Neuroprotection Effect of Astragaloside IV from 2-DG-Induced Endoplasmic Reticulum Stress

Objective. Astragaloside IV shows neuroprotective activity, but its mechanism remains unclear. To investigate whether astragaloside IV protects from endoplasmic reticulum stress (ERS), we focus on the regulation of glycogen synthase kinase-3 beta (GSK-3 beta) and mitochondrial permeability transition pore (mPTP) by astragaloside IV in neuronal cell PC12. Methods and Results. PC12 cells treated with different concentrations of ERS inductor 2-deoxyglucose (2-DG) (25-500 mu M) showed a significant increase of glucose-regulated protein 78 (GRP 78) and GRP 94 expressions and a decrease of tetramethylrhodamine ethyl ester (TMRE) fluorescence intensity and mitochondrial membrane potential ( increment psi m), with the peak effect seen at 50 mu M, indicating that 2-DG induces ERS and the mPTP opening. Similarly, 50 mu M of astragaloside IV increased the GSK-3 beta phosphorylation at Ser9 most significantly. Next, we examined the neuroprotection of astragaloside IV by dividing the PC12 cells into control group, 2-DG treatment group, astragaloside IV plus 2-DG treatment group, and astragaloside IV only group. PC12 cells treated with 50 mu M 2-DG for different time courses (0-36 hr) showed a significant increase of Cleaved-Caspase-3 with the peak at 6 hr. 2-DG significantly induced cell apoptosis and increased the green fluorescence intensity of Annexin V-FITC, and these effects were reversed by astragaloside IV. Such a result indicates that astragaloside IV protected neural cell survival from ERS. 2-DG treatment significantly increased the expressions of inositol-requiring ER-to-nucleus signal kinase 1 (IRE1), phosphor-protein kinase R-like ER kinase (p-PERK), but not affect the transcription factor 6 (ATF6) expression. 2-DG treatment significantly decreased the phosphorylation of GSK-3 beta and significantly reduced the TMRE fluorescence intensity and increment psi m, following mPTP open. Astragaloside IV significantly inhibited the above effects caused by 2-DG, except the upregulation of ATF6 protein. Taken together, astragaloside IV significantly inhibited the ERS caused by 2-DG. Conclusion. Our data suggested that astragaloside IV protects PC12 cells from ERS by inactivation of GSK-3 beta and preventing the mPTP opening. The GRP 78, GRP 94, IRE1, and PERK signaling pathways but not ATF6 are responsible for GSK-3 beta inactivation and neuroprotection by astragaloside IV.

Study on Hydrolysis of Magnesium Hydride by Interface Control

Magnesium hydride (MgH2) is one of the competitive hydrogen storage materials on account of abundant reserves and high hydrogen content. The hydrolysis of MgH2 is an ideal and controllable chemical hydrogen generation process. However, the hydrolyzed product of MgH2 is a passivation layer on the surface of the magnesium hydride, which will make the reaction continuity worse and reduce the rate of hydrogen release. In this work, hydrogen generation is controllably achieved by regulating the change of the surface tension value in the hydrolysis, a variety of surfactants were systematically investigated for the effect of the hydrolysis of MgH2 In the meantime, the passivation layer of MgH2 was observed by scanning electron microscope (SEM), and the surface tension value of the solution with different surfactants were monitored, investing the mechanism of hydrolysis adding different surfactants. Results show that different surfactants have different effects on hydrogen generation. The hydrogen generation capacity from high to low is as follows: tetrapropylammonium bromide (TPABr), sodium dodecyl benzene sulfonate (SDBS), Ecosol 507, octadecyl trimethyl ammonium chloride (OTAC), sodium alcohol ether sulfate (AES), and fatty methyl ester sulfonate (FMES-70). When the ratio of MgH2 to TPABr was 5 : 1, the hydrogen generation was increased by 52% and 28.3%, respectively, at the time of 100 s and 300 s. When hydrolysis time exceeds 80 s, the hydrogen generation with AES and FMES-70 began to decrease; it was reduced by more than 20% at the time of 300 s. SEM reveals that surfactants can affect the crystalline arrangement of Mg(OH)(2) and make the passivation layer three-dimensionally layered providing channels for H2O molecules to react with MgH2.

