The unique combination of high-performance size-exclusion chromatog. with hyphenated electrospray ionization mass spectrometry (MS), atm. pressure chem. ionization-MS, and high-temperature gas chromatog.-MS techniques allows differentiating between radical coupling species and Diels-Alder cycloadducts. Targeted anal. of thermally degraded sunflower oils confirms the exact structures of various acyclic oligomers accompanied by less-abundant products of pericyclic transformations. A series of model experiments simulate the impact of dienophile nature on the course of Diels-Alder reactions. Thus, α-tocopherylquinone, δ-tocopherylquinone, and methyl-(E)-11-oxoundec-9-enoate are synthesized as naturally occurring dienophiles bearing electron-withdrawing groups. The geometry of poor dienophiles does not affect concerted cyclization, while the structure of electron deficient dienophiles can overcome low reactivity. In the absence of oxygen, heat-induced degradation of polyunsaturated triacylglycerols proceed predominantly via a radical pathway, whereas concerted reactions represent minor mechanisms. Sunflower oil triacylglycerol mols. in the system without propagation stage can be effectively protected by natural and/or synthetic antioxidants. Application of chelates is also recommended. However, antioxidant-derived quinones, such as α-tocopherylquinone, can enter the Diels-Alder reaction even more easily than dienophiles without electron-withdrawing groups. Unsaturated core aldehydes possess the same reactivity. Examination of the mechanism controlling high-temperature degradation of triacylglycerols is especially important for processing engineers in edible oil refineries and food technologists. New perspective may help them to minimize undesirable changes in polyunsaturated species.

Hyperbranched aliphatic polycarbonates (HBPCs) have attracted significant attention in the field of biomedical application owing to their abundant end groups, biocompatibility, and nontoxic degradation products. However, their practical application is hampered by tedious synthesis procedures. The present work described a novel organo-catalyzed “”A1+B2″”-ring-opening polymerization (ROP) using a monofunctional primary alc. (A1) as an initiator and a bicyclic carbonate monomer (B2) for the straightforward synthesis of HBPCs under mild conditions. No gelation was observed up to 90% conversion of the cyclic carbonate ring. HBPCs with molar masses in the range from 7 to 20 kg/mol were obtained. Based on the “”A1+B2″”-ROP, HBPCs bearing a variety of functionalities including alkene, alkyne, aldehyde, furan, azide, and mPEG groups, which are of great significance in the field of click chem., have been successfully prepared by using functional alc. initiators. This polymerization strategy allowed for precise control over the HBPC structure: (1) the hyperbranched polymer (HBP) backbone can be adjusted by design of a bicyclic carbonate monomer with various linkages; (2) the use of an appropriate initiator leads to the introduction of functional end groups. Overall, the “”A1+B2″”-ROP provides an efficient method for preparing a variety of HBPs like hyperbranched polyester, polyether, polyphosphate, and poly(amino acid) with complex architectures in a single step.

We show that DMF and various fumaric and maleic acid esters (FMAEs) enhance viral infection of cancer cell lines as well as human tumor biopsies with several oncolytic viruses (OVs), improving therapeutic outcomes in resistant syngeneic and xenograft tumor models. This results in durable responses, even in models otherwise refractory to OV and drug monotherapies. The ability of DMF to enhance viral spread results from its ability to inhibit type I interferon (IFN) production and response, which is associated with its blockade of nuclear translocation of the transcription factor nuclear factor κB (NF-κB). This study demonstrates that unconventional application of U.S. Food and Drug Administration-approved drugs and biol. agents can result in improved anticancer therapeutic outcomes.

To elucidate the function of oxidative phosphorylation (OxPhos) during B cell differentiation, we employ CD23Cre-driven expression of the dominant-neg. K320E mutant of the mitochondrial helicase Twinkle (DNT). DNT-expression depletes mitochondrial DNA during B cell maturation, reduces the abundance of respiratory chain protein subunits encoded by mitochondrial DNA, and, consequently, respiratory chain super-complexes in activated B cells. Whereas B cell development in DNT mice is normal, B cell proliferation, germinal centers, class switch to IgG, plasma cell maturation, and T cell-dependent as well as T cell-independent humoral immunity are diminished. DNT expression dampens OxPhos but increases glycolysis in lipopolysaccharide and B cell receptor-activated cells. Lipopolysaccharide-activated DNT-B cells exhibit altered metabolites of glycolysis, the pentose phosphate pathway, and the tricarboxylic acid cycle and a lower amount of phosphatidic acid. Consequently, mTORC1 activity and BLIMP1 induction are curtailed, whereas HIF1α is stabilized. Hence, mitochondrial DNA controls the metabolism of activated B cells via OxPhos to foster humoral immunity.

