Synthesis and biological activities of simplified aplysiatoxin analogs focused on the CH/pi interaction

Debromoaplysiatoxin (DAT) is a potent protein kinase C (PKC) activator with tumor-promoting and proinflammatory activities. Irie and colleagues have found that 10-methyl-aplog-1 (1), a simplified analog of DAT, has strong anti-proliferative activity against several cancer cell lines with few adverse effects. Based on the synthetic route of 1, 2 was convergently synthesized in 26 linear steps from 6-hydroxy-1-naphthoic acid with an overall yield of 0.18%. Although the anti-proliferative activity of 2 was more potent than that of 1, the binding potency of 2 to the PKC delta-C1B domain did not exceed that of 1. Molecular dynamics simulation indicated the capability of 2 to simultaneously form hydrogen bonds and CH/pi interactions with the PKC delta-C1B domain. Focusing on the hydrogen bonds, their geometry in the binding modes involving the CH/pi interactions seemed to be sub-optimal, which may explain the slightly lower affinity of 2 compared to 1. This study could be of help in optimizing such interactions and synthesizing a promising lead cancer compound.

Molecular docking and nonbonding interactions have been performed against lanosterol 14 alpha-demethylase (CYP51A1), A. flavus (1R51), and E. coli (4XO8) to search the binding affinity and interactions of MDM esters with the receptor proteins. ADMET prediction also performed to evaluate the absorption, metabolism, and toxic properties of SEs. Structure activity relationship (SAR) study along with in vitro, and in silico results indicated that amongst the acyl chains, caproic chain (C6) in combination with acetic (C2), caprylic (C8) or lauric (C12) chains were found the most promising against fungal pathogens. Columnar self-assembly, electrochemical and luminescence properties of basket-shaped liquid crystalline derivatives of Schiff-base-moulded p-tert-butyl-calix[4]arene

A new family of supramolecular liquid crystalline compounds with a calix[4]arene core and two linking groups appended on four sides, exhibiting a wide range of hexagonal columnar phases, has been synthesised and well characterised. The liquid crystal behaviour of the novel materials was studied by using polarising optical microscopy (POM), differential scanning calorimetry (DSC) and thermal gravimetric analysis (TGA), and was further confirmed using high-temperature powder X-ray diffraction (XRD). An electrochemical study was carried out using cyclic voltammetry (CV) and the results were compared with theoretical values obtained from a DFT study. These compounds showed light sky-blue luminescence in solution and as a thin film under long-wavelength UV light. It was observed that all the prepared target compounds exhibited enantiotropic mesophases with good thermal stability. The structural and conformational characterisation of these new compounds was achieved using various spectroscopic techniques, including FT-IR, H-1 NMR, C-13 NMR and MALDI-TOF. The relationship between the structure and the mesomorphic behaviour of the compounds containing an ethyl bridge with linked ester groups (5a-5d) is discussed. This new class of supramolecular derivatives is promising, especially considering the emissive nature and stabilisation of their columnar hexagonal mesophases, and their high range of thermal stability.

CO2-assisted co-pyrolysis of textile dyeing sludge and hyperaccumulator biomass: Dynamic and comparative analyses of evolved gases, bio-oils, biochars, and reaction mechanisms

The CO2-activated co-pyrolysis technology presents promising potential to mitigate the environmental pollution and climate change. The dynamic analyses of evolved syngas, bio-oils, biochars, interaction effects, and reaction mechanisms of the co-pyrolysis of textile dyeing sludge (TDS) and Pteris vittata (PV) (hyperaccumulator biomass) were characterized and quantified comparatively in the three atmospheres. In the CO2-assisted atmosphere, the gasification of PV began to prevail between 600 and 900 degrees C, while in the N-2 atmosphere, PV and TDS were stable at 750 degrees C. The CO2-assisted co-pyrolysis reduced the apparent activation energy. The higher CO2 concentration during gasification led to the higher activation energy. The CO emission level of the CO2 and mixed atmospheres was almost 20 and 14 times that of the N-2 atmosphere, respectively. The CO release from the CO2 atmosphere was 1.4 times that from the mixed atmosphere. CO2 significantly changed the production pathway of biochar in the N-2 atmosphere, as was evidenced by the enhanced temperature sensitivity of O-C = O/hydroxy (- OH) in ester. Our findings research can provide new insights into the effectiveness of the CO2-assisted co-pyrolysis associated with reduced costs and hazardous wastes.

