Direct interaction of dithizone with 7-chloro-6-fluoro-8-nitro-4-oxoquinoline-3-carboxylic acid and its ester delivered the corresponding novel 1,3,4-thiadiazino[5,6-h]quinoline-3-carboxylic acid and its ester I [R = H, Et] resp., alternatively latter ester derivative I [R = Et] was hydrolyzed to its acid derivative I [R = H]. Structures of the latter tricyclic hybrids I [R = H, Et] were deduced from anal. and spectral data and confirmed by single crystal X-ray structure determination of the compound I [R = Et]. The synthesized title compounds I [R = H, Et] were evaluated for antibacterial activity against Staphylococcus aureus ATCC 6538 and Escherichia coli ATCC 8739, antifungal activity against Candida albicans. Compound I [R = H] showed moderate activity against Staphylococcus aureus with a MIC value of 25 μg mL-1 and compounds I [R = H, Et] were inactive against Candida albicans.

INTRODUCTION: Early disease control with disease-modifying drugs is important for improving the prognosis of multiple sclerosis (MS) in children. Dimethyl fumarate (DMF) is an oral disease-modifying drug for MS in adults with relatively stable disease; however, its use in young children has not been heavily documented in the current literature. We report the case of a pediatric patient with relapsing-remitting MS who was treated with DMF. CASE REPORT: A 3-year-old boy with a history of common cold symptoms developed unsteadiness and somnolence. Magnetic resonance imaging revealed multiple white matter lesions. Symptoms were recurrent, and DMF was prescribed at 6 years of age due to a relapse episode with oculomotor disability and facial paralysis. However, disease progression continued, and new lesions were noted at age 7; thus, the dose of DMF was increased to 240 mg/day. No relapse has been observed for over three years; sequelae or severe side effects were absent. CONCLUSIONS: DMF may be a useful oral disease-modifying drug for preventing recurrence in young children with MS.

The multi-component reaction of Et acetoacetate with each of malononitrile (3) benzaldehyde (1) in ethanol containing triethylamine gave the Et 6-amino-5-cyano-2-methyl-4-phenyl-4H-pyran-3-carboxylate (4). The latter compound reacted with hydrazine hydrate to give the hydrazide derivative 6. Compound 6 underwent a series of hetero-cyclization reactions to give pyrzole, hydraide-hydrazone, thiazole derivatives The produced compounds tested against cancer cell lines six cancer cell lines and showed that compounds 8b, 10b, 11a, 17a, 21 and 24a were the most cytotoxic compounds Further tests of the latter compounds toward the five tyrosine kinases and Pim-1 kinase showed that compounds 10b, 21 and 24a were the most potent of the tested compounds and compounds 10a, 11a and 17a were of the highest inhibitions toward Pim-1 kinase. The high inhibitions of most of the tested compounds toward the selected cancer cell lines and the tyrosine kinases encourage for future work to be done.

Hydrothermal liquefaction (HTL) of fractionated two types herbaceous biomass (kenaf and miscanthus) by dilute acid, organosolv, alk., or demineralization process was carried out under ethanol water co-solvent at 350°C for 30 min to examine the biocrude yield and characteristics. The biocrude properties were comprehensively characterized by HPLC, elemental, GC-MS, and TGA anal. Fractionation technologies before HTL effectively increased biocrude yield up to 38% compared to that of untreated herbaceous biomass (31%), except for organosolv fractionation of miscanthus, especially, HTL after alk. fractionation showed high yield and energy recovery ratio up to 70%. Elemental anal. showed that HHV of biocrude was neg. affected by hydrolysis reaction of high lignin content after dilute acid fractionation. The GC-MS anal. revealed that carbohydrates-derived compound significantly increased in the biocrude obtained after organosolv and alk. fractionation due to holocellulose increases through fractionation process. Addnl., TGA results indicated that the ratio of high-boiling-point compounds in biocrude obtained after demineralization was expanded compared with untreated due to ash removal, which could act as a catalyst.

Quorum sensing is a mechanism of chem. communication among bacteria that enables collective behaviors. In V. cholerae, the etiol. agent of the disease cholera, quorum sensing controls group behaviors including virulence factor production and biofilm formation. The major V. cholerae quorum-sensing system consists of the extracellular signal mol. called CAI-1 and its cognate membrane bound receptor called CqsS. Here, the ligand binding activity of CqsS is probed with structural analogs of the natural signal. Enabled by the discovery of a structurally simplified analog of CAI-1, a focused library was prepared and analyzed. The mols. were designed to probe the effects of conformational and structural changes along the length of the fatty acid tail of CAI-1. The results, combined with pharmacophore modeling, suggest a mol. basis for signal mol. recognition and receptor fidelity with respect to the fatty acid tail portion of CAI-1. These efforts provide novel probes to enhance discovery of anti-virulence agents for the treatment of V. cholerae.

The adaptor protein STING plays a major role in innate immune sensing of cytosolic nucleic acids, by triggering a robust interferon response. Despite the importance of this protein as a potential therapeutic target for serious unmet medical conditions including cancer and infectious disease there remains a paucity of STING ligands. Starting with a benzothiazinone series of weak STING activators (human EC50 ∼10μM) we identified several chemotypes with sub-micromolar STING activity across all the major protein polymorphs. An example compound I, based on an oxindole core structure, demonstrated robust on-target functional activation of STING (human EC50 185 nM) in immortalized and primary cells and a cytokine induction fingerprint consistent with STING activation. Our study has identified several related series of potent small mol. human STING activators with potential to be developed as immunomodulatory therapeutics.
