AL amyloidosis is a life-threatening disease characterized by the deposition of amyloidogenic Ig light chain secreted from clonal plasma cells. Here we established an in-vitro screening system of amyloid inhibition of a variable domain in λ6 light chain mutant (Vλ6), Wil, and screened a food-additive compound library to identify compounds inhibiting the fibril formation. We found gossypetin and isoquercitrin as novel inhibitors. NMR anal. showed that both compounds directly interacted with natively-folded Wil, and proteolysis experiments demonstrated that these compounds conferred proteolytic resistance, suggesting that the compounds enhance the kinetic stability of Wil. Since gossypetin and isoquercitrin specifically interacted with the protein at micromolar concentrations, these compounds could be used as lead to further develop inhibitors against AL amyloidosis.

The photodissociation of several N,N-diethyldithiocarbamate derivatives (RDC), i.e., benzyl, 1-phenylethyl, 2-phenylethyl, Bu, sec-, and tert-Bu N,N-diethyldithiocarbamates, was investigated by the spin trapping technique to elucidate the polymerization mechanism of styrene (I) with RDC as iniferters. The scission of RDC occurred at two different C-S bonds depending on the structure of the alkyl groups of RDC and the cleavage manner was dominated by both resonance and steric effects. RDC generating an N,N-diethyldithiocarbamyl radical acts as an effective iniferter and induces the living radical polymerization of I in homogeneous system. The polymerizations of I with tetraethylthiuram disulfide and p-xylylene bis(N,N-diethyldithiocarbamate) were also discussed on the basis of the results of the spin trapping for RDC as the model compounds of the polystyrene chain ends.

A segmented polyurethane (SPU) film was chloromethylated and subsequently dithiocarbamated. The treated films were immersed in solutions containing poly(ethylene glycol) methacrylate (PEGMA) or N,N-dimethylacryl amide (DMAAm) and irradiated with UV light. The resultant surfaces were highly wettable with water. The surface chem. compositions, as determined by XPS, indicated that surface graft polymerization, initiated from surface-derivatized dithiocarbamate, had occurred and had altered the surface properties and the composition of SPU. The platelet adhesion test using platelet-rich plasma showed minimal adhesion for the poly(PEGMA)-grafted surface, followed by the poly(DMAAm)-grafted surface.

A series of asym. bent-core liquid crystals I (X = COO; Y = H, Cl), bearing the stilbene moiety in the side wing, was synthesized and characterised. Structural modifications of the non-stilbene wing resulted in formation of various mesophases. Compounds I (X = COO; Y = H) and I (X = COO; Y = Cl) with an ester linking group between the Ph rings in this wing have an enantiotropic non-polar smectic-type mesophase, while compound I (Y = H), incorporating the biphenyl moiety, has a ferroelec. SmCaPF phase. The mols. of I (Y = H) undergo photoisomerisation in solution under UV irradiation, while the mesophases are unstable and UV irradiation leads to irreversible changes. The mesophases of I (X = COO; Y = H) and I (X = COO; Y = Cl) do not show any electro-optical switching and polarisation current. Applying low frequency AC voltage on a planarly aligned film of compound I (Y = H), smectic domains grow, while the birefringence and an image flickering increase. The observed features of these compounds identify stilbene as an attractive moiety in the design of advanced materials based on bent-core mols.

Early lymphopenia is a known side effect of di-Me fumarate, a disease-modifying therapy for MS. However, the long-term effects on immune response and the impact on lymphocyte counts when another disease-modifying treatment is introduced, remain unknown. To better understand these specific aspects, we reviewed cases that develop prolonged grade 2 to 4 lymphopenia under di-Me fumarate in Lausanne MS clinic. Retrospective anal. of the 12 patients (10.1%) who discontinued di-Me fumarate because of prolonged lymphopenia amongst the 119 patients treated with di-Me fumarate. We reviewed their demographics, as well as their clin., biol. and MRI characteristics compared to the non-lymphopenic patients, during di-Me fumarate therapy, and within a timeframe of up to 18 mo after switching to another disease-modifying treatment. We also focused on lymphocyte subsets in a subgroup of patients. Compared to non-lymphopenic patients, lymphopenic patients were older at di-Me fumarate initiation (51.4 yo vs 39.7, p = 0.0003) and the majority were male (p = 0.037). Three of them (25%) developed delayed lymphopenia, more than one year after treatment onset. Despite persistent lymphopenia, three patients experienced disease activity. Amongst the nine patients (75%) who were switched to another therapy, five (55.6%) presented recurrent lymphopenia, predominantly with a CD8+ T cell decrease. Di-Me fumarate has a long-term impact on lymphocyte biol., even after its discontinuation, with a sustained reduction in CD8+ T cells that may increase opportunistic infection risk, and should be taken in consideration when switching therapies after di-Me fumarate.

