The present study was designed to investigate the consumer preference and the quality attributes of five brands of Chinese traditional first-grade fava pastes, following which their quality differences were evaluated in order to identify the key drivers or factors, which can be applied to explain the behavior of consumer preferences. Consumers ranked these samples based on the 9-point method. An obvious preference for sample P1 was observed with 35% of participants settling on extremely liked option, giving the said sample the highest overall preference score of 7.8 ± 0.2. Further sensory and physicochem. properties, and volatiles were conducted to identify the major drivers of the consumer behavior. Sensory evaluation showed that sample P1 also produced the highest overall sensory score (85 ± 3). Though all the samples exhibited similar physicochem. attributes, significant differences were observed in the species and the concentration of volatile compounds Accordingly, the content of free amino acids (FAAs) and low-mol.-weight (MW) fraction (≤5 kDa) represented the trend that fell in line with sensory score. PLS2 correlation anal. indicated that physicochem. indexes and volatiles could be used to explain the consumer preference, which was likely to be synergistically affected by FAAs, protein, amino nitrogen, dietary fiber, Ca, P, Fe, δ-Ε, and some volatiles, especially the pos. drivers of FAAs and low MW proportion to sample P1. The work would not only provide valuable information for grading of fava pastes in the industry but also guide actual producing toward the expectation of consumers, especially increasing those pos. drivers.

A novel nucleating agent (NA) 1-N’,3-N’-dibenzoylbenzene-1,4-dicarbohydrazide (PTADH-Ph-3) for isotactic polypropylene (iPP) was synthesized and the structure of the synthesized product was characterized by Fourier transform IR spectroscopy (FT-IR), proton NMR spectroscopy (1H-NMR) and high resolution mass spectrometry (HRMS). The results showed that the target compound was successfully synthesized. Differential scanning calorimeter (DSC) was used to study the influence of the addition concentration of PTADH-Ph-3 and the final heating temperature (Tf) on the crystallization behavior of iPP. The results showed that the crystallization behavior of nucleated iPP had no relationship with Tf no matter how much PTADH-Ph-3 was added. This shows that PTADH-Ph-3 is not a solution-type NA, so its nucleation mechanism belongs to the traditional heterogeneous crystallization The nucleation mechanism of PTADH-Ph-3 in iPP was explored through Materials Studio (MS) simulation. Firstly, the mol. dynamics (MD) of PTADH-Ph-3 and iPP were calculated mainly by MS. The calculation results showed that the interaction force between PTADH-Ph-3 and iPP mols. was 14.447 kcal mol-1, indicating that PTADH-Ph-3 has a better adsorption effect on the iPP surface, which can promote the epitaxial crystallization of iPP. The calculated results show that the lattice mismatch between PTADH-Ph-3 and iPP is only 0.03% and 2.28%, which is far lower than the strict upper limit of 10-15% geometric mismatch in the crystal lattice matching theory.

The European grapevine moth, Lobesia botrana, uses (E,Z)-7,9-dodecadienyl acetate as its major sex pheromone component. Through in vivo labeling experiments we demonstrated that the doubly unsaturated pheromone component is produced by Δ11 desaturation of tetradecanoic acid, followed by chain shortening of (Z)-11-tetradecenoic acid to (Z)-9-dodecenoic acid, and subsequently introduction of the second double bond by an unknown Δ7 desaturase, before final reduction and acetylation. By sequencing and analyzing the transcriptome of female pheromone glands of L. botrana, we obtained 41 candidate genes that may be involved in sex pheromone production, including the genes encoding 17 fatty acyl desaturases, 13 fatty acyl reductases, 1 fatty acid synthase, 3 acyl-CoA oxidases, 1 acetyl-CoA carboxylase, 4 fatty acid transport proteins and 2 acyl-CoA binding proteins. A functional assay of desaturase and acyl-CoA oxidase gene candidates in yeast and insect cell (Sf9) heterologous expression systems revealed that Lbo_PPTQ encodes a Δ11 desaturase producing (Z)-11-tetradecenoic acid from tetradecanoic acid. Further, Lbo_31670 and Lbo_49602 encode two acyl-CoA oxidases that may produce (Z)-9-dodecenoic acid by chain shortening (Z)-11-tetradecenoic acid. The gene encoding the enzyme introducing the E7 double bond into (Z)-9-dodecenoic acid remains elusive even though we assayed 17 candidate desaturases in the two heterologous systems.