Derivatives of nitrogen mustard anticancer agents with improved cytotoxicity

In previous studies, we demonstrated that esters of bendamustine containing a basic moiety are far more cytotoxic anticancer agents than their parent compound and that the substitution of the labile ester moiety by a branched ester or an amide markedly increases stability in the blood plasma. In the current study, we showed that this substitution was bioisosteric. Aiming at increased cytotoxicity, we introduced the same modification to related nitrogen mustards: 6-isobendamustine, chlorambucil, and melphalan. The synthesis was accomplished using the coupling reagents N,N ‘-dicyclohexylcarbodiimide or 2-(1H-benzotriazole-1-yl)-1,1,3,3-tetramethylaminium tetrafluoroborate. Cytotoxicity against a panel of diverse cancer cells (carcinoma, sarcoma, and malignant melanoma) was assessed in a kinetic chemosensitivity assay. The target compounds showed cytotoxic or cytocidal effects at concentrations above 1 mu M: a striking enhancement over bendamustine and 6-isobendamustine, both ineffective against the selected cancer cells at concentrations up to 50 mu M, and a considerable improvement over chlorambucil, showing some potency only against the sarcoma cells. Cross-sectional analysis of plasma and CSF metabolomic markers in Huntington’s disease for participants of varying functional disability: a pilot study

Huntington’s Disease (HD) is a progressive, fatal neurodegenerative condition. While generally considered for its devastating neurological phenotype, disturbances in other organ systems and metabolic pathways outside the brain have attracted attention for possible relevance to HD pathology, potential as therapeutic targets, or use as biomarkers of progression. In addition, it is not established how metabolic changes in the HD brain correlate to progression across the full spectrum of early to late-stage disease. In this pilot study, we sought to explore the metabolic profile across manifest HD from early to advanced clinical staging through metabolomic analysis by mass spectrometry in plasma and cerebrospinal fluid (CSF). With disease progression, we observed nominally significant increases in plasma arginine, citrulline, and glycine, with decreases in total and d-serine, cholesterol esters, diacylglycerides, triacylglycerides, phosphatidylcholines, phosphatidylethanolamines, and sphingomyelins. In CSF, worsening disease was associated with nominally significant increases in NAD(+), arginine, saturated long chain free fatty acids, diacylglycerides, triacylglycerides, and sphingomyelins. Notably, diacylglycerides and triacylglyceride species associated with clinical progression were different between plasma and CSF, suggesting different metabolic preferences for these compartments. Increasing NAD(+) levels strongly correlating with disease progression was an unexpected finding. Our data suggest that defects in the urea cycle, glycine, and serine metabolism may be underrecognized in the progression HD pathology, and merit further study for possible therapeutic relevance.

Infectious Effect of Organic Small Molecules on Photo-oxidative Aging of Polypropylene

Aging of a polymer is generally affected by adjacent polymers, known as infectious behavior of aging. Organic small molecules generated during aging are considered to be infection agents. However, the way they influence aging process and the difference among various infection agents are still unknown. In this paper, the influence of 18 representative organic small molecules on photo-oxidative aging of polypropylene (PP) was studied. The small molecules included acids, esters, aldehydes, ketones and alcohols, corresponding to typical photo-oxidative degradation products of polymers. When they were placed near PP during aging, the carbonyl index (CI) of PP was higher compared to the case when PP was aged individually. It was demonstrated that photo-oxidative aging of PP was accelerated by all the small molecules. The acceleration effects of acids, aldehydes and ketones were outstanding. Especially for propanoic acid, isobutyric acid, methacrylic acid, formaldehyde, acetaldehyde and butanone, CI was increased by more than 700% when PP was aged with them. Further, infection mechanism of acetone and acetic acid was investigated , since they had been identified as typical infection agents. CI and concentrations of gaseous degradation products of PP with and without acetone or acetic acid were measured with aging time. When PP was aged with acetone, CI was increased from as early as 24 h at an extremely high rate. With acetic acid, CI was increased after an obvious induction period and the increasing rate was lower than that with acetone. Similar phenomenon was observed in gaseous degradation products. The results showed that acetone acted as an initiator, photolyzed to generate CH3 center dot that could initiate the oxidation of PP; while acetic acid acted as a catalyzer, facilitating the decomposition of hydroperoxides and the accumulation of oxidation products.

