Glioblastoma multiforme (GBM) is a grade IV, lethal, and the most common type of brain tumor. GBM can acquire resistance to temozolomide (TMZ) recommended for its treatment. Naringenin (NAG), a flavonoid generally found in grapefruit, has antioxidant, anti-proliferative, and anti-inflammatory properties. It has been reported that phytochems. can reduce resistance and improve the efficacy of a chemo-resistant drug. The combinatorial effect of TMZ and NAG on cell proliferation was evaluated using 3-4,5-dimethylthiazol-2-yl-2,5-diphenyltetrazolium bromide (MTT) assay, and the apoptosis in the U87MG and LN229 GBM cells were evaluated by change in fluorescence intensity. The effect of NAG and TMZ on anchorage-independent single-cell colony formation and cell migration was investigated. NAG and TMZ demonstrated enhanced cytotoxic effects on U87MG and LN229 cell lines. The combination index value being less than one indicated the synergistic action of the two drugs in restricting the growth of the cells. The NAG and TMZ together resulted in higher fluorescence intensity as compared to the alone drug. Further, the study showed a marked reduction in the migration of the cells and the formation of a single cell colony.

Oxathiapiprolin is one of the best active fungicides discovered for oomycetes control. To develop a fungicide candidate with a broad spectrum of activity, 22 new piperidinylthiazole derivatives were designed and synthesized. Compound (I) showed the best activity against Pseudoperonospora cubensis (Berk. et Curt.) Rostov and Phytophthora infestans in vivo with 100% and 80% of inhibition, resp., at 1 mg/L, and 72.87% of field efficacy against P. cubensis at 1 g ai/667 m2 validated these results. Compound (II) exhibited a broad spectrum of excellent activity against Sclerotinia sclerotiorum with EC50 = 0.30 mg/L (>10 times more active than oxathiapiprolin and azoxystrobin in vitro), good activity against Botrytis cinerea, Cercospora arachidicola, and Gibberella zeae with EC50 of 14.54, 5.57, and 14.03 mg/L in vitro and against P. cubensis and P. infestans with 60% and 30% inhibition rates, resp., at 1 mg/L in vivo. Mode of action studies by RNA sequencing anal. discovered oxysterol-binding protein (OSBP), chitin synthase (CHS1), and (1,3)-尾-glucan synthase (FKS2) as the potent target of II against S. sclerotiorum. Quenching studies validated that OSBP was the same target of both II and oxathiapiprolin; it was quenched by both of them. We present the synthesis of reactive polymer brushes prepared by surface reversible addition-fragmentation chain transfer polymerization of pentafluorophenyl acrylate. The reactive ester moieties can be used to functionalize the polymer brush film with virtually any functionality by simple post-polymerization modification with amines. Dithiobenzoic acid benzyl-(4-ethyltrimethoxylsilyl) ester was used as the surface chain transfer agent (S-CTA) and the anchoring group onto the silicon substrates. Reactive polymer brushes with adjustable mol. weight, high grafting d., and conformal coverage through the grafting-from approach were obtained. Subsequently, the reactive polymer brushes were converted with amino-spiropyrans resulting in reversible light-responsive polymer brush films. The wetting behavior could be altered by irradiation with UV or visible light. Furthermore, a patterned surface of polymer brushes was obtained using a lithog. technique. UV irradiation of the S-CTA-modified substrates leads to a selective degradation of S-CTA in the exposed areas and gives patterned activated polymer brushes after a subsequent RAFT polymerization step. Conversion of the patterned polymer brushes with 5-((2-aminoethyl)amino)naphthalene-1-sulfonic acid resulted in patterned fluorescent polymer brush films. Inorganic perovskite quantum dots (QDs) have attracted great scientific attention in the field of luminescent materials, but the application has been limited by the inferior stability that results from highly dynamic capping ligands. In this work, we use a rare-earth complex to modify perovskite QDs with ligand exchange to realize perovskite functionalization; meanwhile, the stability of perovskite QDs is greatly improved. D. functional theory calculation results show that the adsorption energy of the europium complex to QDs is higher than that with traditional ligands, which provides a thermodn. basis for stability improvement. Furthermore, the modified QDs exhibit attractive dual-response property, including temperature and pH response ascribed to QDs and europium complexes, resp. The superior property can be applied to multi-stimuli-responsive optical encoding, which is further capable of enhancing the security of encrypted information. This study not only affords a strategy for the synthesis of highly stable perovskites but also provides a method for the functionalization of perovskites.

