Retinal gene therapy has had unprecedented success in generating treatments that can halt vision loss. However, immunogenic response and long-term toxicity with the use of viral vectors remain a concern. Non-viral vectors are relatively non-immunogenic, scalable platforms that have had limited success with DNA delivery to the eye. MRNA (mRNA) therapeutics has expanded the ability to achieve high gene expression while eliminating unintended genomic integration or the need to cross the restrictive nuclear barrier. Lipid-based nanoparticles (LNPs) remain at the forefront of potent delivery vectors for nucleic acids. Herein, we tested eleven different LNP variants for their ability to deliver mRNA to the back of the eye. LNPs that contained ionizable lipids with low pKa and unsaturated hydrocarbon chains showed the highest amount of reporter gene transfection in the retina. The kinetics of gene expression showed a rapid onset (within 4 h) that persisted for 96 h. The gene delivery was cell-type specific with majority of the expression in the retinal pigmented epithelium (RPE) and limited expression in the Muller glia. LNP-delivered mRNA can be used to treat monogenic retinal degenerative disorders of the RPE. The transient nature of mRNA-based therapeutics makes it desirable for applications that are directed towards retinal reprogramming or genome editing. Overall, non-viral delivery of RNA therapeutics to diverse cell types within the retina can provide transformative new approaches to prevent blindness. 4-Acetamidophenyl 2-acetoxybenzoate (cas: 5003-48-5) belongs to esters. Esters are also usually derived from carboxylic acids. It may also be obtained by reaction of acid anhydride or acid halides with alcohols or by the reaction of salts of carboxylic acids with alkyl halides. Many esters have the potential for conformational isomerism, but they tend to adopt an s-cis (or Z) conformation rather than the s-trans (or E) alternative, due to a combination of hyperconjugation and dipole minimization effects. The preference for the Z conformation is influenced by the nature of the substituents and solvent, if present. Synthesis, spectral properties and thermal behaviour of zinc(II) acetylsalicylate was written by Lambi, John N.;Nsehyuka, Alfred T.;Egbewatt, Nkongho;Cafferata, Lazaro F. R.;Arvia, Alejandro J.. The thermal behavior of Zn(II) acetylsalicylate [Zn(acsa)2(H2O)2] with respect to phase transitions, pyrolysis both in air and inert (N2) atm., and product identification was studied. The complex was synthesized by metathesis in hot EtOH solution using aspirin (acetylsalicylic acid) as precursor and characterized via electronic and IR spectral analyses. Optical observations showed that the white salt does not undergo a direct transition from the solid to the liquid phase but rather goes slowly through an intermediate mesophase around 80° before melting rapidly to the brick-brown isotropic liquid around 134-136°. No liquid crystalline phases are however formed. This result was complemented by that from thermogravimetric (TG) studies in the ∼25-600° range, which showed three main weight-loss phases of 8.0, 50.0 and 14.0% (around 200, 250 and 400°) corresponding, resp., to the elimination of CO2, xanthone and HOAc. The pyrolysis products, as identified using a combination of instrumental (GC-MS) and wet chem. techniques are: CO2; nonstoichiometric Zn oxide, most likely in the form: Zn1+xO (0.0000 ≤ x ≤ 0.0003); and a mixture of organic products resulting from further decomposition, charring and other attendant thermal effects at the relatively high temperatures (∼600°) involved. Six of the principal organic products were identified and included salsalate and benorylate which are pro-drugs of salicylic acid, a known pharmaceutical. To investigate the differences in the major aroma components between medium- and late-maturing Yantai apple cultivars, headspace solid phase microextraction (HS-SPME) combined with gas chromatog. mass spectrometry (GC-MS) was employed. The apple cultivars and their aroma components were analyzed using principal component anal. (PCA) and cluster anal. (CA). The results showed that 59 aroma compounds were identified from 11 apple cultivars, and 21 of these were common to all apple cultivars. Among the common aroma components, acetate esters, butyrate esters, hexanoate esters, hexanal (including 2-hexenal), and some higher alcs. showed higher concentrations The PCA results indicated that the comprehensive scores of Yang Guang and Dan Xia apples were the highest, while that of the Extremely Precocious Fuji apple was the lowest. The comprehensive score ranking reflected the differences of the major aroma components between the apple cultivars. The CA results showed that 11 apple cultivars could be classified into four clusters. Yantai Fuji 3, Dou Nan, Gan Hong, Extremely Precocious Fuji, Pinova, Yantai Fuji 1, and Best Short Fuji were classified into a cluster; Dan Xia and Yang Guang were classified into a cluster; and Liang Xiang and Hua Shuai were classified into two clusters. Through an effective combination of PCA and CA, the aroma characteristics of four apple cultivars (Dan Xia, Yang Guang, Liang Xiang, and Hua Shuai) were more prominent, providing a basis for parent selection in future apple breeding. And the article was included in Molecules in 2020.Application In Synthesis of Isopentyl hexanoate The following contents are mentioned in the article:

