L-Iduronic acid (IdoA) plays a pivotal role in glycosaminoglycan (GAG) protein interactions. However, the structural microheterogeneity of GAG appears to impede the systematic investigation of IdoA functions. Under such conditions, oligo-Iduronic acid (Oligo-IdoA) are ideal and straightforward heparin mimetics to unravel the relationship between IdoA structure and functions. Herein, we report for the first-time linear synthesis of rare oligo-IdoA precursor utilizing anhydrous β-L-idopyranosyl and IdoA thiophenol building block. After screening various synthetic strategies, we have installed successive IdoA by 5 step reactions with 25-26 % overall yield. These oligo-IdoA are expected to be excellent probes to understand conformational plasticity of IdoA and fine tune carbohydrate-protein interactions.

Wu-tou decoction has been used as a traditional Chinese medicine prescription for thousands of years. It comprises five herbs, namely Radix Aconiti Preparata, Ephedrae Herba, Astragali Radix, Glycyrrhiza Radix, and Paeoniae Radix Alba. In addition, the original prescription contains honey, but in modern research, the existence of honey is commonly ignored. The aim was to investigate the effect of absorption in rats after oral wu-tou decoction with or without honey. In this research, a rapid and sensitive UPLC-MS/MS method was investigated for the quant. anal. of ephedrine, pseudoephedrine, paeoniflorin, calycosin-7-glucoside, glycyrrhizic acid, liquiritin, and benzoylmesaconine in rat plasma after single and continuous oral decoctions. The results of the pharmacokinetic parameters showed that Cmax, CL/F, AUC0-t, and AUC0-∞ in the honey group were significantly increased than those in the non-honey group except for ephedrine and pseudoephedrine. The same trend was observed regardless of single or continuous oral administrations. Research studies showed that honey could promote the absorption of some effective components in wu-tou decoction in rats, enhance bioavailability, and provide a theor. basis for the scientific and rational compatibility of the original prescription.

Reported herein is a fluorometric and colorimetric sensor for the presence of trace amounts of water in organic solvents, using syn-bimane based boronate ester 1. This sensor responds to the presence of water with a highly sensitive turn-off fluorescence response, with detection limits as low as 0.018% water (volume/volume). Moreover, analogously high performance was observed when compound 1 was adsorbed on filter paper, with the paper-based sensor responding both to the presence of liquid water and to humid atmospheres. Reusability of the paper-based sensor up to 11 cycles was demonstrated, albeit with progressive decreases in the performance, and 1H NMR and mass spectrometry analyses were used to explain the observed, hydrolysis-based sensor response.

UV curable polyurethane acrylate (PUA) can be quickly solidified into film and directly coated on the surface of hydraulic machinery with excellent anti-wear, adhesion properties and low cost. In this work, a hyperbranched polyester synthesized by the stepwise reaction of trimethylolpropane (TMP) and dimethylolpropionic acid (DMPA) was as a ‘nucleus’, and the polyurethane prepolymer was obtained by grafting IPDI and PTMG. The UV-curing hyperbranched polyester polyurethane acrylate (PUA-1) was prepared by capping with HEA. Another UV-curing hyperbranched polyester polyurethane acrylate (PUA-2) was prepared as comparison by using TDI and PTMG as the main raw materials. Fourier Transform IR Spectrometer, 1H-NMR spectroscopy, Gel Permeation Chromatog., thermogravimetric anal. and dynamic thermomech. anal. were used to characterize its structure and properties. The wear rate of the two hyperbranched polyesters measured by the underwater steel ball method is below 3%, The swelling rate of PUA-1 is between 250%-300%, and the swelling rate of PUA-2 is around 260%, indicating that PUA-2 has better solvent resistance than PUA-1, and the internal structure of the film is more dense. TG anal. showed that the thermal stability of the two materials was good.

