Peptide therapeutics are increasingly used in the treatment of disease, but their administration by injection reduces patient compliance and convenience, especially for chronic diseases. Thus, oral administration of a peptide therapeutic represents a significant advance in medicine, but is challenged by gastrointestinal instability and ineffective uptake into the circulation. Here, we have used glucagon-like peptide-1 (GLP-1) as a model peptide therapeutic for treating obesity-linked type 2 diabetes, a common chronic disease. We describe a comprehensive multidisciplinary approach leading to the development of MEDI7219, a GLP-1 receptor agonist (GLP-1RA) specifically engineered for oral delivery. Sites of protease/peptidase vulnerabilities in GLP-1 were removed by amino acid substitution and the peptide backbone was bis-lipidated to promote MEDI7219 reversible plasma protein binding without affecting potency. A combination of sodium chenodeoxycholate and Pr gallate was used to enhance bioavailability of MEDI7219 at the site of maximal gastrointestinal absorption, targeted by enteric-coated tablets. This synergistic approach resulted in MEDI7219 bioavailability of ∼ 6% in dogs receiving oral tablets. In a dog model of obesity and insulin resistance, MEDI7219 oral tablets significantly decreased food intake, body weight and glucose excursions, validating the approach. This novel approach to the development of MEDI7219 provides a template for the development of other oral peptide therapeutics.

Living radical polymerization with reversible addition-fragmentation chain transfer (RAFT polymerization) can be achieved with the use of dithiocarbamate derivatives that have the nonbonded electron pair of the nitrogen included as part of an aromatic system. These compounds have been shown to be highly effective in RAFT polymerization of styrene and (meth)acrylate esters to produce polymers of predetermined mol. weight and narrow polydispersity (usually <1.2). By contrast, simple N,N-dialkyl dithiocarbamates (those compounds previously described as “photoiniferters”) are ineffective as RAFT agents. The reasons for the different behavior are discussed.

Mass trapping of Spodoptera frugiperda (Smith) in Central America required a high performance lure with consistent capture rates. S. frugiperda lures from North America and England gave erratic capture rates under field conditions. A reinvestigation of four acetate attractants (Z9-14Ac, Z11-16Ac, Z7-12Ac and Z9-12Ac) for S. frugiperda present in Costa Rica revealed that Z7-12Ac and Z9-12Ac were highly attractive to S. frugiperda when presented alone. Binary combination of Z7-12Ac with Z9-14Ac significantly increased attraction and was at least 10 times more attractive to S. frugiperda in Costa Rica than North American or English lures. Addition of Z11-16Ac to binary combinations of Z9-14Ac and Z7-12Ac marginally increased capture rates. If the concentration of Z7-12Ac is increased to 5% in lures containing Z9-14Ac, Z11-16Ac and Z7-12Ac capture rates significantly decrease. The optimized lure contains Z7-12Ac that is a component not previously reported in S. frugiperda from the Caribbean region.

While monitoring for the seasonal occurrence of the tea tortrix moth, Homona magnanima Diakonoff (Lepidoptera: Tortricidae), a number of Neocalyptis angustilineata (Walsingham) were attracted to traps baited with H. magnanima attractant, a 9:1 blend of (Z)-11-tetradecenyl acetate (Z11-14Ac) and (Z)-9-dodecenyl acetate (Z9-12Ac). We evaluated a 1:1 blend (1 mg) and a 9:1 blend (1 mg) of Z11-14Ac and Z9-12Ac for their attractiveness to the two moth species. H. magnanima was attracted only to the 9:1 blend. However, N. angustilineata was equally attracted to both blends. Thus, we report the 9:1 blend as a co-attractant for N. angustilineata and H. magnanima. This blend is the first finding for the attractant for N. angustilineata.

The same compounds were also found in field-collected females and in effluvia. Z-9-Undecenyl acetate, which is a male attractant on its own, was also found in a sample of female effluvia. A hierarchy in the ethol. function of the pheromone components was observed Z9-12:Ac was an attractant for E. ambiguella males. Dodecyl acetate (12:Ac) was not attractant on its own, but augmented male catch when added to the main attractant. Addition of 18:Ac augmented attraction only when both Z9-12:Ac and 12:Ac were present. Wind-tunnel tests demonstrated that 18:Ac also raised the disorientation threshold, as previously shown for 12:Ac. Other compounds, with the possible exception of addnl. saturated acetates, had either no effect on trap catch or, in the case of E9-12:Ac, Z9-12:OH, and E-9,11-dodecadienyl acetate, were inhibitory above a certain level. A blend of roughly equal parts of Z9-12:Ac, 12:Ac, and 18:Ac provided the best blend for E. ambiguella known to date.

AN-9 and various DNA-specific antineoplastic agents synergistically inhibited the growth of Mm-A cells, and simultaneous treatment was required to evoke the maximum growth-inhibitory effect. On the other hand, there was no synergy between butyrate and these drugs, or between AN-9 and several antimetabolic agents, in inhibiting the growth of the cells, suggesting that the synergistic effect is specific to AN-9 and DNA-reacting agents. AN-9, given alone, dose-dependently prolonged the survival of mice inoculated with Mm-A cells. Moreover, administration of AN-9 plus daunorubicin (DNR) further prolonged survival.
