With five histone deacetylase (HDAC) inhibitors approved for cancer treatment, proteolysis-targeting chimeras (PROTACs) for degradation of HDAC are emerging as an alternative strategy for HDAC-targeted therapeutic intervention. Herein, three bestatin-based hydroxamic acids (P1, P2 and P3) were designed, synthesized and biol. evaluated to see if they could work as HDAC degrader by recruiting cellular inhibitor of apoptosis protein 1 (cIAP1) E3 ubiquitin ligase. Among the three compounds, the bestatin-SAHA hybrid P1 exhibited comparable even more potent inhibitory activity against HDAC1, HDAC6 and HDAC8 relative to the approved HDAC inhibitor SAHA. It is worth noting that although P1 could not lead to intracellular HDAC degradation after 6 h of treatment, it could dramatically decrease the intracellular levels of HDAC1, HDAC6 and HDAC8 after 24 h of treatment. Intriguingly, the similar phenomenon was also observed in the HDAC inhibitor SAHA. Cotreatment with proteasome inhibitor bortezomib could not reverse the HDAC decreasing effects of P1 and SAHA, confirming that their HDAC decreasing effects were not due to protein degradation Moreover, all three bestatin-based hydroxamic acids P1, P2 and P3 exhibited more potent aminopeptidase N (APN, CD13) inhibitory activities than the approved APN inhibitor bestatin, which translated to their superior anti-angiogenic activities. Taken together, a novel bestatin-SAHA hybrid was developed, which worked as a potent APN and HDAC dual inhibitor instead of a PROTAC.

Summary : The antioxidant effects of gallic acid (GA) and its alkyl esters including Me gallate, Et gallate, Pr gallate, Bu gallate (GA-C4), octyl gallate, lauryl gallate, hexadecyl gallate and octadecyl gallate in frying oil (soybean oil) and oyster during frying were investigated. Rancimat induction period (RIP) assay indicated GA and its alkyl esters effectively improved the oxidative stability of soybean oil according to the ‘polar paradox’; i.e., the antioxidant effects of the polyphenols decreased in lipid system with the increasing of hydrophobicity. Results also indicated that GA and its alkyl esters all effectively retarded lipid oxidation in oyster tissue according to parameters including RIP, peroxide value and ESR assays. Moreover, the antioxidant efficiency in oyster increased with the alkyl chain length until GA-C4, and thereafter, the antioxidant effects decreased, similar to the ‘cut-off effect’. This study provides basic data for improving the oxidative stability of seafood during frying.

In addition, the host-guest complexation of NP5 extended the initiating wavelength of Iod from ultrashort UV to near-UV, which could better match the environment-friendly LED light source. It is anticipated that a supra-photoinitiator may open a new route for designing novel photoinitiators with high performance.

A tetraester, i.e. tetra-Me piperazine-N,N’-bis(2-methylene butanedioate) (TMPB), was synthesized through a Michael addition of di-Me itaconate and piperazine at 90°C. Bulk polycondensation of TMPB with butanediamine or hexamethylenediamine was conducted at 130-170°C under atm. pressure, and several cPITAs with tunable properties were prepared The Michael addition and the polycondensation were monitored by FT-IR and 1H NMR spectra. The cPITA films were characterized by DSC, WAXS, TGA, dynamic mech. anal., and tensile test. cPITAs exhibited Tg ranging from 58 to 72°C, tensile strength up to 85 MPa, and strain at break from 9% to 17%.

The InNi3C0.5/Ni-foam catalyst is obtainable by hydrothermal growth of NiC2O4 onto the Ni-foam, impregnation with In2O3 precursor, subsequent calcination and carburization in a syngas. Despite DMO cascade hydrogenation to MG, then to EG, and finally to EtOH, such catalyst hydrogenates DMO dexterously until to EG with a high turnover frequency of 636 h-1, because the 3Ni-In and 3Ni-C sites on InNi3C0.5(1 1 1) effectively activate DMO but hinder EG over-hydrogenation to EtOH. Favorable reaction pathway on the InNi3C0.5(1 1 1) surface predicted theor. is DMO* → CH3OCOCO* → CH3OCOCHO* → CH3OCOCHOH* → MG* → CH3OCHOCH2OH* → CHOCH2OH* → HOCHCH2OH* → EG*. Moreover, the neutral Ni-foam diminishes the formation of ethers and diols. The volatile composition of dichloromethane extracts of preputial glandular secretions (PGS) from intact and castrated male Brandt’s voles Lasiopodomys brandtii were analyzed by using gas chromatog.-mass spectrometry (GC-MS). Thirty-three volatile compounds were detected and were present in nearly all scent donors, and 27 had previously been reported as insect pheromone components. Castration did not eliminate any compounds; however, it significantly suppressed the relative quantity of ten early-eluting, low mol. weight components. These included seven saturated and unsaturated acetates, one hexanoate, and two saturated octanoates. In particular, E,E-farnesyl acetate (FA) was the most abundant compound in the PGS. FA and other components might be candidates for male-produced pheromones. Two-choice behavioral assays of females revealed that low concentrations of raw PGS (0.056%) and FA (5 ppm in water v/w) were attractive, whereas high concentrations (0.56% and 50 ppm, resp.) were repellent. This suggested that FA might act as a male pheromone in a dosage-dependent manner. Extremely high variability of the relative quantities of almost all detected compounds among individuals indicated the inter-individual dissimilarities of PGS and consequent possibilities to communicate individuality to other conspecific members.

The generality of the approach was further demonstrated by the stereoselective creation of vicinal quaternary carbon stereocenters with both centers in acyclic systems using acyclic β-ketoesters as coupling partners. Computational studies elucidate the origins of both diastereo- and enantioselectivity.
