In multiple sclerosis (MS), the effect of di-Me fumarate (DMF) treatment is primarily attributed to its capacity to dampen pathogenic T cells. Here, we tested whether DMF also modulates B cells, which are newly recognized key players in MS, and to which extent DMF restricts ongoing loss of oligodendrocytes and axons in the central nervous system (CNS). Therefore, blood samples and brain tissue from DMF-treated MS patients were analyzed by flow cytometry or histopathol. examination, resp. Complementary mechanistic studies were conducted in inflammatory as well as non-inflammatory CNS demyelinating mouse models. In this study, DMF reduced the frequency of antigen-experienced and memory B cells and rendered remaining B cells less prone to activation and production of pro-inflammatory cytokines. Dissecting the functional consequences of these alterations, we found that DMF ameliorated a B cell-accentuated exptl. autoimmune encephalomyelitis model by diminishing the capacity of B cells to act as antigen-presenting cells for T cells. In a non-inflammatory model of toxic demyelination, DMF limited oligodendrocyte apoptosis, promoted maturation of oligodendrocyte precursors and reduced axonal damage. In a CNS biopsy of a DMF-treated MS patient, we equivalently observed higher numbers of mature oligodendrocytes as well as a reduced extent of axonal damage when compared to a cohort of treatment-naive patients. In conclusion, we showed that besides suppressing T cells, DMF dampens pathogenic B cell functions, which probably contributes to its clin. effectiveness in relapsing MS. DMF treatment may furthermore limit chronically ongoing CNS tissue damage, which may reduce long-term disability in MS apart from its relapse-reducing capacity.

Neuropathic pain (NP) that contributes to the comorbidity between pain and depression is a clin. dilemma. Neuroinflammatory responses are known to have potentially important roles in the initiation of NP and depressive mood. In this study, we aimed to investigate the effects of paeoniflorin (PF) on NP-induced depression-like behaviors by targeting the hippocampal neuroinflammation through the toll-like receptor 4 (TLR4)/nuclear factor-kappa B (NF-魏B) signaling pathway. We used a murine model of NP caused by unilateral sciatic nerve cuffing (Cuff). PF was injected i.p. once a day for a total of 14 days. Pain and depression-like behavior changes were evaluated via behavioral tests. Pathol. changes in the hippocampus of mice were observed by H&E staining. The levels of proinflammatory cytokines in the hippocampus were detected using ELISA. Activated microglia were measured by immunohistochem. staining. The TLR4/NF-魏B signaling pathwayassocd. protein expression in the hippocampus was detected by western blotting. We found that the PF could significantly alleviate Cuff-induced hyperalgesia and depressive behaviors, lessen the pathol. damage to the hippocampal cell, reduce proinflammatory cytokines levels, and inhibit microglial over-activation. Furthermore, PF downregulated the expression levels of TLR4/NF-魏B signaling pathwayrelated proteins in the hippocampus. These results indicate that PF is an effective drug for improving the comorbidity between NP and depression.

The catalytic activity of TMP-PECH-Amine-G1 was studied by synthesizing 4-aryl-1H-1,2,3-triazoles I (R = H, 3-Cl, 4-N(CH3)2, 3,4-(OMe)2, etc.) by one pot condensation reaction of aromatic aldehydes RCHO, nitromethane and sodium azide. The synthesized catalyst offered high yield of products with low catalyst loading in green reaction media without the presence of any external additives. The catalyst was regenerated for six cycles of reaction without substantial loss in activity. This is the first reported method where homogeneous amine functionalized dendronized polymer was used as an organo catalyst for the synthesis of 4-substituted triazoles I.

In the present study, the percentage, and components of essential oil, total phenol, flavonoids, anthocyanins, carotenoids and antioxidant capacity of 24 accessions of cultivated Damask rose have been investigated. These accessions had a 0.010-0.055 (v/w) percentage of essential oil. GC/MS anal. revealed forty-nine constituents that formed more than 90% of detected compounds Geraniol, citral, Me linoleate, n-heneicosane, and n-octane were the major components of essential oils. Acc.1 had the highest total phenol (165 mg Gallic acid/g DW) and flavonoid content (81 mg Quercetin/g DW). Total anthocyanin ranged from 0.5 to 3.5 (mg cyanidin-3-glucoside/g DW). Carotenoid content ranged from 0.002 to 0.055 mg/g DW, the highest being observed in Acc.18. Based on cluster anal. and, the accessions were categorized into seven groups. The antioxidant activities of the samples were determined using DPPH free radical scavenging activity and ferric reducing antioxidant power (FRAP) assay. All the accessions (4-12渭g/mL) had lower IC50 than ascorbic acid (18渭g/mL). In FRAP assay, they had high antioxidant activity in a range of 10-25渭mol Fe+2/g DW. Based on the high and valuable source of bioactive compounds of Damask rose the elite accessions can be recommended for boosting cultivation and food applications.
