Synthesis, Biological Evaluation and In silico Studies of Compounds Based on Tryptophan-Naproxen-Triazole Hybrids

A hybrid of three different frameworks e. g. tryptophan, naproxen and triazole has been explored for the identification of potential antibacterial / anticancer agents. A library of new compounds, designed based on this hybrid framework was synthesized via a multi-step sequence using the Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC) as the key reaction step. Thus the terminal alkyne obtained via the reaction of tryptophan ester with naproxen followed by N-propargylation of the indole ring was coupled with a range of organic azides to give the desired products (through the formation of triazole ring) in good to acceptable yields. Ultrasound-Assisted Method for the Synthesis of Tertiary Fatty Aliphatic Esters with Potential Antimicrobial Activity

This study aims to demonstrate a simple and rapid green method for the ultrasound-assisted synthesis of t-aliphatic esters by reacting t-butanol with saturated (capric lauric, palmitic) and unsaturated (oleic and undecylenic) acids. The spectral characterization of esters as performed by IR and NMR spectroscopies. The impact of ultrasonic irradiation on the reaction in comparison with the conventional method was analyzed based on the reaction time and the yield of the resulting esters. After spectroscopic studies. the synthesis of the corresponding esters after the ultrasound -assisted irradiation was proven. The yields of the t-butyl fatty acid esters increased between 2 and 10% in comparison with the conventional method for their synthesis. It was also established that the application of ultrasonic irradiation intensifies the esterification process significantly compared to the conventional method. The advantages of the ultrasound-assisted esterification include shortening the reaction time: the process is carried out at room temperature instead of at the conventional reaction temperature 67-70 degrees C. How olive washing and storage affect fruit ethanol and virgin olive oil ethanol, ethyl esters and composition

BACKGROUND Olives are stored for a short time after harvesting pending processing in the oil mills. Furthermore, olives are often washed prior to fruit storage. In this work we study how washing and storage affect fruit ethanol content and the effect on virgin olive oil ethanol content and quality. RESULTS Olive storage produced an increase in the fruit ethanol content, achieving values six times higher when storage was in silos. Washing the olives resulted in an increase in fruit ethanol content, although when washed olives were processed immediately no difference was found. The increase in fruit ethanol content during storage was reflected in higher oil ethanol concentration. Similarly, olive washing resulted in oils with higher ethanol concentration. Industrial conditions gave more important increases in oil ethanol content than that from olives processed by hand. For quality parameters all the olive oils were classified as ‘extra virgin’. In general, oils showed a slight decrease in some sensory attributes. At industrial scale after 24 h storage oils were classified as ‘virgin’ because sensory defects were found. CONCLUSION Olive storage should be avoided or reduced to less than 12 h; if possible, olives should not be washed before storage since this practice favors losses in sensory characteristics and the synthesis of ethanol, a precursor of ethyl esters. Extreme Low-Temperature Combustion Chemistry: Ozone -Initiated Oxidation of Methyl Hexanoate

The accelerating chemical effect of ozone addition on the oxidation chemistry of methyl hexanoate [CH3(CH2)(4)C(=O)OCH3] was investigated over a temperature range from 460 to 940 K. Using an externally heated jet-stirred reactor at p = 700 Torr (residence time tau = 1.3 s, stoichiometry phi = 0.5, 80% argon dilution), we explored the relevant chemical pathways by employing molecular-beam mass spectrometry with electron and single-photon ionization to trace the temperature dependencies of key intermediates, including many hydroperoxides. In the absence of ozone, reactivity is observed in the so-called low-temperature chemistry (LTC) regime between 550 and 700 K, which is governed by hydroperoxides formed from sequential O-2 addition and isomerization reactions. At temperatures above 700 K, we observed the negative temperature coefficient (NTC) regime, in which the reactivity decreases with increasing temperatures, until near 800 K, where the reactivity increases again. Upon addition of ozone (1000 ppm), the overall reactivity of the system is dramatically changed due to the time scale of ozone decomposition in comparison to fuel oxidation time scales of the mixtures at different temperatures. While the LTC regime seems to be only slightly affected by the addition of ozone with respect to the identity and quantity of the observed intermediates, we observed an increased reactivity in the intermediate NTC temperature range. Furthermore, we observed experimental evidence for an additional oxidation regime in the range near 500 K, herein referred to as the extreme low-temperature chemistry (ELTC) regime. Experimental evidence and theoretical rate constant calculations indicate that this ELTC regime is likely to be initiated by H abstraction from methyl hexanoate via O atoms, which originate from thermal O-3 decomposition. The theoretical calculations show that the rate constants for methyl ester initiation via abstraction by O atoms increase dramatically with the size of the methyl ester, suggesting that ELTC is likely not important for the smaller methyl esters. Experimental evidence is provided indicating that, similar to the LTC regime, the chemistry in the ELTC regime is dominated by hydroperoxide chemistry. However, mass spectra recorded at various reactor temperatures and at different photon energies provide experimental evidence of some differences in chemical species between the ELTC and the LTC temperature ranges.