The cetane number, a widely used diesel fuel quality parameter related to the ignition delay time (and combustion quality) of a fuel, has been applied to alternative diesel fuels such as biodiesel and its components. In this work, the cetane numbers of 29 samples of straight-chain and branched C1-C4 esters as well as 2-ethylhexyl esters of various common fatty acids were determined The cetane numbers of these esters are not significantly affected by branching in the alc. moiety. Therefore, branched esters, which improve the cold-flow properties of biodiesel, can be employed without greatly influencing ignition properties compared to the more common Me esters. Unsaturation in the fatty acid chain was again the most significant factor causing lower cetane numbers Cetane numbers were determined in an ignition quality tester (IQT) which is a newly developed, automated rapid method using only small amounts of material. The IQT is as applicable to biodiesel and its components as previous cetane-testing methods.

We investigated the relationship between the self-assembled morphol. of poly(tert-Bu acrylate)-block-poly(6-[4-(4′-methoxyphenylazo)phenoxy]hexyl methacrylate) (PtBA-b-PAzoMA) block copolymers and their photoresponsive and fluorescence behaviors. The morphol. of PtBA-b-PAzoMA copolymers was manipulated by dissolving them in mixed DMF (DMF)/hexanol solvents. When PtBA-b-PAzoMA was dissolved in DMF-rich (neutral) solvents, a favorable interaction between the DMF mols. and both blocks resulted in a random-coiled conformation. The unconfined morphol. facilitated the formation of both nonassocd. and head-to-head organized azobenzene mesogens, which promoted fluorescence emission. When hexanol, a PtBA-selective solvent, was added to DMF, the solvency of PtBA-b-PAzoMA worsened, leading to its assembly into micelles, with PAzoMA in the micelle core. The confinement of azobenzene moieties in the micelle core hindered their trans-to-cis photoisomerization, thereby considerably decreasing the kinetics of photoisomerization and the population of cis isomers. Addnl., a nanoconfined geometry resulted in compactly packed chromophores, causing fluorescence loss. When PtBA-b-PAzoMA was exposed to UV light, the increased number of cis isomers hampered the closely packed mesogens, resulting in a substantial enhancement of fluorescence emission. When the mole fraction of the PAzoMA block was increased, PtBA-b-PAzoMA formed clusters, causing the slow kinetics of photoisomerization and fluorescence quenching.

Nano-structured hyperbranched cationic star polymers, called star vectors, were molecularly designed for a novel gene delivery non-viral vector. The linear and 3, 4 or 6 branched water-soluble cationic polymers, which had same mol. weight of ∼18,000, were synthesized by iniferter (initiator-transfer agent-terminator)-based photo-living-radical polymerization of 3-(N,N-dimethylamino)propyl acrylamide, initiated from resp. multi-dithiocarbamate-derivatized benzenes as an iniferter. All polymers produced polyion complexes ‘polyplexes’ by mixing with pDNA (pGL3-control plasmid), in which the particle size was ∼250 nm in diameter [the charge ratio < 2/1 (vector/pDNA)] and ∼150 nm (the charge ratio > 2.5/1), and the ζ-potential was ∼+10 mV (the charge ratio > 1/1). When COS-1 cells were incubated with the polyplexes 12h after preparation under the charge ratio of 5/1, higher gene expression was obtained as an increase in branching, with a little cytotoxicity. The relative gene expression to the linear polymer was about 2, 5, and 10 times in 3-, 4-, and 6-branched polymers, resp. The precise change in branching of polymers enabled the control of the gene transfer activity.