Biodiesel is a renewable biobased fuel obtained from transesterification of plant seed oil with methanol. This fuel has phys. properties that make it attractive as an alternative fuel for compression-ignition (diesel) engines. In the United States (US), there is a need to increase the production of sustainable and environmentally friendly biofuels, including biodiesel, to supplement fossil fuels. To meet these challenges, emphasis is being placed on expanding the use of low-cost non-food oils as feedstocks for biodiesel. Low-erucic acid pennycress (LEAP) was developed from field pennycress as a cover crop for use in the Upper Midwestern US. The objective of the present study was to convert LEAP oil to biodiesel (fatty acid Me esters [FAME]) and evaluate its fuel properties. The same process was applied to FAME made from natural field pennycress (FPC) oil as a baseline for comparison of results. The LEAP oil-FAME (LEAP-ME) had a kinematic viscosity at 40°C (KV40) that was lower than that of FPC oil-FAME (FPC-ME), and nearly equivalent to the KV40 of soybean oil-FAME (SME). The total saturated-FAME (SFAME) concentration of LEAP-ME (5.40 mass%) was greater than that of FPC-ME. Since SFAME have high m.ps., this caused LEAP-ME to have a higher cloud point (CP) than FPC-ME. However, the CP of LEAP-ME (-6.8°C) was lower than those of canola oil-FAME (CaME; -2.5°C) and SME (-2.8°C). The oxidative induction period at 110°C (IP110) of LEAP-ME was poor (0.83 h), indicating that it would need to be treated with antioxidants before distribution as an alternative diesel fuel.

During cooking, aromatic rice has a pleasant and characteristic aroma, a relevant factor to add sale value and attract consumer interest. This work studied the volatile compounds of aromatic rice (IAC 500) aiming at identifying those responsible for the aroma and flavor of the cooked rice. The description of the aromatic notes of the IAC 500 rice was carried out by a trained and selected sensory panel, followed by olfactometry (OSME) and identification by GC-MS of the rice volatile compounds extracted by SPME. A total of 80 volatiles was sensorially perceived and/or detected in the chromatog. effluent, of which 65 were identified, 44 presented some odor, and 36 were odorless. Among the odorous compounds, 15 were not detected by GC-FID or GC-MS. This study confirmed the compound 2-acetyl-1-pyrroline as the impacting volatile compound to the aroma of aromatic rice since it presented a very low percentage of area in the chromatogram and a high odor intensity. Other 43 compounds presented odor in lower intensities, but also contributed to the overall aroma of IAC 500 rice. From the 11 aromatic notes mentioned by the trained panel (cooked vegetable/seed, corn, hominy, green, porridge, popcorn, fresh baked cake/bread, milk, caramel, tapioca flour and flower), eight were related to the volatile compounds responsible for their aroma.

The incorporation of TiO2 nanoparticles into long (several micrometers) fibers of a gel-forming poly(para-phenylene vinylene) (PPV) derivative is described. For that purpose, a PPV-block-dopamine polymer is synthesized, which is used to coat TiO2 nanoparticles leading to well-dispersed nanoparticles and fluorescence quenching of the polymer. Independently, the homopolymer gels in various organic solvents are observed to form long fibers. By mixing the PPV homopolymer with the TiO2-PPV hybrid, we are able to incorporate the nanoparticles into the fibrous network, as shown in transmission electron microscopy (TEM) images. Photoinduced charge separation is measured by Kelvin probe force microscopy. Upon illuminating the polymer fibers, pos. charges are observed, even at distances of around 300 nm. From these results, charge transport along the polymer fibers is inferred.
