Repaglinide, a member of the meglitinide class of drugs, is a new anti-diabetic agent that is utilized as an oral hypoglycemic agent. Cytokines are important immunotherapeutics with approved drugs for the treatment of human cancers. However, systemic administration of cytokines often fails to achieve adequate concentrations to immune cells in tumors due to dose-limiting toxicity. Thus, developing localized therapy that directly delivers immune-stimulatory cytokines to tumors may improve the therapeutic efficacy. In this study, we generated novel lipid nanoparticles (LNPs) encapsulated with mRNAs encoding cytokines including IL-12, IL-27 and GM-CSF, and tested their anti-tumor activity. We first synthesized ionizable lipid materials containing di-amino groups with various head groups (DALs). The novel DAL4-LNP effectively delivered different mRNAs in vitro to tumor cells and in vivo to tumors. Intratumoral injection of DAL4-LNP loaded with IL-12 mRNA was most potent in inhibiting B16F10 melanoma tumor growth compared to IL-27 or GM-CSF mRNAs in monotherapy. Furthermore, intratumoral injection of dual DAL4-LNP-IL-12 mRNA and IL-27 mRNA showed a synergistic effect in suppressing tumor growth without causing systematic toxicity. Most importantly, intratumoral delivery of IL-12 and IL-27 mRNAs induced robust infiltration of immune effector cells, including IFN-γ and TNF-α producing NK and CD8+ T cells into tumors. Thus, intratumoral administration of DAL-LNP loaded with IL-12 and IL-27 mRNA provides a new treatment strategy for cancer. SBA-15 with a large pore size and a high sp. surface area was suitable as catalyst carrier, and the average diameter of Pb-Bi oxide particles was ∼3.57 nm. The addition of Bi led to the improved dispersion of active components, which were highly and evenly dispersed on the SBA-15 surface. Furthermore, an appropriate proportion of Pb and Bi on SBA-15 was conducive to the considerable enhancement of the catalytic activity. Besides, Pb-Bi/SBA-15 also exhibited good durability and recyclability, making it a promising catalytic material for the synthesis of various carbonates. The qRT-PCR and enzyme activity assay revealed that the transcription and expression of GhPEPC1 was dramatically down-regulated in transgenic lines. Consequently, the cottonseed oil content in several transgenic lines showed a significant (P < 0.01) increase (up to 16.7%) without obvious phenotypic changes under filed condition when compared to the control plants. In order to elucidate the mol. mechanism of GhPEPC1 in the regulation of seed oil content, we quantified the expression of the carbon metabolism related genes of transgenic GhPEPC1 RNAi lines by transcriptome anal. This anal. revealed the decrease of GhPEPC1 expression led to the increase expression of triacylglycerol biosynthesis-related genes, which eventually contributed to the lipid biosynthesis in cotton. In modern therapy, cutting side effects caused by the administration of drugs is one of the core challenges. One possible strategy is the delivery of a drug to the disease site by encapsulation into a polymeric matrix. Especially, light-responsive polymers are regarded as promising materials due to, e.g., the capability of tailored on-demand release in illuminated areas. In this work, a series of light-responsive backbone-degradable (co-)polymers were synthesized by polycondensation and polyaddition All obtained polymers showed rapid degradation in solution upon exposure to UV light as observed by size-exclusion chromatog. Depending on the incorporated comonomers, the functional co-polyurethanes exhibited either enhanced hydrophilicity or dual-responsiveness to light and redox environments, which was detected by SEC after treatment with a reducing agent. The formulation of nanoparticles from light-responsive polyurethanes proved the processability of the synthesized polymers, and dynamic light scattering (DLS) measurements confirmed the photo-induced degradation of the prepared particles. This work addresses the mol. thermodn. and artificial neural network (ANN) modeling of surface tensions of several fatty acid esters and biodiesels. Two biodiesels were considered as pure fluid and the other as a binary mixture The mol. thermodn. model is based on the statistical mech. expression according to Fowler-Kirkwood-Buff approximation Regarding this, contributions to surface tension from the hard-chain repulsions, Lennard-Jones dispersion forces, and dipolar interactions were considered and assumed to be additive in the model development. The mol. thermodn. model used three mol. parameters reflecting the hard-core diameter, dispersive energy and segment number as well as the liquid densities for which the values were predicted from perturbed Yukawa-chain equation of state. Further, the model used dipole moment as an adjustable parameter for the accurate calculation of surface tensions. The model could predict 149 surface tension data points for 9 FAEs and 3 biodiesels in 268.6-393 K range with the average absolute relative deviation (AARD) of 1.82%. The degree of accuracy of proposed model has also been compared with some empirical equations. Concerning ANN modeling, a network comprising two hidden layers and 9 neurons for each layer has been trained, according to the constructive approach. The result of the training was quite good, the AARD of the pure fluid dataset of 137 points was found to be 0.44%.