The aim of this study was to evaluate the effect of β-glucosidase (Lb0241) derived from Lactobacillus brevis on the aroma of strawberry wine. The recombinant Lb0241 was cloned and expressed in Escherichia coli BL21 (DE3). The effect of temperature, pH, and additives on the activity of Lb0241 and its substrate specificity and kinetic parameters were analyzed. Furthermore, the effects of Lb0241 on the aroma properties of strawberry wine were evaluated by SPME-GC-MS and sensory evaluation. Lb0241 showed remarkable specificity for β-glucoside and displayed the highest activity at pH 5.0 and 30°C, with a relative activity of 55% in 12% ethanol. Ni2+ and K+ could increase the activity of Lb0241, but Fe2+, urea, 2-hydroxy-1-ethanethiol, and sodium dodecyl sulfate showed the opposite effect. Lb0241 could increase the aroma-active volatiles of esters, terpenoids, and phenols, and decrease alcs. and volatile acids. Overall, Lb0241 showed promising prospects in terms of aroma release in strawberry wine. Practical applications : A notable part of non-volatile glycosidic precursors present in fruit wine constitutes a reserve of potential aroma-active mols. Thus, the hydrolysis of aromatic precursors could significantly enhance the aroma of fruit wine. Our results showed that the new β-glucosidase Lb0241, which was derived from Lactobacillus brevis T61, exhibited strong acid and ethanol resistance. Moreover, Lb0241 could effectively hydrolyze aromatic precursors and enhance the level of aroma compounds in strawberry wine. Therefore, Lb0241 has potential application prospects and economic value to improve the flavor of fruit wine.

Idiopathic pulmonary fibrosis (IPF) is a progressive fibrotic disease with pathophysiol. characteristics of transforming growth factor-β (TGF-β), and reactive oxygen species (ROS)-induced excessive fibroblast-to-myofibroblast transition and extracellular matrix deposition. Macrophages are closely involved in the development of fibrosis. Nuclear factor erythroid 2 related factor 2 (Nrf2) is a key mol. regulating ROS and TGF-β expression. Therefore, Nrf2 signaling modulation might be a promising therapy for fibrosis. The inhalation-based drug delivery can reduce systemic side effects and improve therapeutic effects, and is currently receiving increasing attention, but direct inhaled drugs are easily cleared and difficult to exert their efficacy. Therefore, we aimed to design a ROS-responsive liposome for the Nrf2 agonist di-Me fumarate (DMF) delivery in the fibrotic lung. Moreover, we explored its therapeutic effect on pulmonary fibrosis and macrophage activation. We synthesized DMF-loaded ROS-responsive DSPE-TK-PEG@DMF liposomes (DTP@DMF NPs). DTP@DMF NPs had suitable size and neg. zeta potential and excellent capability to rapidly release DMF in a high-ROS environment. We found that macrophage accumulation and polarization were closely related to fibrosis development, while DTP@DMF NPs could attenuate macrophage activity and fibrosis in mice. RAW264.7 and NIH-3T3 cells coculture revealed that DTP@DMF NPs could promote Nrf2 and downstream heme oxygenase-1 (HO-1) expression and suppress TGF-β and ROS production in macrophages, thereby reducing fibroblast-to-myofibroblast transition and collagen production by NIH-3T3 cells. In vivo experiments confirmed the above findings. Compared with direct DMF instillation, DTP@DMF NPs treatment presented enhanced antifibrotic effect. DTP@DMF NPs also had a prolonged residence time in the lung as well as excellent biocompatibility. DTP@DMF NPs can reduce macrophage-mediated fibroblast-to-myofibroblast transition and extracellular matrix deposition to attenuate lung fibrosis by upregulating Nrf2 signaling. This ROS-responsive liposome is clin. promising as an ideal delivery system for inhaled drug delivery.
