Rigidly and covalently linked porphyrin quinones are well-suited as biomimetic model compounds for studying the photoinduced electron transfer (PET) reaction occurring in primary processes of photosynthesis. In this context, the synthesis of new porphyrin quinones with a cis- or trans-1,4-disubstituted cyclohexylene bridge linking the electron donor and the electron acceptor is reported. To study the dependence of the PET rate of the difference of the free enthalpy of the PET reaction, four quinones with different structures and therefore redox potentials were used as electron acceptor components. As a whole, two series of each four new cis- and trans-1,4-cyclohexylene-bridged porphyrin quinones with variable acceptor strength were synthesized. The most important synthetic steps comprised the free radical addition of the ester functionalized cyclohexylene bridge to the quinone, reduction of the ester to the alc. group with lithium borohydride or DIBALH, oxidation of the alc. to the corresponding aldehyde with PCC or TEMPO (2,2,6,6-tetramethylpiperidine-1-oxyl)/NaOCl, and condensation of these aldehydes with pyrrole and 4-methylbenzaldehyde under equilibrium conditions. Anal. of the 1H NMR spectra unambiguously indicated the chair conformation for the cyclohexane ring of all porphyrin precursors and trans-cyclohexane-bridged porphyrin quinones, whereas the cis-cyclohexane-bridged porphyrin quinones had the cyclohexane ring in the unusual twist-boat conformation. This was addnl. confirmed by an X-ray crystal structure of one of the cis-porphyrin quinones and the corresponding trans-porphyrin quinone. NOE experiments gave information about the spatial arrangement of the diastereomeric target compounds in solution

Liver pathol. changes are as high as 21%-78% in diabetic patients, and treatment options are lacking. Liraglutide is a glucagon-like peptide-1 (GLP-1) receptor that is widely used in the clinic and is approved to treat obesity and diabetes. However, the specific protection mechanism needs to be clarified. In the present study, db/db mice were used to simulate Type 2 diabetes mellitus (T2DM), and they were i.p. injected daily with liraglutide (200μg/kg/d) for 5 wk. Hepatic function, pathol. changes, oxidative stress, iron levels, and ferroptosis were evaluated. First, liraglutide decreased serum AST and ALT levels, and suppressed liver fibrosis in db/db mice. Second, liraglutide inhibited the ROS production by upregulating SOD, GSH-PX, and GSH activity as well as by downregulating MDA, 4-HNE, and NOX4 expression in db/db mice. Furthermore, liraglutide attenuated iron deposition by decreasing TfR1 expression and increasing FPN1 expression. At the same time, liraglutide decreased ferroptosis by elevating the expression of SLC7A11 and the Nrf2/HO-1/GPX4 signaling pathway in the livers of db/db mice. In addition, liraglutide decreased the high level of labile iron pools (LIPs) and intracellular lipid ROS induced by high glucose in vitro. Therefore, we speculated that liraglutide played a crucial role in reducing iron accumulation, oxidative damage and ferroptosis in db/db mice.

Numerous emerging applications in modern society require humidity sensors that are not only sensitive and specific but also durable and intelligent. However, conventional humidity sensors do not have all of these simultaneously because they require very different or even contradictory design principles. Here, inspired by camel noses, we develop a porous zwitterionic capacitive humidity sensor. Relying on the synergistic effect of a porous structure and good chem. and thermal stabilities of hygroscopic zwitterions, this sensor simultaneously exhibits high sensitivity, discriminability, excellent durability, and, in particular, the highest respond speed among reported capacitive humidity sensors, with demonstrated applications in the fast discrimination between fresh, stale, and dry leaves, high-resolution touchless human-machine interactive input devices, and the real-time monitoring humidity level of a hot industrial exhaust. More importantly, this sensor exhibits typical synapse behaviors such as paired-pulse facilitation due to the strong binding interactions between water and zwitterions. This leads to learning and forgetting features with a tunable memory, thus giving the sensor artificial intelligence and enabling the location of water sources. This work offers a general design principle expected to be applied to develop other high-performance biochem. sensors and the next-generation intelligent sensors with much broader applications.

