Purpose: To investigate by in silico screening the anti-inflammatory constituents of Cinnamomum cassia twigs. Methods: Information on the constituents of C. cassia twigs was retrieved from the online Traditional Chinese Medicines (TCM) database and literature. Inflammation-related target proteins were identified from DrugBank, Online Mendelian Inheritance in Man (OMIM), Therapeutic Target Database (TTD), Genetic Association Database (GAD), and PharmGKB. The identified compounds were filtered by Lipinski’s rules with Discovery Studio software. The Libdock module was used to perform molecular docking; LibdockScores and default cutoff values for hydrogen bonds and van der Weals interactions were recorded. LibdockScores between the prototype ligand and target protein were set as the threshold; compounds with higher LibdockScores than threshold were regarded as active compounds. Cytoscape software was used to construct active constituent-target protein interaction networks. Results: Sixty-nine potential inflammatory constituents with good drug-like properties in C. cassia twigs were screened in silico based on molecular docking and network pharmacology analysis. JAK2, mPEGS-1, COX-2, IL-1 beta, and PPAR gamma were considered the five most important target proteins. Compounds such as methyl dihydromelilotoside, hierochin B, dihydromelilotoside, dehydrodiconiferyl alcohol, balanophonin, phenethyl (E)-3-[4-methoxyphenyl]-2-propenoate, quercetin, and luteolin each interacted with more than six of the selected target proteins. Conclusion: C. cassia twigs possess active compounds with good drug-like properties that can potentially be developed to treat inflammation with multi-components on multi-targets.

Regioselectivity for intramolecular Diels-Alder (IMDA) reactions of 6-acetoxy-6-alkenylcyclohexa-2,4-dien-1-ones that were formed by oxidation of 2-alkenylphenols with lead tetraacetate in acetic acid were studied. Bridged regioselectivity was observed in the IMDA reactions of 6-acetoxy-6-alkenylcyclohexa-2,4-dien-1-ones having a dienophile part which could conjugate with an aromatic group. Bridged seven-and eight-membered rings and bicyclo[2.2.2]octane skeletons were constructed by the present IMDA reactions. Density functional theory (DFT) calculations suggested that conjugation of the dienophile with neighboring aromatic groups lowered the highest occupied molecular orbital-lowest unoccupied molecular orbital (HOMO-LUMO) energy gap and preceded bridged [4 + 2] adducts.

The aromatic perennial plant Alpinia zerumbet (Pers.) Burtt & Smith (Zingiberaceae) is widely distributed in the tropical and sub-tropical regions of Japan, from southern Kyushu to the Ryukyu Islands. Its essential oil has a distinct aroma and antioxidant activity but is expensive due to low extraction efficiencies. Applying underwater shockwave pretreatment to A. zerumbet leaves is expected to enable more effective essential oil extraction during subsequent steam distillation. Thus, we subjected A. zerumbet leaves to shockwave pretreatment and analyzed its effects on the essential oil yield and components. Multiple cracks formed in the cell walls via spalling destruction induced by the shockwaves, creating permeation pathways that improved the passage of water vapor and greatly increased the essential oil yield. Moreover, the pretreatment increased the antioxidant activity of the essential oil and its water extract. Underwater shockwave processing selectively and effectively destroyed the fiber and/or cell structures of the leaves, resulting in efficient essential oil extraction. This novel process has the potential for application to many extraction processes, such as the extraction of compounds from medicinal plants. The dynamic control of instantaneous high pressures based on underwater shockwaves could pave the way for a new field of industrial technology. (C) 2020 Institution of Chemical Engineers. Agarwood, a species of resinous heartwood, is a precious medicinal plant and a type of rare natural spice, which is widely used in medicine, cosmetics, religious activities, and other fields. In this study, agarwood samples from eight different regions across four countries were analyzed by comprehensive two-dimensional gas chromatography-quadrupole time-of-flight mass spectrometry. A total of 232 species were identified (the match factors of these compounds were above 750). The main compounds of agarwood are oxygenated sesquiterpenes and chromones. The compositions of India1 and Malaysia2 were significantly different from those of other samples, which might be attributed to the different production processes of agarwood. For further investigation, factor analysis was conducted for six agarwood samples. The results showed that the data classification possessed a regional characteristic; according to the retention time and relative content, characteristic compositions were determined by factor scores. Finally, the differences of characteristic compositions were simply analyzed, and the reasons were speculated.

