPS3) and δ-elemene (FcTPS5) while one sesquiterpene synthase FcTPS4 from the TPS-b clade showed nerolidol synthase activity. This study provides new insights into fig-insect interactions and understanding the mol. mechanisms of terpene biosynthesis and could provide the foundations for sustainable pest management strategies.