Background: In the perspective of ART-free HIV remission, vertically infected children treated with suppressive ART from early infancy represent an optimal population model to better understand the genetic complexity of the reservoir. Objectives: To evaluate the proportion of defective viral population and the genotypic resistance patterns in cell-associated HIV DNA. Methods: In a cohort including 93 ART-treated vertically HIV-infected (VHIV) children in Mali with plasma HIV-1 RNA ≤50 copies/mL for at least 6 mo, we studied total HIV DNA, percentage of defective genomes and resistance by reverse transcriptase and protease bulk sequencing from whole blood in dried blood spots. Results: Children had a median age of 9.9 years at the time of inclusion (IQR = 7.6-13.4) and 3.3 years (IQR = 2-7) at ART initiation; median ART duration was 5.5 years (IQR = 3.7-7.3). The median level of total HIV DNA was 470 copies/106 cells with one patient presenting undetectable HIV DNA (<66 copies/106 cells). We observed the presence of at least one stop codon in viruses from 34 patients (37%). The presence of stop codons was not correlated with the level of HIV DNA or duration of ART. We showed a high prevalence of HIV-1 resistance in DNA with 26% of children harbouring virus resistant to at least one NRTI and 40% to at least one NNRTI. Conclusions: While these VHIV children were successfully treated for a long time, they showed high prevalence of resistance in HIV DNA and a moderate defective HIV reservoir. This method is based on formal C-H azidation of 1,3-disubstituted benzenes via regioselective borylation followed by deborylative azidation with subsequent transformations, such as that of a one-carbon unit on the benzene ring to an azidomethyl group. The functional groups of the diazido compounds were efficiently transformed to various connecting groups, including carboxy, (succinimidyloxy)carbonyl, hydroxymethyl, formyl, bromomethyl, tosylthiomethyl, ethynyl, diazoacetyl, bromoacetyl, boryl, hydroxy, aminocarbonyl, amino, and isothiocyanato groups, leaving the azido groups untouched. Several diazido building blocks were used to prepare diazido compounds by forming amide, thiourea, and sulfide bonds via conjugation at the connecting groups. These results show that the method described here would facilitate diazido probe syntheses and bistriazole library construction.

The purpose of this study was to investigate the preparation of formulated water- in-soybean oil-in-water emulsions by repeated premix membrane emulsification method using a cellulose acetate membrane. The effect of selective membrane emulsification process parameters (concentration of the emulsifiers, number of passes of the emulsions through the membrane and storage temperature) on the properties and stability of the developed emulsions were also investigated. 1, 3, 6, 8-pyrenetetrasulfonic acid tetrasodium salt (PTSA) was used as a hydrophilic model ingredient for the encapsulation of bioactive substances. W/O emulsions with 7 wt% (weight percentage) PGPR displays homogeneous and very fine dispersions, with the median diameter at 0.640 μm. Meanwhile, emulsions prepared by membrane emulsification (fine W/O/W) showed the highest stability at Tween 80 concentrations of 0.5 weight% (weight percentage). It concluded that at 7 weight% (weight percentage) PGPR concentration and 0.5 weight% (weight percentage) Tween 80 concentrations, the most uniform particles with min. mean size of oil drops (9.926 μm) were obtained after four passes through the membrane. Thus, cellulose acetate membrane can be used for preparing a stable W/O/W emulsions by repeated premix ME due to low cost and relatively easy to handle.

The rates of phenylephrine formation on photolysis were estimated from the decay rates of aci-nitro intermediates detected by absorbance between 380 and 500 nm. IV displayed the highest quantum yield (0.28) and most rapid photolysis rate (1980 s-1) measured under near physiol. conditions, pH 8.0, 22°. Biol. properties of the compounds were examined in smooth muscle from rat caudal artery. Laser pulse photolysis of IV at 347 nm initiated a maximal contraction in Krebs buffer, pH 7.1, 25°, that mimicked the response to 50 μM phenylephrine but was faster in onset. Photoinitiated contractions were characterized by a delay of 0.93 s followed by a rising phase with a 10-90% rise time of 3.56 s. Responses were fully blocked by the α1-selective antagonist prazosin. Similar results were obtained using I-III, but a slowly developing sustained phase of smooth muscle contraction was altered in the presence of I-III. IV represents a useful new biol. inactive caged phenylephrine that should facilitate temporally and spatially resolved studies of α1-adrenergic receptor mechanisms in cells and tissues.
