Brucellosis is one of the most widespread bacterial zoonoses worldwide. Here, our aim was to identify the effector mechanisms controlling the early stages of intranasal infection with Brucella in C57BL/6 mice. During the first 48 h of infection, alveolar macrophages (AMs) are the main cells infected in the lungs. Using RNA sequencing, we identified the aconitate decarboxylase 1 gene (Acod1; also known as Immune responsive gene 1), as one of the genes most upregulated in murine AMs in response to B. melitensis infection at 24 h post-infection. Upregulation of Acod1 was confirmed by RT-qPCR in lungs infected with B. melitensis and B. abortus. We observed that Acod1-/- C57BL/6 mice display a higher bacterial load in their lungs than wild-type (wt) mice following B. melitensis or B. abortus infection, demonstrating that Acod1 participates in the control of pulmonary Brucella infection. The ACOD1 enzyme is mostly produced in mitochondria of macrophages, and converts cis-aconitate, a metabolite in the Krebs cycle, into itaconate. Di-Me itaconate (DMI), a chem.-modified membrane permeable form of itaconate, has a dose-dependent inhibitory effect on Brucella growth in vitro. Interestingly, structural anal. suggests the binding of itaconate into the binding site of B. abortus isocitrate lyase. DMI does not inhibit multiplication of the isocitrate lyase deletion mutant ΔaceA B. abortus in vitro. Finally, we observed that, unlike the wt strain, the ΔaceA B. abortus strain multiplies similarly in wt and Acod1-/- C57BL/6 mice. These data suggest that bacterial isocitrate lyase might be a target of itaconate in AMs.

The reactions occur in parallel, under the same reaction conditions in anisole, a solvent with an impressive sustainability rank comparable to those of ethanol and water. The products contain formyl and acetoxy moieties, both established olfactory groups in flavor and fragrance industry, and present potential as new fragrance components with less allergenic properties. To the best of our knowledge, this is the first time that a one-pot process involving hydroformylation combined with further functionalization in a remote site is described.

Beauty salons are considered occupational environments where the staff personnel and clients are exposed to high levels of airborne pollutants. In this study, air quality monitoring was carried out in Leon, Spain. Temperature, relative humidity, CO2, CO and particulate matter were continuously monitored inside and outside areas of the salon. PM10 was simultaneously collected onto filters. Volatile organic compounds and carbonyls were sampled inside the salon. Indoor PM10 concentrations exceeded the recommended 24-h guideline of 50 μg m-3. A detailed PM10 chem. characterization included organic and elemental carbon, trace elements, water-soluble ions and organic speciation. Indoor vs outdoor ratios of PM10, as well as the majority of PM10-bound organic compounds, were >1. During work periods, organic carbon accounted for 29.5 ± 1.8 and 16.2 ± 4.5%weight of PM10 indoors and outdoors, resp. More than 200 individual organic compounds were detected in the PM10, including aliphatic alcs., fatty acids, phthalates, glycerol derivatives, fatty acid alkyl esters, phenolic compounds, alkanes and polycyclic aromatic hydrocarbons, among others. Many of them are part of formulations widely used in hair cosmetics, personal care and cleaning products. Inadequate ventilation, working activities and the use of specific products can greatly contribute to high indoor levels of organic air pollutants. Formaldehyde revealed a cancer risk of 4.6 x 10-6, higher than the guideline level, suggesting a “possible risk” for workers. The total excess lifetime cancer risk from exposure to multiple compounds was 9.3 x 10-6, which is lower than the acceptable risk, but not negligible.

The in vivo hepatoprotective potential of methanolic extract of Ceasalpinia bonduc (CBLM) has been explored against carbon tetrachloride (CCl4) induced acute liver injury in rats. Treatment of plant extract on CCl4 intoxicated liver significantly reduced the hepatoxicity, along with serum enzymes GPT and GOT. To explore the chem. constituents from CBLM extract, it was fractionated into non-polar to moderately polar fractions (CBLM-H, CBLM-HEt, CBLM-Et, CBLM-EtM, CBLM-M) and subjected to GC/GC-MS anal. Altogether twenty seven (~71%) phytochems. were identified from different fractions by using Electronic Mass Spectral Library GC-MS (NIST 20). Out of which twenty one are first time reported from Ceasalpinia bonduc, fourteen from genus Caesalpinia and ten from family Fabaceae. The identified phytochems. 2-ethyl-2-hydroxy-1,3-dimethylcyclopentanecarboxylic acid, Et ester (21) and 1,3,5-triazine-2,4-diamine,6-hydroxy-N,N-dicyclohexyl (23) are first time identified as plant metabolites. To explore the antimicrobial potential four strains of Gram-pos. and eight strains of Gram-neg. bacteria were used along with pure cultures of five saprophytic fungus (molds) and two strains of yeast were utilized. CBLM-H and CBLMHEt were exhibited praiseworthy antimicrobial potential. CBLM-H showed complete growth inhibition of P. mirabilis and V. cholerae at the concentration of 0.1μg/mL while CBLM-HEt at 0.05μg/mL halted the growth of S. aureus.

A novel phosphorus, nitrogen and sulfur containing monomer, di-Et (acryloyloxy) ethylthiophosphoramidate was synthesized in a two step reaction. The monomer was polymerized and grafted onto cotton fabric by the living radical polymerization method using benzyl N,N-di-Et thiocarbamate as iniferter and sur-iniferter resp. The monomer, its precursor, iniferter and the polymer were characterized by spectroscopic techniques. Flame retardant property of the modified cotton fabric was studied by the Shirley Manual Flammability Tester and it was found that the incorporation of phosphorus and nitrogen together exerts a synergistic effect in imparting flame retarding property to the fabric.