Retinal vein occlusion (RVO) is a severe retinal vascular disease involving several complications, leading to weakening of vision and even blindness. Globally, over 16 million patients with RVO were found in the middle-aged population. Paeoniflorin (PF), a monomer of Taohong Siwu decoction, was reported to exhibit many pharmacol. activities including anti-inflammatory, antioxidant, cardioprotective, and neuroprotective effects. However, the effect of PF on the progression of RVO remains unclear. In the current study, CCK8 assay was performed to investigate the cell viability. In addition, transwell assay and western blot were used to measure cell invasion and protein expression, resp. Moreover, a mouse model of oxygen-induced dischemic retinopathy (OIR) was established. We found PF was able to inhibit the migration and angiogenesis of human retinal capillary endothelial cells under normoxia. Addnl., PF notably prevented hypoxia-induced angiogenesis of human retinal capillary endothelial cells via inhibiting hypoxia-inducible factor-1伪 (HIF-1伪)/vascular endothelial growth factor (VEGF)/STAT3 pathway. Eventually, PF significantly alleviated the retinal lesions in the mouse with OIR. All in all, PF was able to alleviate the progression of retinal vein occlusion via inhibiting HIF-1伪/VEGF/STAT3 pathway. These findings might provide some theor. knowledge for exploring novel effective treatment for patients with RVO.

Pretreatment drug resistance (PDR) can limit the effectiveness of HIV antiretroviral therapy (ART). The aim of this study was to assess the prevalence of PDR among HIV-pos. individuals that initiated antiretroviral therapy in 2014-2020 in southwestern China. Consecutive cross-sectional surveys were conducted in Qinzhou, Guangxi. We obtained blood samples from individuals who were newly diagnosed with HIV in 2014-2020. PDR and genetic networks analyses were performed by HIV-1 pol sequences using the Stanford HIV-database algorithm and HIV-TRACE, resp. Univariate and multivariate logistic regression models were used to explore the potential factors associated with PDR. In total, 3236 eligible HIV-pos. individuals were included. The overall prevalence of PDR was 6.0% (194/3236). The PDR frequency to NNRTI (3.3%) was much higher than that of NRTI (1.7%, p < 0.001) and PI (1.2%, p < 0.001). A multivariate logistic regression anal. revealed that PDR was significantly higher among individuals aged 18-29 (adjusted odds ratio (aOR): 1.79, 95% CI 1.28-2.50) or 30-49 (aOR: 2.82, 95% CI 1.73-4.82), and harboring CRF08_BC (aOR: 3.23, 95% CI 1.58-6.59). A total of 1429 (43.8%) sequences were linked forming transmission clusters ranging in size from 2 to 119 individuals. Twenty-two individuals in 10 clusters had the same drug resistant mutations (DRMs), mostly to NNRTIs (50%, 5/10). The overall prevalence of PDR was medium, numerous cases of the same DRMs among genetically linked individuals in networks further illustrated the importance of surveillance studies for mitigating PDR. Herein, the active ester monomer pentafluorophenyl acrylate (PFPA) acts as a general hydrophobic masking group for N-alkyl acrylamides. Amidation reactions with different amines allow the transformation of one PFPA-based hydrophobic precursor network into a multitude of different APCNs that possess optical transparency and nanophase-separated morphologies. Accordingly, the properties of these ACPNs can be tailored to the desired application by simple variation of the amide functionality. Furthermore, combination of PFPA with another hydrophobically masked monomer allows for the functionalization of the hydrophilic phase of APCNs in precisely defined amounts Controlled integration of pendant groups such as pH-responsive imidazole, fluorescent dyes, and biotin for specific protein binding is achieved, greatly expanding the functionality of the APCNs. Such functionalized APCNs could find application as stimuli-responsive drug delivery membranes, smart hydrogels, biosensors, or as matrixes for biocatalysis.

We demonstrate the usefulness of our APTAC (amphiphilic polymer-facilitated transformations under anhydrous conditions) approach by performing several challenging transformations on DNA-conjugated small mols. in pure organic solvents: the addition of a carbanion equivalent to a DNA-conjugated ketone in THF, the synthesis of saturated heterocycles using the tin (Sn) amine protocol (SnAP) in dichloromethane, and the dual-catalytic (Ir/Ni) metallaphotoredox decarboxylative cross-coupling of carboxylic acids to DNA-conjugated aryl halides in DMSO. In addition, we demonstrate the feasibility of the latter in multititer-plate format.
