Demekhin, Oleg D.;Burov, Oleg N.;Kletskii, Mikhail E.;Lisovin, Anton V.;Kurbatov, Sergey V.;Bereznyak, Elena A.;Trishina, Alena V. research published 《 New 13-vinyl derivatives of berberine: synthesis and characterization》, the research content is summarized as follows. The possibility of obtaining electroneutral substituted 13-vinylberberines I [R = H, CH2C(O)CH3; R1 = dicyanomethyl, 1,3-dimethoxy-1,3-dioxopropan-2-yl, 2,4,6-trioxo-1,3-diazinan-5-yl, etc.] was demonstrated exptl. and via quantum-chem. DFT/B3LYP calculations in the 6-31++G(d,p) basis set. The introduction of pharmacophoric fragments conjugated through vinyl moiety opened new possibilities for structural modification of berberine, enabling pronounced changes in tropicity toward supramol. biol. structures. The newly synthesized 13-vinylberberines I were stable in their reduced form due to significant intramol. electron d. transfer from berberine ring system to the vinyl moiety bearing electron-withdrawing groups. It was demonstrated that berberine derivatives I may exist not only in ion pair form consisting of organic cation and inorganic anion, but also as zwitterionic structures featuring significant intramol. charge transfer. The obtained 13-vinylberberines I exhibited biol. activity against the highly pathogenic Vibrio cholerae.

Dube, Phelelisiwe S.;Legoabe, Lesetja J.;Jordaan, Audrey;Jesumoroti, Omobolanle J.;Tshiwawa, Tendamudzimu;Warner, Digby F.;Beteck, Richard M. research published 《 Easily accessed nitroquinolones exhibiting potent and selective anti-tubercular activity》, the research content is summarized as follows. Nitro based DprE1 inhibitors exemplified by benzothiazinones have been reported to elicit potent anti-tubercular activity. Poor PK properties associated with benzothiazinones have inspired the discovery of alternative nitro based DprE1 inhibitors. Quinolone based antibiotics on the other hand have good PK properties. The potent anti-tubercular activity of nitro compounds and the good PK properties of the quinolones have elicited an interest in us to construct a new class of nitro containing compounds around the quinolone scaffold with the aim of identifying novel DprE1 inhibitors with potent anti-tubercular activity. Thus, we report herein the anti-tubercular activity of novel 6-nitroquinolone-3-carboxamide derivatives achieved using less than five cheap synthetic transformations. Among the 23 target compounds evaluated for anti-tubercular activity, 12 were active against Mtb- exhibiting activity in the range of <0.244-31.865 μM. Compound 25 having a mol. weight of 399 Da and ClogP value of 2.7 is the most active (MIC90: <0.244 μM) in this series. The SAR analyses suggest that anti-tubercular activity was influenced by substituents at position N-1 (R2) and C-3 (R3) of the quinolone ring. The activity data suggest that the nature of R3 has a stronger influence on the SAR compared to R2; with a fluorobenzyl and chlorobenzyl moiety at R2 being the most favored when R3 is an aliphatic amine. Docking study confirms that compound 25 binds to the same hydrophobic pocket as does TCA1, and other nitro based DprE1 inhibitors, with its nitro group in close proximity with Cys387 residue.

Kinetic resolution provided a highly enantioselective method to access a range of 3-aryl-3,4-dihydro-2H-1,4-benzoxazines using n-butyllithium and the chiral ligand sparteine. The enantioenrichment remained high on removing the tert-butoxycarbonyl (Boc) protecting group. The intermediate organolithium underwent opening to an enamine. The kinetic resolution was extended to give enantiomerically enriched substituted 1,2,3,4-tetrahydroquinoxalines and was applied to the synthesis of an analog of the antibiotic levofloxacin that was screened for its activity against the human pathogen Streptococcus pneumoniae.

The oxycyclohexyl acid BMS-986278 (33) is a potent lysophosphatidic acid receptor 1 (LPA1) antagonist, with a human LPA1Kb of 6.9 nM. The structure-activity relationship (SAR) studies starting from the LPA1 antagonist clin. compound BMS-986020 (1), which culminated in the discovery of 33, are discussed. The detailed in vitro and in vivo preclin. pharmacol. profiles of 33, as well as its pharmacokinetics/metabolism profile, are described. On the basis of its in vivo efficacy in rodent chronic lung fibrosis models and excellent overall ADME (absorption, distribution, metabolism, excretion) properties in multiple preclin. species, 33 was advanced into clin. trials, including an ongoing Phase 2 clin. trial in patients with lung fibrosis (NCT04308681).

870-50-8, Di-tert-butyl azodicarboxylate, also known as Di-tert-butyl azodicarboxylate, is a useful research compound. Its molecular formula is C₁₀H₁₈N₂O₄ and its molecular weight is 230.26 g/mol. The purity is usually 95%.
Di-tert-butyl azodicarboxylate is a reagent used in the electrophilic amination of β-keto esters catalyzed by an axially chiral guanidine. Building block in an enantioselective synthesis of 3,6-dihyropyridazines employing organocatalysts such a L-proline or (S)-2-pyrrolidinyl tetrazole. Utilized in the asymmetric Friedel-Crafts amination via a chiral organocatalyst.
Di-tert-butyl azodicarboxylate is a reagent used in the preparation of acyl hydrazinedicarboxylates via photoorganocatalytic hydroacylation of dialkyl azodicarboxylates with aldehydes in presence of phenylglyoxylic acid as photocatalyst., Application In Synthesis of 870-50-8