We report herein cobalt-catalyzed enantioselective and chemodivergent reactions between a cyclopropanol and an oxabicyclic alkene via a cobalt homoenolate, which afford either an alkylative ring-opening product or a hydroalkylation product, with the counterion of the cobalt catalyst being a major chemoselectivity-controlling factor. A catalyst generated from cobalt(II) chloride and a chiral diphosphine promotes alkylative ring opening to afford 1,2-dihydronaphthalen-1-ol derivatives in good yields with high enantioselectivity. By contrast, a catalyst generated from cobalt(II) acetate and the same diphosphine ligand, with the assistance of methanol, selectively affords hydroalkylation products with retention of the bicyclic structure at a comparable level of enantioselectivity.

We identified the most popular electronic cigarette (EC) refill fluids using an Internet survey and local and online sales information, quantified their flavor chemicals, and evaluated cytotoxicities of the fluids and flavor chemicals. Berries/Fruits/Citrus was the most popular EC refill fluid flavor category. Twenty popular EC refill fluids were purchased from local shops, and the ingredient flavor chemicals were identified and quantified by gas chromatography-mass spectrometry. Total flavor chemical concentrations ranged from 0.6 to 27.9 mg/ml, and in 95% of the fluids, total flavor concentration was greater than nicotine concentration. The 20 most popular refill fluids contained 99 quantifiable flavor chemicals; each refill fluid contained 22 to 47 flavor chemicals, most being esters. Some chemicals were found frequently, and several were present in most products. At a 1% concentration, 80% of the refill fluids were cytotoxic in the MTT assay. Six pure standards of the flavor chemicals found at the highest concentrations in the two most cytotoxic refill fluids were effective in the MTT assay, and ethyl maltol, which was in over 50% of the products, was the most cytotoxic. These data show that the cytotoxicity of some popular refill fluids can be attributed to their high concentrations of flavor chemicals.

Objectives: The incidence of acute kidney injury (AKI) in infective endocarditis (IE), its risk factors and consequences on patient and renal survival remain debated. Methods: Patients hospitalized for a first episode of IE (possible or definite according to modified Duke criteria) between 2013 and 2016 were included. The primary endpoint was to determine risk factors for early AKI (E-AKI) during the first week of management of IE. Results: A total of 276 patients were included: 220 (79.7%) had definite IE and 56 (20.3%) had possible IE. E-AKI occurred in 150 patients (53%). IE due to Staphylococcus aureus (OR 3.41; 95% CI 1.83-6.39; p < 0.01), history of diabetes (OR 2.34; 95% CI 1.25-4.37; p < 0.01), peripheral arterial disease (OR 2.59; 95% CI 1.07-6.23; p < 0.05), immunological manifestations (OR 3.11; 95% CI 1.31-7.39; p = 0.01), and use of norepinephrine (OR 3.44; 95% CI 1.72-7.02; p < 0.01) were associated with E-AKI. In subgroup analysis, infectious disease consultation was associated with a lower risk of AKI at day 7 (OR 0.41; 95% CI 0.16-0.88; p = 0.04). E-AKI was associated with 1-year mortality (OR 1.65; 95% CI 1.03-2.64; p = 0.04) and chronic kidney disease progression (OR 2.23; 95% CI 1.30-3.82; p < 0.01). Conclusions: E-AKI is common in IE and often associated with non-modifiable variables. Multidisciplinary management should be mandatory, and awareness of AKI diagnosis and etiological explorations should be raised. (C) 2020 The Authors. Alpinia galanga is a wild medicinal plant having tremendous industrial importance. However, lack of quality planting material in natural resources restricted its wide exposure in industrial sector. Thus, a standardised protocol for stable supply of quality planting materials with genetical and phytochemical stability is need of the hour to meet commercial requirements. The current study reports an efficient protocol to regenerate micropropagated plantlets with long term stability in molecular as well as phytochemical level. Murashige and Skoog (MS) media augmented with 3 mg/l 6-Benzyladenine (BA), 3 mg/l Kinetin (Kn) and 1 mg/l Napthalene Acetic acid (NAA) was found optimum for regeneration and multiplication of plantlets. In order to establish genetic stability molecular marker-based profiling of micropropagated plants were done at every 6 months intervals up to 6 years. Monomorphic banding pattern were found that are identical to the mother plants. After six years of in vitro conservation, the regenerated plants were transplanted to the field and its drug yielding potential were evaluated through phytoconstituent analysis along with bioactivity studies. A comparative Gas chromatography (GC) and mass spectroscopy (MS) analysis of essential oils showed no significant differences in phytoconstituents between the conventional and in vitro propagated plants. The bioactivities like antioxidant, antimicrobial and anticancerous study along with total phenolic and total flavonoid content of in vitro plants were compared with mother plants and found stable with negligible variation. Thus, the present study has enough significance towards commercial exploitation of A. galanga with phytochemical and molecular stability.
