Theoretical and experimental studies of pyranopyrazoles and their tribological compatibility with a borate ester

The tribological properties of fused heterocyclic ring systems have been scarcely studied. Generally, increased heteroatoms in such molecules should make them better additives. Accordingly, antiwear behavior of substituted pyranopyrazoles (PPz-R, where R=H, methyl and methoxy), compounds with fused heterocyclic rings, have been investigated in the present research. These compounds were characterized by FTIR and NMR (H-1 and C-13) spectroscopy. Their tribological activity followed the order PPz-OMe > PPz-Me > PPz-H > ZDDP (standard). Atomic force microscopy (AFM) and scanning electron micrographs (SEM) of wear scar lubricated with pyranopyrazoles exhibited surface smoothness according to tribological results. The energy-dispersive X-ray (EDX) spectroscopy analysis revealed the heteroatoms nitrogen and oxygen of additive on the surface. XPS analysis showed that interaction of the best additive, PPz-OMe with a borate ester (Van lube 289), increased the efficiency synergistically. Results of DFT calculations on the interaction between adsorbent and substituted pyranopyrazoles are in agreement with the experimental data. The order of adsorption energies found using molecular dynamics (MD) simulations, correlated very well with the additive activity order. Furthermore, MD studies gave the conformation of the additive molecules on the adsorbing surface, paving the way to an understanding of the mechanism of adsorption and triboactivity.

Comparison of three different analytical protocols for 2019 updated D2425 method for renewable jet fuel product certification analysis

ASTM standard specification D7566 covers the manufacture of synthetic aviation turbine fuel components and their blends with conventional Jet fuel (Jet A or Jet A-1). One of the components is renewable jet fuel (RJF) which is synthetic paraffinic kerosene (SPK) made from hydroprocessed esters and fatty acids (HEFA). The specification D7566 dictates property requirements for the SPK-HEFA, including concentration limits for selected hydrocarbon types (paraffins, cycloparaffins, and aromatics), which are analyzed by using the mass spectrometry (MS) based standard method D2425. The most recent update for D2425 released in 2019 includes the synthetic hydrocarbon sample type (e.g., SPK-HEFA) and defines various analytical procedures for the analysis. Notably, the procedures differ considerably from each other, and the experimental conditions are not defined in details. This leads to laboratories setting up analytical schemes for D2425 that are likely to differ from each other, which may result in variation in the quality of the results obtained in different laboratories. In the present study, the performances of D2425 analytical protocols set up by three laboratories were tested in certification analysis (D7566) of SPK-HEFA type RJF. The tested analytical protocols were proven to comply with the requirements of the 2019 version of the D2425 standard. Furthermore, the precisions of the protocols did not differ significantly from each other. However, a significant bias was found for the results obtained for cycloparaffins and aromatics. Further, considerable differences were found in the bias values between the laboratories. Based on the results of this study, the guidelines of the 2019 updated D2425 standard may result in setting up an analytical protocol for D2425 which may not be optimal for RJF certification. DHA 12-LOX-derived oxylipins regulate platelet activation and thrombus formation through a PKA-dependent signaling pathway

Background The effects of docosahexaenoic acid (DHA) on cardiovascular disease are controversial and a mechanistic understanding of how this omega-3 polyunsaturated fatty acid (omega-3 PUFA) regulates platelet reactivity and the subsequent risk of a thrombotic event is warranted. In platelets, DHA is oxidized by 12-lipoxygenase (12-LOX) producing the oxidized lipids (oxylipins) 11-HDHA and 14-HDHA. We hypothesized that 12-LOX DHA-oxylipins may be involved in the beneficial effects observed in dietary supplemental treatment with omega-3 PUFAs or DHA itself. Objectives To determine the effects of DHA, 11-HDHA, and 14-HDHA on platelet function and thrombus formation, and to elucidate the mechanism by which these omega-3 PUFAs regulate platelet activation. Methods and results DHA, 11-HDHA, and 14-HDHA attenuated collagen-induced human platelet aggregation, but only the oxylipins inhibited IIb beta 3 activation and decreased -granule secretion. Furthermore, treatment of whole blood with DHA and its oxylipins impaired platelet adhesion and accumulation to a collagen-coated surface. Interestingly, thrombus formation was only diminished in mice treated with 11-HDHA or 14-HDHA, and mouse platelet activation was inhibited following acute treatment with these oxylipins or chronic treatment with DHA, suggesting that under physiologic conditions, the effects of DHA are mediated through its oxylipins. Finally, the protective mechanism of DHA oxylipins was shown to be mediated via activation of protein kinase A. Conclusions This study provides the first mechanistic evidence of how DHA and its 12-LOX oxylipins inhibit platelet activity and thrombus formation. These findings support the beneficial effects of DHA as therapeutic intervention in atherothrombotic diseases.