The present work deals with the characterization of volatile organic compounds in wines from the Slovak Tokaj wine region. Studied wine samples were divided into 3 groups-varietal wines from registered Tokaj vine varieties, film wines Tokajske samorodne dry, and naturally sweet botrytized wines Tokaj selections. The VOCs from wines were extracted using optimized solid phase microextraction and analyzed by comprehensive two-dimensional gas chromatog. coupled to high-resolution time-of-flight mass spectrometry (HRTOF-MS). In total, 176 VOCs were identified in all 46 studied samples. It was found that the total number of VOCs in varietal wines was generally higher than in botrytized wines. All three studied categories showed characteristic VOC profiles with significant differences. Varietal wines were characterized by higher concentrations of esters and terpenoids originating from grapes. The presence of γ-octalactone, (E)-6-methylhept-2-en-4-one, and lack of benzaldehyde were typical for Tokajske samorodne dry. Tokaj selections expressed the highest concentration of di-Et malate, benzaldehyde, and furfurals. Several interesting trends were also observed The concentration of fermentation products was highest in varietal wines, while long-term matured Tokaj special wines were typified by the presence of compounds related to noble-rotten raisins (2-phenylacetaldehyde, Et 2-phenylacetate, and 2-phenylethanol), wood (cis-whisky lactone), and aging (1,1,6-trimethyl-2H-naphthalene, furfural, and 5-methylfurfural). The disruptive effect of organic solvents on microbial membranes represents a significant challenge to the economical production of green fuels and value-added chems. from lignocellulosic feedstocks. One route to overcoming this challenge is to engineer microbes with membranes capable of resisting organic solvent stresses. In this regard, it is useful to understand the mechanisms by which organic solvents disrupt typical biomembranes. Here, mol. dynamics (MD) simulation, complemented by small-angle X-ray and neutron scattering (SANS/SAXS), provide a mol.-scale view of the disruption of a microbial model membrane by 1-butanol and THF (THF), two common water-organic cosolvent mixtures of importance in biofuel production Solvent interactions at the interface between the head-group and fatty acid tail regions lead to more dramatic membrane changes than interactions solely at the head-groups or tails. Although both organic solvents are found to partition into the membrane, the depth of solvent penetration into the membrane is quite different. Specifically, 1-butanol localizes near the interface between the lipid heads and tails at low concentrations, but partitions into both the head and tail regions at high concentrations In contrast, THF, overall, partitions less than 1-butanol and prefers the lipid tail regions. Importantly, the presence of 1-butanol near the head/tail interface introduces drastic membrane changes not seen with THF. The organic solvent interactions with the lipids lead to membrane thinning. Anthocyanins-based colorimetric volatile amines sensors have recently received great attention for monitoring meat and fish freshness. However, most of the sensors suffered from anthocyanins leaching problems in humid packaging environments, severely limiting their indication ability. The present study developed a hydrogel-in-oleogel bigel to protect anthocyanins from leaching. The internal hydrogel was composed of water, agar and purple sweet potato anthocyanins (PSPA), and the external oleogel was made up of sunflower oil, beeswax and glyceride monooleate (GMO). The agar concentration and water-to-oil-phase volume ratio (W/O) were optimized to be 1.0% and 2:8, resp. The optimal pH value of hydrogel was determined to be 6.50, taking into consideration of the gas sensitivity and color stability of the bigel. The bigel was extruded onto polyvinylidene fluoride (PVDF) film through a 3D printing method to form the composite film. The results of the study revealed that PVDF-bigel composite film showed good stability in water, indicative of strong anti-leaching properties. Meanwhile, the PVDF-bigel film was sensitive to trimethylamine (TMA) with a limit of detection of 2.52μM. The