Liver fibrosis is a reversible wound-healing response to acute or chronic liver injury that can progress to cirrhosis and liver cancer. Finding new strategies for prevention and management of liver fibrosis is urgently needed. It is known that hepatic stellate cell (HSC) is the primary source of extracellular matrix that drives liver fibrosis progression. Herein, we carried out a comprehensive secretome profiling to identify NMN-induced changes in secretory proteins and found that NMN suppressed the secretion of profibrotic protein and oxidoreductase in activated HSC (LX-2) cells, while real-time quant. PCR anal. revealed that NMN downregulated profibrotic gene expression, resulting in HSC inactivation. Next, we demonstrated that NMN (NMN) reduced the accumulation of liver extracellular matrix in thioacetamide (TAA) and carbon tetrachloride (CCl4) induced mouse models for liver fibrosis. Furthermore, we determined that NMN inhibited oxidation-mediated 15-PGDH degradation to promote prostaglandin E2 degradation and suppress HSC activation. In summary, our results propose that NMN supplementation is a new therapeutic approach for liver fibrosis prevention. The removal of organic pollutants was examined by new zeolitic imidazolate framework-8/carboxyl-functionalized multi-walled carbon nanotubes (ZIF-8/MWCNTs-COOH) nanoadsorbents, which were prepared via controlling the reaction sequence of MWCNTs-COOH with the precursors of ZIF-8. Structural morphol. and chem. composition characterizations confirmed the successful preparation of ZIF-8/MWCNTs-COOH nanoadsorbents. The obtained samples manifested a preferable adsorption capacity for Congo red with the maximum adsorption capacity of 1185.58 mg g-1 with an adsorption removal rate of 97.93% at equilibrium adsorption. Further analyses indicated that the adsorption process conformed to the pseudo-second-order kinetic model and Langmuir adsorption isotherm. The thermodn. analyses were depicted as endothermic, entropy-increasing, and spontaneous. The effects of pH value, interfering ions, and dosage on the adsorption properties were systematically explored. The nanoadsorbent can be expediently reused for eight cycles without a prominent reduction in adsorption activity. The improved performance and high reusability can be ascribed to the increasing sp. surface, the presence of electrostatic interaction, hydrogen bonding, and π-π interaction between ZIF-8/MWCNTs-COOH and organic mols. In addition, the prepared composites also showed a better adsorption performance for tetracycline with the equilibrium adsorption capacity of 471.22 mg g-1 compared to pure ZIF-8 and MWCNTs-COOH. Chondrosarcoma is the second most common primary bone sarcoma. Treatment of chondrosarcoma is limited to surgery due to radiation and chemotherapy resistance of this cancer. An ideal treatment for chondrosarcoma would be a well-tolerated, minimally invasive local or systemic treatment modality to halt or slow tumor growth prior to resection of local, unresectable local, or metastatic disease. Palovarotene, an agonist of nuclear retinoic acid receptor γ (RARγ) has shown therapeutic action for treatment of heterotopic ossification and osteochondroma without serious adverse effects in animal models. We hypothesized that selective agonists of RARγ would have an inhibitory effect on chondrosarcoma. All human chondrosarcoma specimens expressed RARγ as determined by immunohistochem. staining. The ΗCS-2/8 chondrosarcoma cell line, established from low-grade human chondrosarcoma, was used to examine the actions of RARγ agonists. In ΗCS2/8 pellet cultures, RARγ agonist treatment reduced the mass size and significantly decreased total glycosaminoglycan, protein amounts, and gene expression levels of cartilage matrix mols. when compared with control groups. Systemic treatment with RARγ agonists significantly inhibited the growth of ΗCS-2/8 cell transplants in vivo. Furthermore, local injection of RARγ agonist-loaded poly-lactic acid nanoparticles induced regression of the mass size of the transplants. Histol. anal. demonstrated that RARγ agonist treatment inhibited cell proliferation activity and stimulated encapsulation of the tumor. These findings indicate that RARγ agonists, including palovarotene, may have an anti-tumor effect on low-grade chondrosarcomas.
