Copolymers of pentafluorophenylhexafluoroisopropyl methacrylate (FPPMA) with trifluoroethyl methacrylate (TFEMA) were prepared in THF solution and in bulk using azobisisobutyronitrile as a free radical initiator. The monomer reactivity ratios of TFEMA (M1) and FPPMA (M2) were calculated as r1 = 0.55 and r2 = 0.07. The refractive indexes of poly(TFEMA) and poly(FPPMA) are very similar as 1.435 and 1.430, resp., at 532 nm, and the copolymer films were transparent. Abstract: Increasing resistance in antibiotic and chemotherapeutic treatments has been pushing studies of design and evaluation of bioactive peptides. Designing relies on different approaches from minimalist sequences and endogenous peptides modifications to computational libraries. Evaluation relies on microbiol. tests. Aiming a deeper understanding, we chose the octapeptide Jelleine-I (JI) for its selective and low toxicity profile, designed small modifications combining the substitutions of Phe by Trp and Lys/His by Arg and tested the antimicrobial and anticancer activity on melanoma cells. Biophys. methods identified environment-dependent modulation of aggregation, but critical aggregation concentrations of JI and analogs in buffer show that peptides start membrane interactions as monomers. The presence of model membranes increases or reduces the partial aggregation of peptides. Compared to JI, analog JIF2WR shows the lowest tendency to aggregation on bacterial model membranes. JI and analogs are lytic to model membranes. Their composition-dependent performance indicates preference for the higher charged anionic bilayers in line with their superior performance toward Staphylococcus aureus and Streptococcus pneumoniae. JIF2WR presented the higher partitioning, higher lytic activity and lower aggregated contents. Despite these increased membranolytic activities, JIF2WR exhibited comparable antimicrobial activity in relation to JI at the expenses of some loss in selectivity. Busulfan-based conditioning is the most commonly used high-dose conditioning regimen for allogeneic hematopoietic cell transplant (HCT). The alkylating agent busulfan has a narrow therapeutic index, with busulfan doses personalized to a target plasma exposure (targeted busulfan). Using a global pharmacometabonomics approach, we sought to identify novel biomarkers of relapse or acute graft vs. host disease (GVHD) in a cohort of 84 patients receiving targeted busulfan before allogeneic HCT. A total of 763 endogenous metabolomic compounds (EMCs) were quantitated in 230 longitudinal blood samples before, during, and shortly after i.v. busulfan administration. We performed both univariate linear regression and pathway enrichment analyses using global testing. The cysteine/methionine pathway and the glycine, serine, and threonine metabolism pathway were most associated with relapse. The latter be explained by the fact that glutathione S-transferases conjugate both busulfan and glutathione, which contains glycine as a component. The D-arginine and D-ornithine metabolism pathway and arginine and proline metabolism pathway were most associated with acute GVHD. None of these associations were significant after correcting for false discovery rate (FDR) with a strict cutoff of FDR-adjusted p < 0.1. Although larger studies are needed to substantiate these findings, the results show that EMCs may be used as predictive biomarkers in HCT patients. D limonene (L) is a natural phenolic compound with high antioxidant and antimicrobial activity. However, poor solubility and high volatility limit its applications in the food and pharmaceutical industries. In this study, L was incorporated in β-cyclodextrin nanosponges (CD-NS) to overcome these limitations. For preparation of NS, different molar ratios of β-CD and di-Ph carbonate (DPC) as crosslinker were used. The synthesized NSs were characterized by FTIR, SEM, TGA, and XRD. The release of encapsulated L was evaluated in different food simulants and simulated gastrointestinal conditions. The encapsulation of L in NS was confirmed by FTIR. The L-NS showed cubic structures with an average size around 1528 ± 195 nm. The solubility and thermal stability of L were enhanced after encapsulation in NS. Encapsulated L showed higher antibacterial activity compared to free L and min. inhibitory concentration (MIC) of free L was decreased significantly after encapsulation in CD-NS. L indicated a controlled release in gastrointestinal conditions and different food simulants. Our results propose that CD-NS is a suitable carrier for hydrophobic and sensitive compounds and L-NS can be used as a potential preservative with enhanced antibacterial activity in food applications. It is important to note that the results of this study were obtained in vitro and further studies related to their toxicity (in vivo) are needed for confirmation of their application in the era of nutrition and genomics. Pears (Pyrus communis L.) cv. Packham′s Triumph are very traditional for human consumption, but pear is a highly perishable climacteric fruit with a short shelf-life affected by several diseases with a microbial origin. In this study, a protective effect on the quality properties of pears was evidenced after the surface application of chitosan-Ruta graveolens essential oil coatings (CS + RGEO) in four different concentrations (0, 0.5, 1.0 and 1.5 %, volume/volume) during 21 days of storage under 18 °C. After 21 days of treatment, a weight loss reduction of 10% (from 40.2 ± 5.3 to 20.3 ± 3.9) compared to the uncoated pears was evident with CS + RGEO 0.5%. All the fruits′ phys.-chem. properties evidenced a protective effect of the coatings. The maturity index increased for all the treatments. However, the pears with CS + RGEO 1.5% were lower (70.21) than the uncoated fruits (98.96). The loss of firmness for the uncoated samples was higher compared to the coated samples. The pears′ most excellent mech. resistance was obtained with CS + RGEO 0.5% after 21 days of storage, both for compression resistance (7.42 kPa) and force (22.7 N). Microbiol. studies demonstrated the protective power of the coatings. Aerobic mesophilic bacteria and molds were significantly reduced (in 3 Log CFU/g compared to control) using 15 μL/mL of RGEO, without affecting consumer perception. The results presented in this study showed that CS + RGEO coatings are promising in the post-harvest treatment of pears. Using these fragmented ions, we characterized individual and isomeric species in each class of mitochondrial phospholipids, including unusual species, such as PS, containing an ether linkage and species containing odd-numbered acyl chains in cardiolipin, PS, PI, and PG. We also characterized monolysocardiolipin and dilysocardiolipin, the least abundant but nevertheless important mitochondrial phospholipids. The results clearly show the power of HPLC-MS/MS for identification and characterization of phospholipids, including minor species. Volcanic deposits increase soil organic carbon storage. However, little is known about the effect of volcanic deposits on forest soil nitrogen (N) dynamics and microbial communities. We explored gross and net N transformation rates and microbial community structure using a phospholipid fatty acid (PLFA) method across eight forests with soils derived from different parent material in Japan. Volcanic mineral soils had approx. three-fold greater total N and inorganic N contents and gross nitrification, ammonium immobilization, and nitrate immobilization rates that were one order of magnitude higher than in non-volcanic soils. Moreover, volcanic mineral soils had a 1.7-fold lower N turnover rate, which was estimated as net N mineralization rate per soil N. This was likely caused by a higher gross N immobilization rate, potentially due to the mineral traits of volcanic soils. Volcanic mineral soils had approx. four- and three-fold greater microbial biomass-N and bacterial PLFA contents, resp., and the fungal:bacterial PLFA ratio was lower in volcanic mineral soils than in non-volcanic soils. Microbial community structure, analyzed using non-metric multidimensional scaling, was distinct between volcanic and non-volcanic soils, and was significantly affected by soil characteristics. This study demonstrates that aside from soil carbon storage, volcanic deposits are related to increases in soil N immobilization, N content, bacterial biomass, and N transformation rates in forest ecosystems.
