Com. sparkling wine production represents a relatively low but important part of the Croatian wine production, especially in the Zagreb county. This study presents the results of volatile aroma compounds profile and organic acid composition of com. sparkling wine samples from three vine-growing regions in Zagreb county. In total, 174 volatile aroma compounds were identified, separated between their chem. classes (aldehydes, higher alcs., volatile phenols, terpenes, C13-norisoprenoids, lactones, esters, fatty acids, sulfur compounds, other compounds, other alcs.). Higher alcs. such as phenylethyl and isoamyl alc. as well as 2-methyl-1-butanol, and esters such as di-Et succinate, Et hydrogensuccinate, and Et lactate had the strongest impact on the volatile compounds profile of Zagreb county sparkling wine. The presence of di-Et glutarate and di-Et malonate, compounds whose concentrations are influenced by yeast autolysis or caused by chem. esterification during the ageing process, was also noted. The influence of every single volatile aroma compound was evaluated by discriminant anal. using forward stepwise model. The volatile profiles of traditional sparkling wines from Croatia were presented for the first time. A convenient method for the preparation of polybasic ternary hybrid cryogels consisting of Halloysite nanotubes (HNTs) and tertiary amine functional groups by freezing-induced gelation is proposed. Ternary hybrid gels were produced via one-shot radical terpolymn. of 2-hydroxyethyl methacrylate (HEMA), 2-acrylamido-2-methyl-1-propane sulfonic acid (AMPS), and DEAEMA in the presence of HNTs. The equilibrium swelling in various swelling media and the mech. properties of the produced ternary hybrid gels were analyzed to investigate their network structure and determine their final performance. The swelling ratio of HNT-free gels was significantly higher than the ternary hybrid gels composed of high amount of HNTs. The addition of HNTs to terpolymer network did not suppress pH- and temperature-sensitive behavior. While DEAEMA groups were effective for pH-sensitive swelling, it was determined that both HEMA and DEAEMA groups were effective in temperature-sensitive swelling. Ternary hybrid gels simultaneously demonstrated both neg. and pos. temperature-responsive swelling behavior. The swelling ratio changed considerably according to swelling temperature Both DEAEMA and HEMA monomers in terpolymer structure were dominant in temperature-sensitive swelling. Mech. tests in compression of both as-prepared and swollen-state demonstrated that strength and modulus of hybrid cryogels significantly increased with addition of HNTs without significant loss of mech. strength. On the deposition of CQDs on the copolymer, the penetration power and fluorescent property have been improved, which help to track the cells in vitro. This film is worthy to be applied to the skin. It can efficiently load lidocaine hydrochloride monohydrate (LHM). In vitro and ex vivo skin permeation profiles reveal the sustained release behavior of loaded LHM at average skin temperature and pH. In vivo soluble α-synuclein occurs as a monomer and several multimers, the latter of which may be important for the biol. function of α-synuclein. Currently, there is a lack of reproducible methods to compare α-synuclein multimer abundance between complex biol. samples. Here we developed a method, termed “multimer-PAGE,” that combines in-gel chem. crosslinking with several common electrophoretic techniques to measure the stoichiometry of soluble α-synuclein multimers in brain tissue lysates. Results show that soluble α-synuclein from the rat brain exists as several high mol. weight species of approx. 56 kDa (αS56), 80 kDa (αS80), and 100 kDa (αS100) that comigrate with endogenous lipids, detergents, and/or micelles during blue native gel electrophoresis (BN-PAGE). Co-extraction of endogenous lipids with α-synuclein was essential for the detection of soluble α-synuclein multimers. Homogenization of brain tissue in small buffer volumes (>50 mg tissue per 1 mL buffer) increased relative lipid extraction and subsequently resulted in abundant soluble multimer detection via multimer-PAGE. α-Synuclein multimers captured by directly crosslinking soluble lysates resembled those observed following multimer-PAGE. The ratio of multimer (αS80) to monomer (αS17) increased linearly with protein input into multimer-PAGE, suggesting to some extent, multimers were also formed during electrophoresis. Overall, soluble α-synuclein maintains lipid interactions following tissue disruption and readily forms multimers when this lipid-protein complex is preserved. Once the multimer-PAGE technique was validated, relative stoichiometric comparisons could be conducted simultaneously between 14 biol. samples. Multimer-PAGE provides a simple inexpensive biochem. technique to study the mol. factors influencing α-synuclein multimerization.
