T lymphocyte and macrophage infiltration in the aortic wall is critical for abdominal aortic aneurysm (AAA). However, how T lymphocytes interact with macrophages in the pathogenesis of AAA remains largely uncharacterized. In an elastase-induced murine AAA model, we first found that the expression of pyruvate kinase muscle isoenzyme 2 (PKM2), the last rate-limiting enzyme in glycolysis, was increased in infiltrated T lymphocytes of vascular lesions. T lymphocyte-specific PKM2 deficiency in mice (LckCrePKM2fl/fl) or i.p. administration of the sphingomyelinase inhibitor GW4869 caused a significant attenuation of the elastase-increased aortic diameter, AAA incidence, elastic fiber disruption, matrix metalloproteinases (MMPs) expression, and macrophage infiltration in the vascular adventitia compared with those in PKM2fl/fl mice. Mechanistically, extracellular vesicles (EVs) derived from PKM2-activated T lymphocytes elevated macrophage iron accumulation, lipid peroxidation, and migration in vitro, while macrophages treated with EVs from PKM2-null T lymphocytes or pretreated with the lipid peroxidation inhibitors ferrostatin-1 (Fer-1), liproxstatin-1 (Lip-1), or the iron chelating agent deferoxamine mesylate (DFOM) reversed these effects. In vascular lesions of elastase-induced LckCrePKM2fl/fl mice with AAA, the oxidant system weakened, with downregulated 4-hydroxynonenal (4-HNE) levels and strengthened antioxidant defense systems with upregulated glutathione peroxidase 4 (GPX4) and cystine/glutamate antiporter solute carrier family 7 member 11 (Slc7a11) expressions in macrophages. High-throughput metabolomics showed that EVs derived from PKM2-activated T lymphocytes contained increased levels of polyunsaturated fatty acid (PUFA)-containing phospholipids, which may provide abundant substrates for lipid peroxidation in target macrophages. More importantly, upregulated T lymphocyte PKM2 expression was also found in clin. AAA subjects, and EVs isolated from AAA patient plasma enhanced macrophage iron accumulation, lipid peroxidation, and migration ex vivo. Therefore, from cell-cell crosstalk and metabolic perspectives, the present study shows that PKM2-activated T lymphocyte-derived EVs may drive AAA progression by promoting macrophage redox imbalance and migration, and targeting the T lymphocyte-EV-macrophage axis may be a potential strategy for early warning and treating AAA.

Nowadays, the discovery and development of α-glucosidase inhibitors from natural products or their derivatives represents an attractive approach. Here we reported studies on a series of novel N-acyl-2-aminothiazoles fused (+)-nootkatone and evaluation for their α-glucosidase inhibitory activities. Most of (+)-nootkatone derivatives exhibited more potent α-glucosidase inhibitory ability than the pos. drug acarbose. In particular, compounds I and II showed the most promising α-glucosidase inhibitory ability with IC50 values of 13.2 and 13.8 μM. I and II also exhibited relatively low cytotoxicities towards normal LO2 cells. Kinetic study indicated that compounds I and II inhibited the α-glucosidase in a noncompetitive manner, and mol. docking results were in line with the noncompetitive characteristics that I and II did not bind to the known active sites (Asp214, Glu276 and Asp349). Based on our findings, these (+)-nootkatone derivatives could be used as antidiabetic candidates.

Lithium metal batteries are attractive for next-generation energy storage because of their high energy d. A major obstacle to their commercialization is the uncontrollable growth of lithium dendrites, which arises from complicated but poorly understood interactions at the electrolyte/electrode interface. In this work, we use a machine learning-based artificial neural network (ANN) model to explore how the lithium growth rate is affected by local material properties, such as surface curvature, ion concentration in the electrolyte, and the lithium growth rates at previous moments. The ion concentration in the electrolyte was acquired by Stimulated Raman Scattering Microscopy, which is often missing in past exptl. data-based modeling. The ANN network reached a high correlation coefficient of 0.8 between predicted and exptl. values. Further sensitivity anal. based on the ANN model demonstrated that the salt concentration and concentration gradient, as well as the prior lithium growth rate, have the highest impacts on the lithium dendrite growth rate at the next moment. This work shows the potential capability of the ANN model to forecast lithium growth rate, and unveil the inner dependency of the lithium dendrite growth rate on various factors.

The single-atom catalysts (SACs) supported on carbon-based materials are very promising for maximizing their electrocatalytic activity. However, carbon-based SACs are primarily bonded with carbons, resulting in inferior performance in the oxygen evolution reaction (OER). Herein, we develop a novel coordination adsorption strategy for the synthesis of a monodispersed Fe(NO)2 moiety anchored on the nitrogen sites of the nitrogen-doped graphene quantum dots (N-GQDs) to form a Fe(NO)2-N-GQDs complex as an efficient OER catalyst. The resultant Fe(NO)2-N-GQDs complex exhibits a highly stable overpotential of 270 mV at a c.d. of 10 mA cm-2 and a Tafel slope of 48 mV dec-1 together with long-term durability, which greatly outperforms the state-of-the-art ruthenium oxide. Our finding emphasizes the role of electron-transfer resistance changes during a simple synthesis method to enhance electrocatalytic efficiency. Therefore, this work will envision numerous opportunities for creating novel-type carbon-based SACs via nitrogen-metal coordination as highly robust OER catalysts.

The fatty acid elongase elongation of very long-chain fatty acids protein 2 (ELOVL2) controls the elongation of polyunsaturated fatty acids (PUFA) producing precursors for omega-3, docosahexaenoic acid (DHA), and omega-6, docosapentaenoic acid (DPAn-6) in vivo. Expectedly, Elovl2-ablation drastically reduced the DHA and DPAn-6 in liver mitochondrial membranes. Unexpectedly, however, total PUFAs levels decreased further than could be explained by Elovl2 ablation. The lipid peroxidation process was not involved in PUFAs reduction since malondialdehyde-lysine (MDAL) and other oxidative stress biomarkers were not enhanced. The content of mitochondrial respiratory chain proteins remained unchanged. Still, membrane remodeling was associated with the high voltage-dependent anion channel (VDAC) and adenine nucleotide translocase 2 (ANT2), a possible reflection of the increased demand on phospholipid transport to the mitochondria. Mitochondrial function was impaired despite preserved content of the respiratory chain proteins and the absence of oxidative damage. Oligomycin-insensitive oxygen consumption increased, and coefficients of respiratory control were reduced by 50%. The mitochondria became very sensitive to fatty acid-induced uncoupling and permeabilization, where ANT2 is involved. Mitochondrial volume and number of peroxisomes increased as revealed by transmission electron microscopy. In conclusion, the results imply that endogenous DHA production is vital for the normal function of mouse liver mitochondria and could be relevant not only for mice but also for human metabolism

The most important fragment ions in the spectra of 4 dispirohexadecanones could be explained by assuming the same types of fragmentation as found for the monospiro ketones. The composition of the fragment ions was checked by high resolution mass measurements.