Simultaneous Dyeing and Finishing of Cotton Fabric in Presence of Starch Nanoparticles as Reinforcement Agent Using Citric Acid Based Finishing Formulation

To augment high performance, coloration and functional finishing onto cotton fabrics; novel approach for simultaneous dyeing and finishing in addition to reinforcement effect were applied by conventional pad-dry-cure method in one step. This was done to explore the potential of reduction in energy consumption and production time as well as dyes and chemicals. For this purpose, our previously prepared starch nanoparticles (SNPs) as a reinforcement biopolymer were incorporated in cross-linking formulation of cotton fabrics embracing different concentrations of citric acid and cationic dyes, i.e., Methylene Blue and Crystal Violet in presence of sodium hypophosphite as a proficient catalyst at 150(0)C for 2 minutes. Evaluation of the treated fabric performance with respect to color strength (K/S), fastness properties to light, rubbing and perspiration, add on %, dry wrinkle recovery angles, tensile strength, fabric stiffness, antibacterial effect and ultraviolet protection factor were appraised. It is seen from the obtained results that; the simultaneously dyed and finished fabric in presence of SNPs showed higher color strength, dry wrinkle recovery angle, tensile strength, fabric stiffness, antibacterial and UPF functionalities in addition to considerable light, rubbing and perspiration fastness in comparison with untreated fabric. Besides, FTIR spectra exhibited the chemical structure of treated fabrics with respect to the presence of the ester crosslinking and functional groups of cationic dyes in questions in comparison with untreated fabric. The durability of the imparted functional properties was confirmed also by washing test.

CAPE was embedded into the PCL core via hydrophobic interactions, while DOX was loaded within a mixed PAA/PEO corona layer through complexation. The main physico-chemical properties of blank and dual drug-loaded micelles were investigated using dynamic and electrophoretic light scattering and transmission electron microscopy. Loading of CAPE and DOX increased the size and size distribution of carriers to some extent. Highly engineered phytases, which sequentially hydrolyze the hexakisphosphate ester of inositol known as phytic acid, are routinely added to the feeds of monogastric animals to improve phosphate bioavailability. New phytases are sought as starting points to further optimize the rate and extent of dephosphorylation of phytate in the animal digestive tract. Multiple inositol polyphosphate phosphatases (MINPPs) are clade 2 histidine phosphatases (HP2P) able to carry out the stepwise hydrolysis of phytate. MINPPs are not restricted by a strong positional specificity making them attractive targets for development as feed enzymes. Here, we describe the characterization of a MINPP from the Gram-positive bacterium Bifidobacterium longum (BlMINPP). BlMINPP has a typical HP2P-fold but, unusually, possesses a large alpha-domain polypeptide insertion relative to other MINPPs. This insertion, termed the U-loop, spans the active site and contributes to substrate specificity pockets underpopulated in other HP2Ps. Mutagenesis of U-loop residues reveals its contribution to enzyme kinetics and thermostability. Moreover, four crystal structures of the protein along the catalytic cycle capture, for the first time in an HP2P, a large ligand-driven alpha-domain motion essential to allow substrate access to the active site. This motion recruits residues both downstream of a molecular hinge and on the U-loop to participate in specificity subsites, and mutagenesis identified a mobile lysine residue as a key determinant of positional specificity of the enzyme. Taken together, these data provide important new insights to the factors determining stability, substrate recognition, and the structural mechanism of hydrolysis in this industrially important group of enzymes.
