Injectable Hypoxia-Induced Conductive Hydrogel to Promote Diabetic Wound Healing

Injectable hydrogels with the capability to cast a hypoxic microenvironment is of great potentialities to develop novel therapies for tissue regeneration. However, the relative research still remains at the conceptual phase. Herein, we chose diabetic wound as a representative injury model to explore the actual therapeutic results of tissue injury by injectable hypoxia-induced hydrogels. To enhance recovery and widen applicability, the hypoxia-induced system was incorporated with a conductive network by an original sequentially interpenetrating technique based on the combination of a fast click chemistry and a slow enzymatic mediated cross-linking. Hyperbranched poly(beta-amino ester)-tetraaniline (PBAE-TA) was cross-linked with thiolated hyaluronic acid (HA-SH) via a thiol-ene click reaction, contributing to the rapid formation of the first conductive network, where vanillin-grafted gelatin (Geln-Van) and laccase (Lac) with a slow cross-linking rate were employed in casting a hypoxic microenvironment. The as-prepared injectable hydrogels possessed both suitable conductivity and sustainable hypoxia-inducing capability to upregulate the hypoxia-inducible factor-la and connexin 43 expressions of the encapsulated adipose-derived stem cells, which enhanced vascular regeneration and immunoregulation and further promoted the reconstruction of blood vessels, hair follicles, and dermal collagen matrix, eventually leading to the recovery of diabetic rat skin wounds and restoration of skin functions. This work provides a promising strategy to broaden the applicability of diverse hydrogels with a long time-consuming gelation process and to integrate different networks with various biological functions for the therapies of diabetic wounds and other complex clinical symptoms.

Fine root biomass stimulates microbial activity of glucose mineralization in buried humic horizon of volcanic ash soils

Burial of the surface soil by volcanic ash deposition plays roles in organic matter accumulation. Assuming that decomposition of the buried humus is limited solely by labile substrate inputs, glucose inputs are hypothesized to stimulate microbial activity in buried humic horizons. We describe patterns of microbial mineralization of C-14-labeled glucose in volcanic soil profiles to test whether glucose inputs increase microbial activity similarly in the surface soil and buried humic horizons. We found that microbial biomass was dependent on fine root biomass rather than soil carbon concentration in the volcanic soil profiles. Both bacterial and fungal biomass correlated with fine root biomass. The microbial capacities of glucose mineralization in the buried humic horizons were lower than in the surface horizons. The microbial activity of glucose mineralization in the buried humic horizons is limited by (1) the smaller microbial biomass due to smaller fine root biomass and (2) the dominance of microbial community adapted to the low concentrations of substrates with low mineralization capacity. Stimulation of microbial activities in the buried humic horizons requires both fine root biomass and inputs of labile substrates.

Aging is a fundamental and fascinating process. Anti-aging research tried to find the mysteries about the human lifespan. To investigate the longevity-extending role of caffeic acid phenethylester (CAPE) and reveal the possible regulation mechanism, the long-term cultivation under well-defined environments, real-time monitoring, and live imaging is highly desired. In this paper, a well-designed microfluidic device was proposed to analyze the anti-aging effect of CAPE in Caenorhabditis elegans. With the combined use of multiple functional units, including micro-pillar, worm responder, a branching network of distribution channels, and microchambers, the longitudinal measurements of the exact number of worms throughout the whole lifespans is possible. Meanwhile, the brief cooling function of temperature-controllable system can achieve temporary and repeated immobilization of nematodes for fluorescence imaging. Our research data showed that CAPE can increase the survival of worms under normal and stress condition, including heat stress and paraquat-induced oxidative stress. The further studies revealed the anti-aging mechanism of CAPE. This proposed strategy and device would be a useful platform to facilitate future anti-aging studies and the finding of new lead compounds.

Columnar self-assembly, electrochemical and luminescence properties of basket-shaped liquid crystalline derivatives of Schiff-base-moulded p-tert-butyl-calix[4]arene

An electrochemical study was carried out using cyclic voltammetry (CV) and the results were compared with theoretical values obtained from a DFT study. These compounds showed light sky-blue luminescence in solution and as a thin film under long-wavelength UV light. It was observed that all the prepared target compounds exhibited enantiotropic mesophases with good thermal stability. The structural and conformational characterisation of these new compounds was achieved using various spectroscopic techniques, including FT-IR, H-1 NMR, C-13 NMR and MALDI-TOF. The relationship between the structure and the mesomorphic behaviour of the compounds containing an ethyl bridge with linked ester groups (5a-5d) is discussed. This new class of supramolecular derivatives is promising, especially considering the emissive nature and stabilisation of their columnar hexagonal mesophases, and their high range of thermal stability.

The transfection efficiency of siRNA mediated by cationic polymers is limited due to the instability of polymers/siRNA complexes in the presence of serum. Poly(ethylene glycol) (PEG) is usually applied to modify cationic polymers, so as to reduce protein and cell adsorption and then to improve siRNA transfection efficiency. However, the polymers’ modification with PEG mostly consumes the free amino of the polymers, which can, in turn, reduce the charge density and limit their siRNA transfection efficacy. Here, a new PEG modification strategy that need not consume the surface aminos of polymers is proposed. Catechol-PEG polymers are coated on the surface of phenylboronic acid (PBA)-modified Generation 5 (G5) poly(amidoamine) dendrimers (G5PBA) via reversible boronate esters to establish PEG-modified dendrimer/siRNA nanoassemblies for efficient siRNA delivery. The PEG/G5PBA/siRNA nanoassemblies have positive charge and show excellent gene silencing efficacy in the absence of serum in vitro. More importantly, the PEG/G5PBA/siRNA nanoassemblies also exhibit excellent serum resistance and gene silencing efficacy in serum-containing medium. Furthermore, the effective antiserum and gene silencing efficacy elicited by these nanoassemblies lead to excellent antitumor effects in vivo. The syntheses employ an orthogonally protected trisaccharide as a pivotal building block that can be readily converted into the donor and acceptor for glycosidic coupling. In addition, the internal patterns of protecting groups, involving N-trichloroacetyl for N-acetyl group, benzylidene and benzyl groups for sulfonated hydroxyl groups, and benzoyl and methyl esters for free hydroxyl and carboxylic acid, respectively, ensure stereoselective formation of the glycosidic linkages and sequential transformation into the desired FuCS oligosaccharides.