Metabolomic markers from tumor core biopsies have been shown to reflect cancer metabolic dysregulation and hold prognostic value. Implement and validate a novel ensemble machine learning approach to evaluate survival based on metabolomic biomarkers from tumor core biopsies. Data were obtained from tumor core biopsies evaluated with high-resolution 2DLC-MS/MS. Unlike biofluid samples, anal. of tumor tissue is expected to accurately reflect the cancer metabolism and its impact on patient survival. A comprehensive suite of machine learning algorithms were trained as base learners and then combined into a stacked-ensemble meta-learner for predicting “”short”” vs. “”long”” survival on an external validation cohort. An ensemble method of feature selection was employed to find a reliable set of biomarkers with potential clin. utility. Overall survival (OS) is predicted in external validation cohort with AUROCTEST of 0.881 with support vector machine meta learner model, while progression-free survival (PFS) is predicted with AUROCTEST of 0.833 with boosted logistic regression meta learner model, outperforming a nomogram using covariate data (staging, age, sex, treatment vs. non-treatment) as predictors. Increased relative abundance of guanine, choline, and creatine corresponded with shorter OS, while increased leucine and tryptophan corresponded with shorter PFS. In patients that expired, N6,N6,N6-Trimethyl-L-lysine, L-pyrogluatmic acid, and benzoic acid were increased while cystine, methionine sulfoxide and histamine were decreased. In patients with progression, itaconic acid, pyruvate, and malonic acid were increased. This study demonstrates the feasibility of an ensemble machine learning approach to accurately predict patient survival from tumor core biopsy metabolomic data.

Lignocellulosic biomass, particularly corn fiber, represents a renewable resource that is available in sufficient quantities from the corn wet milling industry to serve as a low cost feedstock for production of fuel alc. and valuable coproducts. Several enzymic and chem. processes have potential for the conversion of cellulose and hemicellulose to fermentable sugars. The hydrolyzates are generally rich in pentoses (D-xylose and L-arabinose) and D-glucose. Yeasts produce a variety of polyalcs. from pentose and hexose sugars. Many of these sugar alcs. have food applications as low-calorie bulking agents. During the screening of 49 yeast strains capable of growing on L-arabinose, we observed that two strains were superior secretors of L-arabitol as a major extracellular product of L-arabinose. Candida entomaea NRRL Y-7785 and Pichia guilliermondii NRRL Y-2075 produced L-arabitol (0.70 g/g) from L-arabinose (50 g/L) at 34° and pH 5.0 and 4.0, resp. Both yeasts produced ethanol (0.32-0.33 g/g) from D-glucose (50 g/l) and only xylitol (0.43-0.51 g/g) from D-xylose (50 g/l). Both strains preferentially utilized D-glucose > D-xylose > L-arabinose from mixed substrate (D-glucose, D-xylose and L-arabinose, 1:1:1, 50 g/L, total) and produced ethanol (0.36-0.38 g/g D-glucose), xylitol (0.02-0.08 g/g D-xylose) and L-arabitol (0.70-0.87 g/g L-arabinose). The yeasts co-utilized D-xylose (6.2-6.5 g/L) and L-arabinose (4.9-5.0 g/L) from corn fiber acid hydrolyzate simultaneously and produced xylitol (0.10 g/g D-xylose) and L-arabitol (0.53-0.54 g/g L-arabinose).

Although all marketed antiretrovirals (ARVs) have proven efficacy, genetic differences can result in varied effectiveness. This study was conducted to determine the effectiveness of different Highly Active Antiretroviral Therapy (HAART) combinations among patients attending HIV clinic at a Major Teaching Hospital in Ghana. The study was a retrospective study involving 500 patients at an HIV clinic in the Ashanti Region of Ghana. Twelve major antiretroviral combinations for HAART were prescribed at the study center. The most prescribed drug combinations were AZT + 3TC + EFV and AZT + 3TC + NVP. The study identified that HAART, irresp. of the kind of drug combination used, was effective at increasing CD4 count within the first 6 mo of therapy initiation in the study population. However, the magnitude of the increases differed from combination to combination. All HAART combinations with zidovudine as one of the drugs resulted in higher CD4 counts compared with combinations containing stavudine. HAART with nevirapine also resulted in a higher CD4 count than those with efavirenz. However, efavirenz-based combinations appeared to be more effective in critically ill patients and patients with mean CD4+T helper cells count below 100 cell/mm3. More importantly, common among all HAART combinations that resulted in treatment failure. There was significant variation in response to different HAART combination among Ghanaian HIV patients. However, there was no statistically significant difference in mean CD4 count between the two most predominately used HAART i. e AZT + 3TC + EFV and AZT + 3TC + NVP. More importantly, efavirenz was common among all HAART combinations that resulted in treatment failure.