Effect of split fuel injection strategies on the diverse characteristics of CRDI diesel engine operated with tamarind biodiesel

The demand for petroleum fuels has been rapidly increasing owing to the wide use of automobiles and other agricultural engines which prefer diesel as a major fossil fuel. This research work investigates the effect of change in the fuel injection strategies on CRDI engine under various loads with tamarind seed methyl ester (TSME) 20% biodiesel blend. Initially, the diesel fuel is injected at 600 bar with 30% pilot injection (P.I) and 100% main injection (M.I). Later, TSME20 is injected at the same pressure with the two injection strategies mentioned. This study discloses that the BTE has boosted by 4.79% with 30% pilot injection than 100% main injection for the tested biodiesel. Further, TSME20 biodiesel at 30% pilot injection has shown significant reduction of hydrocarbons by 12.16%, carbon monoxide by 32.14% and smoke emissions by 19.71% when compared with 100% diesel main injection at peak load conditions. However, the pilot injection for the tested fuels has exposed slight enhancement in NOX emissions, which may be reduced by exhaust gas recirculation (EGR). Hence, the use of 30% pilot injection of TSME20 biodiesel is recommended for diesel engines as an alternative fuel which has improved performance along with reduced emissions.

Soil microbiomes mediate degradation of vinyl ester-based polymer composites

Polymer composites are attractive for structural applications in the built environment due to their lightweight and high strength properties but suffer from degradation due to environmental factors. While abiotic factors like temperature, moisture, and ultraviolet light are well studied, little is known about the impacts of naturally occurring microbial communities on their structural integrity. Here we apply complementary time-series multi-omics of biofilms growing on polymer composites and materials characterization to elucidate the processes driving their degradation. We measured a reduction in mechanical properties due to biologically driven molecular chain breakage of esters and reconstructed 121 microbial genomes to describe microbial diversity and pathways associated with polymer composite degradation. The polymer composite microbiome is dominated by four bacterial groups including the Candidate Phyla Radiation that possess pathways for breakdown of acrylate, esters, and bisphenol, abundant in composites. We provide a foundation for understanding interactions of next-generation structural materials with their natural environment that can predict their durability and drive future designs. The effect of microbial communities on the degradation of polymer composites has not been widely studied. Here, a multi-omics and materials characterization approach reveals the time-dependent effect of naturally occurring microbes on the degradation of vinyl ester composites.

Corrosion-Resistant Cross-Linked Cyanate Ester Coatings Doped by Graphene Oxide for Use in High-Temperature Marine Environments

Organic coatings are widely used to protect metals from corrosion in high-temperature marine environments. Compared with inorganic or metal coatings, organic coatings exhibit favorable properties, such as a reasonable cost, adequate flexibility, and easy fabrication. However, most organic coatings are limited to relatively short protection times and thus fail to meet the increasing demands for industrial applications. In this paper, graphene oxide (GO) was added to cyanate ester (CE) coatings as a corrosion inhibitive filler. Electrochemical analysis shows that a 3 wt % GO/CE coating has favorable corrosion protection properties with a low-frequency resistance higher than 10(6) Omega cm(2) and a corrosion rate of 632.9 x 10(-6) mm per year after 1320 h of immersion in a high-temperature marine environment. Top-view and cross-sectional SEM images identify the electrochemical results. Furthermore, chemical analyses by Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy demonstrate that hydroxyl groups of GO can react with cyanate groups, and therefore, a robust cross-linked structure was formed in the GO/CE coating. The synergistic effect of the physical shielding of GO and the robust cross-linked structure makes the GO/CE coating a promising choice for corrosion protection of steel in high-temperature marine environments. Our study also illustrates a novel method for preparing other chemically cross-linked composite coatings.