Comparison of five membrane filters to collect bioaerosols for airborne microbiome analysis

Aims Recovering DNA of airborne micro-organisms (AM) from air is a challenging task. We compared five membrane filters for bioaerosol sampling-mixed cellulose ester (MCE), polyethersulfone (PES), polyamide (PA), polytetrafluorethylene (PTFE) and polyvinylidene fluoride (PVDF)-based on their bacterial, fungal and eukaryotic DNA recoveries. Methods and results Bacterial, fungal and eukaryotic populations were quantified using quantitative PCR. With a bacterial consortium, PTFE exhibited the best recovery efficiency (113%), followed by PA (92%), PES (86%), MCE (48%) and PVDF (1%). When filters were compared with air, PA was used as a control to normalize results from the others. The bacterial, fungal and eukaryotic DNA recovery ratios were markedly greater in PES (9 center dot 3, 11 center dot 5 and 10 center dot 3 respectively) than in the remaining. Eukaryotic MiSeq sequencing revealed that PES recovered a more diverse and considerably richer assemblage (richness ratios, 4 center dot 97 vs <= 1 center dot 16 for PES vs the others). Rank abundance distribution analysis showed that distribution tails were longer (>4 times) in PES, but these did not differ between the remaining and PA. Community comparison showed that PES exhibited a lower variation across trials than the PA, while the remaining did not. Conclusions PES filter markedly outperformed the other filters in quantitative and qualitative recovery of AM. Significance and Impact of the Study Our findings demonstrated the importance of filter selection for sampling AM.

Nimodipine attenuates dibutyl phthalate-induced learning and memory impairment in kun ming mice: An in vivo study based on bioinformatics analysis

Dibutyl phthalate (DBP), a typical representative of phthalate esters (PAEs), is used as a plasticizer in various industrial applications and has been reported to be responsible for neurobehavioral changes. The results of an in vivo study confirmed that DBP affects the levels of Ca2+-related proteins, up-regulates phosphorylated -ERK1/2 expression and results in hippocampal neuronal damage and apoptosis, whereas Nim as a Ca2+ antagonist, has a certain neuroprotective role to avoid these adverse effects. Our data suggest that Nim could be used to attenuate the learning and memory impairment in DBP-exposed mice, to down-regulate intracellular Ca2+ levels, subordinate the ERK1/2 pathway and attenuate apoptosis in hippocampal tissue.

Maternal Allocation of Lipids to Eggs by Southern Flounder after Diet Changes

Maturation and spawning diets for flatfish broodstock, such as Southern Flounder Paralichthys lethostigma, have received little research attention, despite their critical role in the reproductive performance. During the spawning season, which is when Southern Flounder broodstock produce multiple egg batches, they rely on their diet to supply nutrients to the eggs. Lipids are an important class of nutrients for offspring development. The objective of this brief communication is to describe the changes in egg lipid classes during the spawning season following a diet change. Female broodstock were fed three diets after the first spawn (week 0)-a control diet (equal weights of Spanish Sardines Sardinella aurita and brown shrimp Farfantepenaeus aztecus), a shrimp-only diet, and a sardine-only diet (n = 3, 3, and 4, respectively)-for a period of 6 weeks and the lipid composition of the eggs was measured every 2 weeks. The change to a diet of sardines only or shrimp only altered the concentrations of egg lipid classes, indicating that egg composition is affected by maternal dietary intake over a period of 2 weeks or less. Females that were fed the sardine-only diet produced eggs with higher levels of ketones and acetone-mobile polar lipids and lower levels of hydrocarbons than did females that were fed shrimp, exhibiting changes that were consistent with the differences between the diets. The shift to the sardine-only diet had little effect on total lipids, triacylglycerols, free fatty acids, and sterols in the eggs. The change to the shrimp-only diet resulted in subtle but consistent decreases in all of the lipids, except wax/steryl esters at week 2, followed by gradual increases. This suggests that the shrimp-only diet provided insufficient nutrition and that the females mobilized stored lipids to compensate. These preliminary results revealed interesting patterns that, with further research, could lead to improved diets and feeding protocols for the production of Southern Flounder.
