Lasia spinosa (L.) Thwaites is a widely used ethnomedicinal plant in Bangladesh. In this work, we report the effect of different light emitting diodes (LED) on induction, metabolite production, and antioxidant properties of calluses derived from stem and leaf segments of P. setacea cv BRS Peŕola do Cerrado. Friable calluses were induced on MSM medium supplemented with picloram, under different light qualities (white fluorescent lamps or different types of LEDs) or in the dark. Light quality contributed more significantly for biomass accumulation than the other variables. Callus formation was observed in response to all treatments, although the highest biomass accumulation was induced by red LED. Chromatog. analyses indicated that blue LED induced the highest production of bioactive substances. All samples displayed low antioxidant potential by the DPPH assay, but showed a high capacity to chelate iron. The activity of antioxidant enzymes was also evaluated. The incubation under red LED caused an increase on the activity of superoxide dismutase, catalase and ascorbate peroxidase in calluses derived from internodal segments induced under red LED. This is the first study evaluating the effect of lighting conditions in in vitro systems of P. setacea cv BRS Peŕola do Cerrado, thus opening new possibilities of study and utilization. Retinal gene therapy has had unprecedented success in generating treatments that can halt vision loss. However, immunogenic response and long-term toxicity with the use of viral vectors remain a concern. Non-viral vectors are relatively non-immunogenic, scalable platforms that have had limited success with DNA delivery to the eye. MRNA (mRNA) therapeutics has expanded the ability to achieve high gene expression while eliminating unintended genomic integration or the need to cross the restrictive nuclear barrier. Lipid-based nanoparticles (LNPs) remain at the forefront of potent delivery vectors for nucleic acids. Herein, we tested eleven different LNP variants for their ability to deliver mRNA to the back of the eye. LNPs that contained ionizable lipids with low pKa and unsaturated hydrocarbon chains showed the highest amount of reporter gene transfection in the retina. The kinetics of gene expression showed a rapid onset (within 4 h) that persisted for 96 h. The gene delivery was cell-type specific with majority of the expression in the retinal pigmented epithelium (RPE) and limited expression in the Muller glia. LNP-delivered mRNA can be used to treat monogenic retinal degenerative disorders of the RPE. The transient nature of mRNA-based therapeutics makes it desirable for applications that are directed towards retinal reprogramming or genome editing. Overall, non-viral delivery of RNA therapeutics to diverse cell types within the retina can provide transformative new approaches to prevent blindness. The thermal behavior of Zn(II) acetylsalicylate [Zn(acsa)2(H2O)2] with respect to phase transitions, pyrolysis both in air and inert (N2) atm., and product identification was studied. The complex was synthesized by metathesis in hot EtOH solution using aspirin (acetylsalicylic acid) as precursor and characterized via electronic and IR spectral analyses. Optical observations showed that the white salt does not undergo a direct transition from the solid to the liquid phase but rather goes slowly through an intermediate mesophase around 80° before melting rapidly to the brick-brown isotropic liquid around 134-136°. No liquid crystalline phases are however formed. This result was complemented by that from thermogravimetric (TG) studies in the ∼25-600° range, which showed three main weight-loss phases of 8.0, 50.0 and 14.0% (around 200, 250 and 400°) corresponding, resp., to the elimination of CO2, xanthone and HOAc. The pyrolysis products, as identified using a combination of instrumental (GC-MS) and wet chem. techniques are: CO2; nonstoichiometric Zn oxide, most likely in the form: Zn1+xO (0.0000 ≤ x ≤ 0.0003); and a mixture of organic products resulting from further decomposition, charring and other attendant thermal effects at the relatively high temperatures (∼600°) involved. Six of the principal organic products were identified and included salsalate and benorylate which are pro-drugs of salicylic acid, a known pharmaceutical.
