This research focused on the composition for the essential oils, which was obtained by solvent-free microwave extraction (SFME) from the aerial parts of Scorzonera calyculata, and hydrodistd. oils from the aerial parts and roots of Centaurea irritans, from Astraceae family, were investigated by gas chromatog. (GC) and GC/mass spectrometry (MS). In addition, the biol. activities of the methanolic extracts from the aerial parts of S. calyculata and C. irritans were determined Total phenolic content was determined by the Folin-Ciocalteu procedure. Antibacterial activity of the methanolic extracts was carried out by min. inhibitory concentration (MIC) and min. bactericidal concentration (MBC). Cytotoxicity of the methanolic extract of S. calyculata against human lung cancer cells (A549) and the methanolic extract of C. irritans against breast lung cancer cells (MCF-7) were measured using 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) method. The obtained results of GC/MS technique showed that the SFME oil of S. calyculata, was rich in regard to nonterpenoid and sesquiterpene components. Both oils from the aerial parts and roots of C. irritans were rich in regard to oxygenated monoterpenes. The S. calyculata and C. irritans extracts showed moderate antioxidant activities with an inhibitory concentration (IC50) value of 1.48 and 1.99 mg/mL, using 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay and 73.51 and 44.48μmol Fe (II)/g dry mass using ferric-reducing antioxidant power (FRAP) assay, resp. The extracts showed high toxicity against gram-pos. bacteria and the IC50 value of extracts cytotoxicity was found to be 9.8 and 10.3 mg/mL, resp. It appeared that the investigated samples could be as a promising drug for pharmaceutical industry.

Me palmitate predominates in P. salicifolia oil representing 51.15% whereas thymol (35.76%) is the major constituent in P. sewerzowii essential oil. GC-MS analyses showed that P. bucharica and P. sewerzowii are more closely related comparable to P. salicifolia. The antimicrobial activity of the essential oils was assessed against different microorganisms using agar-disk diffusion and broth microdilution assay. Among the three tested species, the essential oil of P. salicifolia showed the highest antibacterial activity.

The risk of fomite-mediated transmission in the clinic is substantially increasing amid the recent COVID-19 pandemic as personal protective equipment (PPE) of hospital workers is easily contaminated by direct contact with infected patients. In this context, it is crucial to devise a means to reduce such transmission. Herein, we report an antimicrobial, antiviral, and antibiofouling trifunctional polymer that can be easily coated onto the surface of medical protective clothing to effectively prevent pathogen contamination on the PPE. The coating layer is formed on the surfaces of PPE by simple spray-coating of an aqueous solution of the trifunctional polymer, poly(DMA-PEGMA-QA). To establish an optimal ratio of antifouling and antimicrobial functional groups, we performed antifouling, antibacterial, and antiviral tests using four different ratios of the polymers. Antifouling and bactericidal results were assessed using Staphylococcus aureus, a typical pathogenic bacterium that induces an upper respiratory infection. Regardless of the molar ratio, polymer-coated PPE surfaces showed considerable antiadhesion (�5-75%) and antibacterial (�5-87%) efficacies soon after in contact with pathogens and maintained their capability for at least 24 h, which is sufficient for disposable PPEs. Further antiviral tests using coronaviruses showed favorable results with PPE coated at two specific ratios (3.5:6:0.5 and 3.5:5.5:1) of poly(DMA-PEGMA-QA). Moreover, biocompatibility assessments using the two most effective polymer ratios showed no recognizable local or systemic inflammatory responses in mice, suggesting the potential of this polymer for immediate use in the field.

Abstract: The novel human coronavirus (SARS-CoV-2), the causative agent of COVID-19, has quickly become a threat to the public health and economy worldwide. Despite the severity of some cases, there are no current pathogen-specific antivirals available to treat the disease. Therefore, many studies have focused on the evaluation of the anti-SARS-CoV-2 activity of clin. available drugs. Here, we conducted a systematic review to describe the drug repositioning strategy against SARS-CoV-2 and to discuss the clin. impact of this approach in the current pandemic context. The systematic review was performed on March 23, 2020, using PubMed/MEDLINE, Scopus, Cochrane Library, and Biblioteca Virtual de Saud́e (BVS). The data were summarized in tables and critically analyzed. After the database search, 12 relevant studies were identified as eligible for the review. Among the drugs reported in these studies, 57 showed some evidence of antiviral activity. Antivirals, especially antiretrovirals, are the main class of therapeutic agents evaluated against COVID-19. Moreover, studies have reported the anti-SARS-CoV-2 activity of antitumor (16%; 9/57), antimalarial (7%, 4/57), and antibacterial (5%; 3/57) agents. Addnl., seven pharmacol. agents (chloroquine, tetrandrine, umifenovir (arbidol), carrimycin, damageprevir, lopinavir/ritonavir) are in phase IV of clin. trials. Due to the evidence of the anti-SARS-CoV-2 activity of various clin. available agents, drug repositioning stands out as a promising strategy for a short-term response in the fight against the novel coronavirus.