The Protective Effect of Aspirin Eugenol Ester on Paraquat-Induced Acute Liver Injury Rats

Aspirin eugenol ester (AEE) possesses anti-inflammatory and anti-oxidative effects. The study was conducted to evaluate the protective effect of AEE on paraquat-induced acute liver injury (ALI) in rats. AEE was against ALI by decreasing alanine transaminase and aspartate transaminase levels in blood, increasing superoxide dismutase, catalase, and glutathione peroxidase levels, and decreasing malondialdehyde levels in blood and liver. The yield of 10-hydroxy-12-octadecenoic acid (HOD) achieved 85% under optimum hydration conditions. At the same time, the thermal rearrangement reaction of 10-HOD was also preliminary investigated in this research. Value-added chemicals of alpha-octene and 10-oxo-decanoic acid (ODA) were obtained through the thermal cleavage. From this process, a high efficiency carbon production pathway with great potential for producing high value-added chemicals from unsaturated fatty acid was presented.

Quality Markers for Astragali Radix and Its Products Based on Process Analysis

Due to the complex nature of traditional medicines, quality control methods need to cover two aspects: compliance of raw materials with quality standards and process control. Astragali radix (AR), the roots of Astragalus mongholicus Bunge, was selected in this study as an example of a widely used traditional medicine in various formulations. Astragaloside IV (AG IV) and calycosin 7-O-beta-D-glucoside (CG) are used as the markers for the quality control of AR and its products in the Chinese Pharmacopoeia. However, in the raw materials, malic acid esters of the CG and acetate esters of the astragaloside are easily decomposed into CG and AG IV during storage and processing of AR to make extracts for various preparations. The thermal stability of the isoflavonoids and astragalosides in decoction was studied. The level of CG and astragalosides (AG I/AG II/AG IV) was strongly affected by prolonged heat during processing, while calycosin was stable in the conditions. Also the major astragalosides in AR could fully converted into AG IV which eventually reaches a stable level under certain conditions. With calycosin and AG IV as marker components, practical, reproducible, and precise methods were established and applied to the quality analysis of AR from its raw materials to its intermediates and products. This study demonstrates that a full chemical profiles analysis of the whole manufacturing process (from raw materials-intermediates/extracts-final product) is important to identify quality markers (Q-markers) and even to establish proper analysis methods for traditional Chinese medicine products.

Evaluation of the best pH-sensitive linker using norbornene-derived polymers

pH-responsive drug delivery systems that are bearing with acid-sensitive linkages between therapeutic molecules and polymers have been extensively explored in cancer treatment. But there is no systematic study to choose the best acid-sensitive linker in literature. In this report, three polymer-drug conjugates were synthesized with different pH-sensitive linkers such as ester, amide, and hydrazone with the anticancer drug doxorubicin with the same polymer backbone and the same molecular weights. We believe that this study will provide useful decision-making information to choose the best linker for the stimuli-responsive delivery system. First, we synthesized three different norbornene monomers with ester linker (mono 1), amide linker (mono 2), and hydrazone linker (mono 3). Next, the homopolymerization of mono 1 (HP-DOXE), mono 2 (HP-DOXA), and mono 3 (HP-DOXH) were carried out to obtain same molecular weight of homopolymers by using second generation Grubbs’ catalyst. In vitro drug release profile indicated the importance of having the hydrazone linker that helps the drug at the mildly acidic conditions resembling the pH of the cancerous cells, compared to amide and ester linkers with the same polymer backbone and the same molecular weights of polymers. The cell viability experiments in cancerous cell lines demonstrate that the hydrazone linker kills more cells compared to ester and amide linkers up to 500 mu g/mL concentration.

In recent years, polysorbate degradation in biopharmaceutical formulations has been observed. Especially hydrolysis, which can be induced chemically as well as enzymatically, leads to the release of FAs from PS. These FAs are poorly soluble in aqueous buffer systems due to their hydrophobic nature and therefore prone to precipitation and particle formation. Since the emergence of particles in liquid formulations has to be avoided, it is important to prevent their formation. This study evaluates the solubility limits of selected FAs, which are likely to be released during the degradation of PS20 and PS80 in the presence of defined PS concentrations. Our results show that the solubility is highly dependent on the pH, the temperature, the used PS concentration and the aliphatic chain of respective FAs. Solubility of FAs, such as palmitic and oleic acid under the conditions determined in this study, are in the range of 3-130 mu g.ml(-1) (12-460 mu M). Furthermore, the results allow making an estimation to which extent PS may degrade before particle formation in the drug product may be expected.
