Repeated-dose toxicity (RDT) studies are one of the critical studies to assess chem. safety. There have been some studies attempting to predict RDT endpoints based on chem. substructures, but it remains very difficult to establish such a method, and a more detailed characterization of chem. compounds seems necessary. Cytochrome P450s (P450s) comprise multiple forms with different substrate specificities and play important roles in both the detoxification and metabolic activation of xenobiotics. In this study, we investigated possible use of P 450 reactivity of chem. compounds to classify the compounds A total of 148 compounds with available rat RDT test data were used as test compounds and subjected to inhibition assays against 18 human and rat P450s. Among the tested compounds, 82 compounds inhibited at least one P 450 form. Hierarchical clustering analyses using the P 450 inhibitory profiles divided the 82 compounds into nine groups, some of which showed characteristic chem. and biol. properties. Principal component analyses of the P 450 inhibition data in combination with the calculated chem. descriptors demonstrated that P 450 inhibition data were plotted differently than most chem. descriptors in the loading plots. Finally, association analyses between P 450 inhibition and RDT endpoints showed that some endpoints related to the liver, kidney and hematol. were significantly associated with the inhibition of some P450s. Our present results suggest that the P 450 reactivity profiles can be used as novel descriptors for characterizing chem. compounds for the investigation of the toxicity mechanism and/or the establishment of a toxicity prediction model. Polyurethane materials (PUs) have been widely used in industry and daily life due to the versatile chem. However, despite the rapid advance in synthetic chem., it still remains a significant challenge for the facile fabrication of PUs with a single crosslinked network embedded with excellent mech. properties and recyclability. Herein, in this study, we proposed a simple strategy to fabricate a high-performance recyclable crosslinked PU and revealed the relationship between microscopic structure and macroscopic properties. The UPy (2-ureido-4-[1H]-pyrimidione) motifs were incorporated into the backbone chains of PUs, where the quadruple hydrogen bonding interactions between UPy dimers can significantly enhance the mech. strength and toughness. Furthermore, a single small mol. Diels-Alder adduct was utilized as the chem. crosslinker, rendering the final crosslinked PUs healable and recyclable. The thermal reversibility of the Diels-Alder reaction was well verified by DSC and solid-state NMR spectroscopy. Notably, it was found that the incorporation of UPy motifs could enhance the strain-induced crystallization (SIC), leading to a large stress at break. The structural and dynamic changes induced by SIC were quant. addressed by proton multiple-quantum NMR spectroscopy and SAXS experiments, where SIC further imposed restrictions on the mobility of surrounding polymer chains in the soft domain and led to the change of microphase separated nanostructures. Overall, a simple strategy is proposed here for the facile fabrication of high performance recyclable PUs, and the detailed investigation here on the structure-property relationship may further provide insights into developing high performance polymeric materials. Herein, a strategy to design a “hard-soft” hard domain by the cooperation of quadruple hydrogen bonds (HBs) in the mainchain as restriction units and single HBs in the side chain as diffusion units is reported. The resulting transparent supramol. elastomer exhibited fast self-recoverability, good puncture resistance and superior mech. properties with a tensile strength of 20.5 MPa, an extensibility of 2043.7%, a toughness of 146.1 MJ m-3 and a tear resistance of 13.8 kJ m-2.