Essential oils (EOs) extracted from botanical resources have greatly been appraised as antimicrobials against a broad spectrum of microorganisms. A panoply of exptl. studies have pointed out the immense potency of EOs as natural antimicrobial agents at relatively low doses; largely attributed to the synergistic interactions of various bioactive components. In the present study, 10 EOs prepared from two endemic Pittosporum senacia Putterl. subsp. Senacia and Syzygium coriaceum J. Bosser & J. Gueho and seven exotic Cinnamomum camphora (L.) Nees & Eberm, Citrus aurantium L., Curcuma longa L, Morinda citrifolia L., Petroselinum crispum (Mill.) Fuss, Plectranthus amboinicus (Lour.) Sprengel and Syzygium samarangense (Blume) Merr. & L. Perry aromatic medicinal plants from Mauritius, were screened for their growth inhibitory activities against eight bacteria (ATCC strains and clin. isolates) using broth microdilution techniques. The EOs were found to possess varying degree of antibacterial potency. The most active EOs were found to have min. inhibitory concentration (MIC) of 0.25-4 mg/mL and min. bactericidal concentration (MBC) of 0.25-16 mg/mL. In particular, at its MIC values, P. amboinicus EO showed bactericidal effects against four strains. However, all tested bacteria were insensitive to P. senecia and C. aurantium fruit peel EOs. Bacillus spizizenii was found to be the most susceptible strain to the active EOs. Addnl., P. amboinicus and the two Syzygium spp. EOs showed antibiotic potentiating activities. Mostly synergistic and partial synergistic actions of the EOs in combination with the conventional antibiotics (streptomycin, chloramphenicol and ciprofloxacin) at a ratio of 1:1 were obtained against the tested bacteria. Thus, results from this study highlighted the antibacterial potential and the efficacy of the most active EOs as antibiotic potentiating agents that could potentially be used in combinational therapies along with conventional antibiotics for a synergistic approach in the treatment/management of clin. relevant bacterial infections.

The aim of this study is to estimate the nutritive values, metabolites of Hass and Reed Avocado cultivars and evaluate their antimicrobial and anticancer activities. Hass was rich in water soluble vitamins, iron and calcium while, Reed contains more fat soluble vitamins. Benzaldehyde and Bu phenol derivatives were the major volatile components identified by solid phase extraction in Hass and Reed, resp. Naringenin and rutin were the major compounds identified in Hass and Reed by HPLC anal. resp. Hass showed a promising antimicrobial activity, especially, against Methicillin-Resistant S. aureus (MIC = 7.81 μg/mL). Two targets sites were selected to investigate the mechanism of action of Hass, Staphylococcal Enterotoxin-like X and Serine/threonine kinases Proteins (STK). Mol. docking demonstrated high binding affinity of naringenin towards Enterotoxin-like X. However, high levels of rutin in Reed might account for its cytotoxic activity against colorectal adenocarcinoma. Avocado extracts may be used for developing potential antibiotics.

The Drosophila melanogaster enzymes juvenile hormone esterase (DmJHE) and its duplicate, DmJHEdup, present ideal examples for studying the structural changes involved in the neofunctionalization of enzyme duplicates. DmJHE is a hormone esterase with precise regulation and highly specific activity for its substrate, juvenile hormone. DmJHEdup is an odorant degrading esterase (ODE) responsible for processing various kairomones in antennae. Our phylogenetic anal. shows that the JHE lineage predates the hemi/holometabolan split and that several duplications of JHEs have been templates for the evolution of secreted β-esterases such as ODEs through the course of insect evolution. Our biochem. comparisons further show that DmJHE has sufficient substrate promiscuity and activity against odorant esters for a duplicate to evolve a general ODE function against a range of mid-long chain food esters, as is shown in DmJHEdup. This substrate range complements that of the only other general ODE known in this species, Esterase 6. Homol. models of DmJHE and DmJHEdup enabled comparisons between each enzyme and the known structures of a lepidopteran JHE and Esterase 6. Both JHEs showed very similar active sites despite low sequence identity (30%). Both ODEs differed drastically from the JHEs and each other, explaining their complementary substrate ranges. A small number of amino acid changes are identified that may have been involved in the early stages of the neofunctionalization of DmJHEdup. Our results provide key insights into the process of neofunctionalization and the structural changes that can be involved.
