The reactive-vapor permeation-distillation (R-VP-D) hybrid configuration is a newly designed inter-integration concept firstly proposed for the production of Et levulinate (EL) by esterification of ethanol and levulinic acid in this paper. In the R-VP-D configuration, the VP membrane module is inserted to the reaction section of the reactive distillation (RD) column, of which the product water is in-situ removed by the vapor permeation (VP) to promote the performance of esterification RD process. The proposed R-VP-D configuration is modeled by Aspen Plus V8.4 using the two-dimensional VP model built in ACM, and the inserting position of single or double VP modules is found by contrast trials, where both the conversion rate and purity of product can reach the industrial production standards As a result, the proposed novel R-VP-D configuration is feasible, with > 21.6% and 31.4% reduction in total annual cost and total duty as compared to the configuration which only applies an extra distillation column to separation and recycle the excess ethanol without azeotrope separation, and with 13.5 and 21.5% reduction in TAC and total duty as compared to the hybrid configuration where the vapor permeation is coupled outside the RD column. The result also shows the advantage of the proposed R-VP-D process for potentially leading to the development of new integration processes and applications.

Diabetic foot ulcer (DFU) is an invariably common complication of diabetes, characterized by delayed wound healing process and increased inflammation. Evidence has indicated that paeoniflorin exerts an anti-inflammatory effect in diabetic retinopathy. The current work was aimed to investigate the effect of paeoniflorin on inflammation and wound healing in DFU. DFU rat models by streptozotocin and skin biopsy punch, as well as high glucose-treated human immortalized keratinocytes (HaCaT) were established. Levels of blood glucose, wound contraction and proinflammatory cytokine were detected after paeoniflorin administration. Several essential targets associated with the NF-κB and Nod-like receptor protein-3 (NLRP3) signaling pathways were examined Results showed markedly down-regulation of interleukin (IL)-1β IL-18 and tumor necrosis factor-alpha in paeoniflorin-treated DFU rats. Paeoniflorin decreased the expression levels of chemokine receptor CXCR2, nuclear NF-κB and p-IκB (Ser36), as well as increased IκB level. Histol. anal. and immunostaining showed lower inflammatory cells with decreased NLRP3 and cleaved caspase-1 levels following paeoniflorin treatment. Further in vitro evidence confirmed that paeoniflorin efficiently inhibited NLRP3 and NF-κB-mediated inflammation in DFU by inhibiting CXCR2. These findings are suggestive of greatly attenuated wound inflammation and better wound healing in paeoniflorin-treated DFU rats. Our study demonstrates that paeoniflorin is a potential therapeutic agent for DFU.

Herein, a novel method for molecularly imprinted polymers (MIPs) using methacrylic acid functionalized beta-cyclodextrin (MAA-β-CD) monomer is presented, which was designed as a potential water-compatible composite for the controlled release of atropine (ATP). The molecularly imprinted microspheres with pH-sensitive characteristics were fabricated using thermally-initiated precipitation polymerization, employing ATP as a template mol. The effects of different compounds and concentrations of crosslinking agents were systematically investigated. Uniform microspheres were obtained when the ratio between ATP, MAA-β-CD, and trimethylolpropane trimethacrylate (TRIM) was 1:4:20 (mol/mol/mol) in polymerization system. The ATP loading equilibrium data was best suited to the Freundlich and Langmuir isotherm models. The in vitro drug release study was assessed under simulated oral administration conditions (pH 1.5 and 7.4). The potential usefulness of MIPs as drug delivery devices are much better than non-molecularly imprinted polymers (NIPs). The study shows that the prepared polymers are a pH stimuli-responsive system, which controlled the release of ATP, indicating the potential applications in the field of drug delivery.

The ketogenic diet (KD) is a restrictive therapeutic diet, distinguished by being hyperlipidic, normoproteic, and hypoglucidic. This diet simulates biochem. changes related to fasting periods to achieve systemic ketosis. The metabolic particularities of glioma tumors motivated the rise in investigations and nutritional strategies, such as KD, to modulate the glycemic response as a treatment. This systematic review followed the PRISMA recommendations and was published in PROSPERO, with the identification CRD42021264173. The databases used were EMBASE, PubMed/Medline, Scopus, and Web of Science, and the studies were analyzed using the web-based application Rayyan. To analyze the risk of bias, Cochrane RevMan 5 software was used. For the anal. and treatment of statistical data, Microsoft Excel was used. A total of nine original articles were included. Data on survival, symptomol., and quality of life were collected. Mean overall survival was 15.9 mo. Constipation and fatigue were the most reported symptoms. In 44.4% of the studies, an improvement in the quality of life was found. The KD is supported by most published studies as an effective therapy in the treatment of malignant gliomas due to its pos. effects on patient survival. It was not possible to conclude the effectiveness of KD on quality of life.

The KNOG-1101 study showed improved 2-yr progression-free survival (PFS) with temozolomide during and after radiotherapy compared to radiotherapy alone for patients with anaplastic gliomas. This trial investigates the effect of concurrent and adjuvant temozolomide on health-related quality of life (HRQoL). In this randomized, open-label, phase II trial, 90 patients with World Health Organization grade III glioma were enrolled across multiple centers in South Korea between March 2012 to Feb. 2015 and followed up through 2017. The European Organization for Research and Treatment of Cancer Quality of Life Questionnaire 30 (EORTC QLQ-C30) and 20-item EORTC QLQ-Brain Neoplasm (QLQ-BN20) were used to compare HRQoL between patients assigned to concurrent chemoradiotherapy with temozolomide followed by 6 cycles of adjuvant temozolomide (arm A) and radiotherapy (RT) alone (arm B). Of the 90 patients in the study, 84 patients (93.3%) completed the baseline HRQoL questionnaire. Emotional functioning, fatigue, nausea and vomiting, dyspnea, constipation, appetite loss, diarrhea, seizures, itchy skin, drowsiness, hair loss, and bladder control were not affected by the addition of temozolomide. All other items did not differ significantly between arm A and arm B throughout treatment. Global health status particularly stayed consistent at the end of adjuvant temozolomide (p = 0.47) and at the end of RT (p = 0.33). The addition of concurrent and adjuvant temozolomide did not show neg. influence on HRQoL with improvement of PFS for patients with anaplastic gliomas. The absence of systematic and clin. relevant changes in HRQoL suggests that an overall long-term net clin. benefit exists for concurrent and adjuvant temozolomide.

The scope of this method tolerates various functionalized amino acids such as His, Ser, Trp, Tyr, Glu, etc., while the only limitations uncovered so far are restricted to cysteine and free amine residues. Monoalkylated bis(4-hydroxyphenyl)methanes are reported to show weak binding affinity for estrogen receptor (ER). The authors hypothesized that introduction of appropriately located hydrophobic substituents in these compounds would increase the binding affinity. Indeed, the authors found that bis(4-hydroxyphenyl)methane bearing a 3,3-dimethylcyclohexyl group I shows potent ERα binding affinity, comparable to that of estradiol. Bulkier substituents could be introduced at the 3,3-position without decreasing the affinity. However, the position of the substituents was critical: the 4,4-dimethylcyclohexyl derivative showed very weak binding affinity. The compounds with high ER-binding affinity showed predominantly agonistic activity, together with weak antagonistic activity at high concentration, in cell proliferation assay with human breast cancer cell line MCF-7. Further structure-function studies of these compounds and their derivatives might lead to the development of more selective and potent estrogen receptor modulators.