PVDF-bigel film was subjected to in-situ and real-time monitor beef and salmon freshness. The film exhibited a red-to-purple color change upon reacting with volatile amines generated from beef and salmon. Henceforth, this study presented a new method to develop anthocyanins-based colorimetric gas sensors for intelligent food packaging. Depolarizing sodium (Na+) leak currents carried by the NALCN channel regulate the resting membrane potential of many neurons to modulate respiration, circadian rhythm, locomotion and pain sensitivity1-8. NALCN requires FAM155A, UNC79 and UNC80 to function, but the role of these auxiliary subunits is not understood3,7,9-12. NALCN, UNC79 and UNC80 are essential in rodents2,9,13, and mutations in human NALCN and UNC80 cause severe developmental and neurol. disease14,15. Here we determined the structure of the NALCN channelosome, an approx. 1-MDa complex, as fundamental aspects about the composition, assembly and gating of this channelosome remain obscure. UNC79 and UNC80 are massive HEAT-repeat proteins that form an intertwined anti-parallel superhelical assembly, which docks intracellularly onto the NALCN-FAM155A pore-forming subcomplex. Calmodulin copurifies bound to the carboxy-terminal domain of NALCN, identifying this region as a putative modulatory hub. Single-channel analyses uncovered a low open probability for the wild-type complex, highlighting the tightly closed S6 gate in the structure, and providing a basis to interpret the altered gating properties of disease-causing variants. Key constraints between the UNC79-UNC80 subcomplex and the NALCN DI-DII and DII-DIII linkers were identified, leading to a model of channelosome gating. Our results provide a structural blueprint to understand the physiol. of the NALCN channelosome and a template for drug discovery to modulate the resting membrane potential. Precise protein sequencing and folding are believed to generate the structure and chem. diversity of natural channels1,2, both of which are essential to synthetically achieve proton transport performance comparable to that seen in natural systems. Geometrically defined channels have been fabricated using peptides, DNAs, carbon nanotubes, sequence-defined polymers and organic frameworks3-13. However, none of these channels rivals the performance observed in their natural counterparts. Here we show that without forming an atomically structured channel, four-monomer-based random heteropolymers (RHPs)14 can mimic membrane proteins and exhibit selective proton transport across lipid bilayers at a rate similar to those of natural proton channels. Statistical control over the monomer distribution in an RHP leads to segmental heterogeneity in hydrophobicity, which facilitates the insertion of single RHPs into the lipid bilayers. It also results in bilayer-spanning segments containing polar monomers that promote the formation of hydrogen-bonded chains15,16 for proton transport. Our study demonstrates the importance of the adaptability that is enabled by statistical similarity among RHP chains and of the modularity provided by the chem. diversity of monomers, to achieve uniform behavior in heterogeneous systems. Our results also validate statistical randomness as an unexplored approach to realize protein-like behavior at the single-polymer-chain level in a predictable manner. To develop an iron-based solid lipid nanoparticle (SLN) absorbable by the intestinal wall and assess the differential diagnostic value of intestinal lesions in magnetic resonance imaging (MRI). SLNs were prepared with the simultaneous loading of trivalent Fe ions (Fe3+), levodopa Me ester (DM), and fluorescein isothiocyanate (FITC). We evaluated the particle size, loading rate, encapsulation efficiency, and cytotoxicity of SLNs. The T1 contrast effects of the FeDM-FITC-SLNs and gadolinium-based contrast agent (GBCA) were compared in different mouse models: acute ulcerative colitis (AUC), chronic ulcerative colitis (CUC), colon adenocarcinoma (COAD), and normal control. MRI was performed in the same mouse with i.v. injection of GBCA on day 1 and enema of FeDM-FITC-SLNs on day 2. The signal-to-noise ratios (SNRs) were compared using one-way anal. of variance. Tissues were then collected for histol. The average particle size of FeDM-FITC-SLN was 220 nm. The mean FeDM loading rate was 94.3%, and the encapsulation efficiency was 60.3%. The relaxivity was 4.02 mM-1·s-1. After enema with FeDM-FITC-SLNs, MRI showed the following contrast enhancement duration: AUC = COAD > normal > CUC. Confocal fluorescence microscopy confirmed that FeDM-FITC-SLNs were mainly distributed in the intestinal mucosa and tumor capsule. Iron-based SLNs are promising alternatives for contrast enhancement at T1-weighted MRI and will help in the differential diagnosis of intestinal bowel diseases (IBDs).
