The transient receptor potential vanilloid type 4 (TRPV4) is a polymodal receptor. Antagonists of human TRPV4 (hTRPV4) represent a novel therapeutic approach for acute lung injury (ALI). However, the discovery of various hTRPV4 antagonists has been difficult due to the unavailability of 3D-structure of hTRPV4 protein. We constructed the 3D-structure of hTRPV4 protein by homol. modeling, and the binding pocket of antagonist with hTRPV4 was predicted for the first time. The pocket was consistent with the same subfamily rabbit TRPV5. The detailed interactions of different protein-ligand complexes were calculated by mol. docking and mol. dynamics (MD) simulation, and the outcome revealed the rationality of the binding pocket. Based on the docking and MD results of this model and the structure of compound A2, a TRPV4 antagonist reported in literature, two small mol. compounds, B1 and B2, were designed and synthesized as hTRPV4 antagonists. The results of biol. evaluation in vitro showed that these compounds have good inhibitory activity on hTRPV4. Moreover, the results were in good agreement with those predicted by mol. simulation, which in turn suggested that the modeling 3D structure and the predicted active sites of hTRPV4 are reasonable and reliable. Agricultural microplastics (MPs) pollution resulting from the widespread and extensive use of polymer-coated fertilizer (PCF) is of great concern. However, little information is currently available concerning the influence of MPs derived from PCF (PCF-MPs) on the crop-soil system. Therefore, we investigated the impacts of PCF-MPs (0, 0.01, 0.1, 1% weight/weight) on the growth of maize cultivars (ZNT 488 & ZTN 182), rhizosphere metabolites, microbial community, and soil properties. The effects of PCF-MPs on plant growth varied with plant cultivars and exposure doses. MPs had no influence on the growth of ZNT 488, but significantly improved the plant performance of ZTN 182 at higher doses. PCF-MPs insignificantly altered the rhizosphere metabolite profile of two maize cultivars. Although soil PAEs contents and the microbial communities were unchanged, the phyla Fibrobacteres and Verrucomicrobia (associated with plant growth-promotion) were increased significantly in ZTN 182. Moreover, PCF-MPs stimulated the soil urease, acid phosphatase and nitrate reductase activities, especially the phytase activity in ZTN 182. Besides, significant alterations in soil properties were also discovered in a cultivar-dependent manner. Overall, our study highlighted that the potential environmental risk assessment of PCF-MPs is still needed, even though PCF-MPs did not show neg. effect on maize growth and rhizosphere metabolic profiles. Despite lipid nanoparticles′ (LNPs) success in the effective and safe delivery of mRNA vaccines, an inhalation-based mRNA therapy for lung diseases remains challenging. LNPs tend to disintegrate due to shear stress during aerosolization, leading to ineffective delivery. Therefore, LNPs need to remain stable through the process of nebulization and mucus penetration, yet labile enough for endosomal escape. To meet these opposing needs, we utilized PEG lipid to enhance the surficial stability of LNPs with the inclusion of a cholesterol analog, β-sitosterol, to improve endosomal escape. Increased PEG concentrations in LNPs enhanced the shear resistance and mucus penetration, while β-sitosterol provided LNPs with a polyhedral shape, facilitating endosomal escape. The optimized LNPs exhibited a uniform particle distribution, a polyhedral morphol., and a rapid mucosal diffusion with enhanced gene transfection. Inhaled LNPs led to localized protein production in the mouse lung without pulmonary or systemic toxicity. Repeated administration of these LNPs led to sustained protein production in the lungs. Lastly, mRNA encoding the cystic fibrosis transmembrane conductance regulator (CFTR) was delivered after nebulization to a CFTR-deficient animal model, resulting in the pulmonary expression of this therapeutic protein. The present study was conducted to determine the characterization and antibacterial activity of peppermint essential oil-loaded solid lipid nanoparticle (PEO-SLN) and its impact on the quality of trout fillet stored at 4 ± 1°C for 12 days. The SLNs were prepared through a bath sonication technique. PEO-SLNs contained 0.2% (w/v) PEO in 2% of lipid phase glycerol monostearate (GMS) and tween 80 (1% w/v) used as a surfactant in the aqueous phase. The characterization parameter of PEO-SLN was evaluated, and the antibacterial activity of PEO-SLNs was conducted under in vitro conditions. Trout samples were analyzed for inoculated Pseudomonas aeruginosa, Listeria monocytogenes, and Escherichia coli O157:H7 during refrigerated storage. The mean particle size of PEO-SLNs was 154.83 ± 1.21 nm with a polydispersity index (PDI) of 0.35 ± 0.01 and zeta potential was about -24.16 ± 0.51 mV. The results indicated that PEO-SLN had higher antibacterial activity than the free form of PEO and also when used in combination with gelatin coating (gel + PEO-SLN) had a significant effect on preventing microbial growth in trout fillets (p < 0.05). The most decreasing rate of P. aeruginosa (1.92 log CFU/g), E. coli O157:H7 (0.71 log CFU/g), and L. monocytogenes count (1.69 log CFU/g) was seen in gel + PEO-SLN. These findings illustrated that PEO-SLNs could potentially be utilized in the food industry to increase the shelf life of fish fillets. ‘Marselan is a promising wine grapevine with unique qualities, studies on its introduction and characteristics have never stopped. The aims of this paper are to introduce a method to classification of fish sauce origin by means of electronic nose fingerprint and gas chromatog.- mass spectrometry of volatile compounds and the two artificial neural networks are used to predict the origins of fish sauce. Methods: Headspace sampling-solid phase microextraction combined with gas chromatog.-mass spectrometric anal. and electronic nose were used to analysze volatile compounds in different origins of fish sauce, and these dates predicted the origins of fish sauce by artificial neural networks. Results: 94 volatile compounds were identified by Automatic mass spectral deconvolution and identification system, out of which 44 are from Guangdong, 53 from our laboratory, 51 from Vietnam, 47 and 45 from Thailand. Then electronic nose was applied to identify the origin of fish sauce, and the data were analyzed using principal component anal. and load anal. Lastly, two artificial neural networks are used to predict the origins of fish sauce, and the accuracy rates of radial basis and gradient descent both are 93.33%. Conclusion: That illustrates that we can provide a quick method to distinguish fish sauce products of different origins. These results indicated that the combinations of multiple anal. and identification methods could make up the limitations of a single method, enhance the accuracy of identification, and provide useful information for product development.
