The intensity to the m/z 61 ion also can be correlated with the double bond position. Results are generally accurate to ±1 C atom.

A splitless injector for gas chromatog. with a septum purge and liner purge flow for clearing off materials from the septum and minimizing solvent tails was designed and made, which can increase injected volume and sensitivity. The splitless injector was set up on a SP-103 gas chromatograph and used for the determination of insect pheromones. By using the splitless injector and a PEG-20 M capillary column, satisfactory results were obtained in separating geometrical and positional isomers of insect pheromones. The splitless injector is suitable for the determination of insect pheromones and other trace anal.

Besides gravity, roots are also guided by light to grow deep into the soil and sensitivity of roots to light is evidently due to presence of photoreceptors like phototropins. Such light-induced root growth (light-escape growth) presumably involves reactive oxygen species (ROS). Present study explores the possibility of ROS action in this event during early seedling growth of Vigna radiata based on pharmacol. evidences. Germinated (20 h) seeds were incubated in dark or light in presence of general ROS scavenger (Pr gallate), specific scavengers of O·2 (copper chloride; CuCl2), H2O2 [dimethylthiourea (DMTU) and potassium iodide (KI)] and ·OH (sodium benzoate) and ROS-producing enzyme inhibitors [zinc chloride (ZnCl2), inhibitor of NADPH oxidase (NOX); diethyldithiocarbamate (DEDTC), inhibitor of superoxide dismutase (SOD) and salicylhydroxamic acid (SHAM), inhibitor of peroxidase]. Light-induced root growth of 3-day seedlings diminished signifcantly in case of all the treatments suggesting for a pos. role of ROS in light-escape growth. This is supported by elevated level of apoplastic ROS in light grown roots as evident from ROS-specific staining [nitroblue tetrazolium chloride (NBT) for O·2 and 3,3,5,5-tetramethylbenzidine (TMB) for H2O2] and spectrophotometric estimation of apoplastic ROS production (O·2 and H2O2). In addition, higher activity of membrane bound NOX (producing O·2 ) and apoplastic class III peroxidase (Prx, producing ·OH) in light grown roots further corroborates the view that apoplastic ROS (initiated with NOX-generated O·2 , which is converted, either spontaneously or by the activity of SOD, to H2O2 and further metabolized by Prx to ·OH that participates in cell wall relaxation required for growth) is instrumental in light-escape growth of roots.

In field tests, Z9-12:Ac and Z11-14:OH together were required for trap catch, and addition of Z11-14:Al, E11-14:OH, 14:OH, or (Z)-11-tetradecen-1-ol acetate did not increase catch. Ratios in rubber spectra of 0.5:99.5 to 1.5:98.5 (Z9-12:Ac/Z11-14:OH) captured the most males and captures were statistically equivalent for dosages of 10-1000 μg/rubber septum. Traps baited with the synthetic lure produced better catches than those baited with females.

According to ISO 10993-1:2018, the skin sensitization potential of all medical devices must be evaluated, and for this endpoint ISO 10993-10:2010 recommends the use of in vivo assays. The goal of the present study was to determine if the in vitro SENS-IS assay could be a suitable alternative to the current in vivo assays. The SENS-IS assay uses the Episkin Large and SkinEthic RHE reconstructed human epidermis models to evaluate marker genes. In our study, the SENS-IS assay correctly identified 13 sensitizers spiked in a non-polar solvent. In a subsequent anal. six medical device silicone samples previously impregnated with sensitizers were extracted with polar and non-polar solvents. The SENS-IS assay correctly identified five of these extracts, while a sixth extract, which contained the weak sensitizer Ph benzoate, was classified as neg. However, when this extract was concentrated, or a longer exposure time was used, the assay was able to detect Ph benzoate. The SENS-IS assay was transferred to a naive laboratory which correctly identified sensitizers in six blinded silicone samples, including the one containing Ph benzoate. In light of these results, we conclude that the SENS-IS assay is able to correctly identify the presence of sensitizers in medical devices extracts

In addition to its action as a topoisomerase II poison, mitoxantrone is activated by formaldehyde to bind DNA, forming DNA-adducts specifically at 5’CpG and CpA sequences, with an enhancement of adducts at methylated CpG sites. The butyric acid prodrug, AN-9 (pivaloyloxymethyl butyrate), releases formaldehyde upon cellular hydrolysis and our previous studies have shown that mitoxantrone acts synergistically with AN-9 in cytotoxicity assays. In this paper, we investigated the impact of methylation levels in the cell on mitoxantrone-induced cytotoxicity using the colon cancer cell line HCT116 and its derived DNA methyltransferase (DNMT) 1 and DNMT 3a knockout (DKO8) cell line. We found that decreased methylation levels in the DNMT-null cells led to at least a 2-fold reduction in mitoxantrone-induced cytotoxicity. Next, we studied the impact of mitoxantrone alone, and in combination with AN-9, on hypermethylated genes and their mRNA expression in breast cancer cells. Using methylation-specific PCR and RT-PCR, we found that mitoxantrone treatment of breast cancer cell lines resulted in demethylation of the 14.3.3σ, cyclin D2 and ERα genes, followed by re-expression of their mRNA. The effect of mitoxantrone on re-expression of key genes involved in cell cycle regulation, and ensuing death of the cells may be an addnl., previously undiscovered mechanism of action of mitoxantrone.
