Small ribozymes such as Oryza sativa twister spontaneously cleave their own RNA when the ribozyme folds into its active conformation. The coupling between twister folding and self-cleavage has been difficult to study, however, because the active ribozyme rapidly converts to product. Here, we describe the synthesis of a photocaged nucleotide that releases guanosine within microseconds upon photosolvolysis with blue light. Application of this tool to O. sativa twister achieved the spatial (75 μm) and temporal (â‰?0 ms) control required to resolve folding and self-cleavage events when combined with single-mol. fluorescence detection of the ribozyme folding pathway. Real-time observation of single ribozymes after photo-deprotection showed that the precleaved folded state is unstable and quickly unfolds if the RNA does not react. Kinetic anal. showed that Mg2+ and Mn2+ ions increase ribozyme efficiency by making transitions to the high energy active conformation more probable, rather than by stabilizing the folded ground state or the cleaved product. This tool for light-controlled single RNA folding should offer precise and rapid control of other nucleic acid systems.

A facile method for preparing supramol. waterborne polyurethane (WPU) based on quadruple hydrogen bonds (4H-WPU) is reported. Herein, 4H-WPU with quadruple hydrogen bonds in main chain were synthesized by poly (1,4-butylene-neopentylene adipate glycol) as soft segment, 2-ureido-4[1H]-pyrimidinone (UPy) functionalized monomer, isophorone diisocyanate, 2,2-Bis(hydroxymethyl)propionic acid (hydrophilic monomer), isophoronediamine, triethylamine (neutralizer), and monoethanolamine (blocking agent) as hard segment. The mol. weight of 4H-WPU was controlled around 18,000 consistently. The properties of 4H-WPU with different content of hydrophilic group, hard segment, and UPy units were characterized and the results could provide the reference for preparing supramol. WPU with high mech. and self-healing performance while maintaining dispersion stability. The tensile strength of 4H-WPU was 10 times of the blank sample. The healing time of scratched 4H-WPU film was inversely proportional to the content of UPy-functionalized monomer and the shortest healing time is 2.5 h at 80°C. Mech. performance of healed films can be restored to more than 90%.

Gypsophila paniculata is an ornamental crop with medicinal value. To date, limited information has been reported about the natural products in G. paniculata to explain its medicinal function. The current study reports the natural products found in G. paniculata stem for the first time. Thirty-three compounds were isolated from the extract of G. paniculata stem and identified by gas chromatog.-mass spectrometry, 10 of which have contents >2%. These were 2-O-methyl-D-mannopyranose (37.4706%), glycerol (12.5669%), two tetratetracontane isomer (7.6523 + 3.5145%), tetrahygro-4-pyranol (5.3254%), 1,6-anhydro-beta-d-glucopyranos (4.7507%), palmitic acid (4.1848%), 4-hydroxy-3-methoxystyrene (3.7439%), methyl-octadeca-9,12-dienoate (2.7490%), and 2-deoxy-D-galactose (2.6193%). Another bioactive compound, condrillasterol, was identified with 1.3384% content. We also reported that G. paniculata possesses antioxidant activity possibly associated with the presence of a phenolic chem. 4-hydroxy-3-methoxystyrene. Our data collectively demonstrate that G. paniculata contains some bioactive compounds with high contents and antioxidants, consistent with its role as a medicinal herb.

Nitrosyl triflate (TfONO) can be generated in situ from tetra-n-butylammonium nitrite and triflic anhydride (1:1) in CH2Cl2 solution at ca. -30°C. It acts as a powerful and soluble nitrosating agent with a wide range of olefinic or aromatic substrates. Nitrous anhydride (O=N-O-N=O) can be generated in the same way using tetra-n-butylammonium nitrite and triflic anhydride (2:1) in CH2Cl2 solution at ca. -30°C. Each reagent has been isolated and characterized. They react with olefins to give different products. Nitrosyl triflate is an excellent reagent for generating either vinylic or allylic nitroso compounds or their dimeric bisoxazo derivates. Different types of novel xanthates containing a vinyl ether moiety, S-benzyl O-2-(vinyloxy)ethyl carbonodithioate (Xanthate 1) and S-1-(ethoxycarbonyl)ethyl O-2-(vinyloxy)ethyl carbonodithioate (Xanthate 2) were synthesized. In particular, the Xanthate 2 enabled the design of polyvinyl alc. (PVA) stereoblock copolymer via the combination of living cationic vinyl polymerization and RAFT/MADIX polymerization For cationic polymerization of iso-Bu vinyl ether (IBVE) and tert-Bu vinyl ether (TBVE), the polymerizations were conducted under Xanthate 1-HCl adduct/SnCl4 and Xanthate 1 or 2-CF3COOH adduct/EtAlCl2 initiating system in the presence of Et acetate. Both systems proceeded in living polymerization fashion because the calculated Mn of both poly(IBVE) and poly(TBVE) matches with the Mn polymerized assuming that one polymer chain is formed per one mol. of the Xanthate 1 or 2. The resulting poly(TBVE) had a high number average α-end functionality as determined by MALDI-TOF-MS spectrometry. Xanthate 2 is more efficient for the following RAFT/MADIX polymerization of vinyl acetate (VAc). The RAFT/MADIX polymerization of vinyl acetate (VAc) using azobis(isobutyronitrile) (AIBN) at 60 °C was conducted using either poly(IBVE) or poly(TBVE) macro-CTA. The poly(TBVE) macro-CTAs synthesized from the Xanthate 2 were able to polymerize VAc smoothly via RAFT/MADIX polymerization, to prepare well-defined diblock copolymer, poly(TBVE)-b-poly(VAc). The resulting block copolymer was then hydrolyzed using KOH in methanol and followed by acid hydrolysis using HBr gas bubbling. The resulting polymer is and inherently stereoblock-like copolymer, isotactic rich PVA-b-atactic PVA (iPVA-b-aPVA). From the DSC measurement, the iPVA-b-aPVA has one glass transition at 69.5 °C and two m.ps. according to iPVA and aPVA at 237.9 and 198.1 °C, resp. Thus, it can be suggested that the obtained PVA has two different geometries by the combination of living cationic polymerization and RAFT/MADIX polymerization

In the present study, sex pheromones from virgin females were collected by the filter paper adsorption method and the glass-wall adsorption method to investigate the pheromones. The efficiency of collection by the filter paper method was �0-times greater than that by the glass wall method. However, there was no difference in the ratio of pheromonal components between 2 collection methods. The amount of pheromones and their related compounds released from STT was �-times greater than that from SFT, but almost the same quantity of alcs. were discharged by each species.
