The production of valuable fatty acid alkyl ester (FAAE) com. known as biodiesel from used cooking oil (UCO) as feedstock has recently gained global interest. To carry out this production process, an esterification reaction is necessary to minimize the amount of free fatty acid (FFA) in the UCO. The purpose of this research work is to evaluate the performance of the newly snythesized sulfonated hypercrosslinked exchange resin (SHER) for the esterification reaction. The catalyst was first synthesized and characterized using various physicochem. analyses (i.e. FTIR, surface morphol., N2 physisorption anal. and TGA and further subjected to the esterification reaction to determine the reaction kinetics. The esterification reaction was conducted at various catalyst loading (1-8 wt%), reaction temperatures (40-60°C), and methanol to oil molar ratios (6:1-24:1). From the characterization study, SHER was found to have high decomposition temperature (up to 398°C) and sp. surface area (836 m2 g-1). Addnl., SHER had the capability to accommodate high number of active sites which could benefit the esterification reaction. The highest FFA conversion of 97% was achieved at 5 wt% catalyst loading, 60°C of reaction temperature and 12:1 methanol to oil mole ratio in 2 h reaction time. Analyses of the spent catalyst revealed the presence of impurities residue on the SHER’s surface following the esterification reaction, and the catalyst pore remained evident even after several reusability cycles.

Glioblastoma (GBM) is the most common and lethal primary malignant brain tumor, and due to its unique features, its management is certainly one of the most challenging ones among all cancers. N6-isopentenyladenosine (IPA) and its analog N6-benzyladenosine (N6-BA) are modified nucleosides endowed with potent antitumor activity on different types of human cancers, including GBM. Corroborating our previous finding, we demonstrated that IPA and N6-BA affect GBM cell line proliferation by modulating the expression of the F-box WD repeat domain-containing-7 (FBXW7), a tumor suppressor with a crucial role in the turnover of many proteins, such as SREBPs and Mcl1, involved in malignant progression and chemoresistance. Luciferase assay revealed that IPA-mediated upregulation of FBXW7 translates in transcriptional inactivation of its oncogenic substrates (Myc, NFkB, or HIF-1α). Moreover, downregulating MGMT expression, IPA strongly enhances the killing effect of temozolomide (TMZ), producing a favorable sensitizing effect starting from a concentration range much lower than TMZ EC50. Through DNA methyltransferase (DNMT) activity assay, anal. of the global DNA methylation, and the histone modification profiles, we demonstrated that the modified adenosines behave similar to 5-AZA-dC, known DNMT inhibitor. Overall, our results provide new perspectives for the first time, suggesting the modified adenosines as epigenetic tools able to improve chemo- and radiotherapy efficacy in glioblastoma and potentially other cancers.

Semi-liquid catholyte Lithium-Sulfur (Li-S) cells have shown to be a promising path to realize high energy d. energy storage devices. In general, Li-S cells rely on the conversion of elemental sulfur to soluble polysulfide species. In the case of catholyte cells, the active material is added through polysulfide species dissolved in the electrolyte. Herein, we use operando Raman spectroscopy to track the speciation and migration of polysulfides in the catholyte to shed light on the processes taking place. Combined with ex-situ surface and electrochem. anal. we show that the migration of polysulfides is central in order to maximize the performance in terms of capacity (active material utilization) as well as interphase stability on the Li-metal anode during cycling. More specifically we show that using a catholyte where the polysulfides have the dual roles of active material and conducting species, e. g. no traditional Li-salt (such as LiTFSI) is present, results in a higher mobility and faster migration of polysulfides. We also reveal how the formation of long chain polysulfides in the catholyte is delayed during charge as a result of rapid formation and migration of shorter chain species, beneficial for reaching higher capacities. However, the depletion of ionic species during the last stage of charge, due to the conversion to and precipitation of elemental sulfur on the cathode support, results in polarization of the cell before full conversion can be achieved.

Niobium incorporated meso-tetra-(4-carboxyphenyl)-porphyrin (Nb-TCPP) was prepared for the first time and grafted through the axial position by the surface amine groups present on functionalized SBA-15 (SBA-AM). The synthesized TCPP ligand, Nb-TCPP complex, and the grafted Nb-TCPP-SBA-AM complex were thoroughly characterized by various anal. and spectroscopic techniques such as FTIR, UV-visible, DR UV-visible, CHN, 1H NMR, powder XRD, and N2 sorption studies. The catalytic activity of the homogeneous (Nb-TCPP) and the heterogenized (Nb-TCPP-SBA-AM) complex were explored for the esterification of levulinic acid. The studies revealed that Nb-TCPP and Nb-TCPP-SBA-AM showed comparatively good catalytic activity (74-80% conversion) for the esterification of levulinic acid using methanol under mild reaction conditions with the formation of Me levulinate and α-angelica lactone as the major products.

Cancer stem cells, quiescent and drug resistant, have become a therapeutic target. Unlike high-intensity focused ultrasound directly killing tumor, low-intensity pulsed ultrasound (LIPUS), a new noninvasive phys. device, promotes pluripotent stem cell differentiation and is primarily applied in tissue engineering but rarely in oncotherapy. We explored the effect and mechanism of LIPUS on glioma stem cell (GSC) expulsion from quiescence. Here, we observed that LIPUS led to attenuated expression of GSC biomarkers, promoted GSC escape from G0 quiescence, and significantly weakened the Wnt and Hh pathways. Of note, LIPUS transferred sonomech. energy into cytochrome c and B5 proteins, which converted oxygen mols. into singlet oxygen, triggering telomere crisis. The in vivo and in vitro results confirmed that LIPUS enhanced the GSC sensitivity to temozolomide. These results demonstrated that LIPUS “”waked up”” GSCs to improve their sensitivity to chemotherapy, and importantly, we confirmed the direct targeted proteins of LIPUS in GSCs.

Secondary plant metabolites remain one of the key sources of therapeutic agents despite the development of new approaches for the discovery of medicinal drugs. In the current study, chem. anal., and biol. activities of Kei apple (Dovyalis caffra) methanolic extract were evaluated. Chem. anal. was performed using HPLC and GC-MS. Antiviral and anticancer effect were assessed using the crystal violet technique and activity against human liver cells (HepG2), resp. Antibacterial activity was tested with the disk diffusion method. The obtained results showed that chlorogenic acid (2107.96 ± 0.07 μg/g), catechin (168 ± 0.58 μg/g), and gallic acid (15.66 ± 0.02 μg/g) were the main bioactive compounds identified by HPLC techniques. While, compounds containing furan moieties, as well as levoglucosenone, isochiapin B, dotriacontane, 7-nonynoic acid and tert-hexadecanethiol, with different biol. activities were identified by GC-MS. Addnl., inhibition of 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) scavenging was 79.25% at 2000 μg/mL, indicating its antioxidant activity with IC50 of 728.20 ± 1.04 μg/mL. The tested extract exhibited potential anticancer activity (58.90% toxicity) against HepG2 cells at 1000 μg/mL. Potential bacterial inhibition was observed mainly against Escherichia coli and Proteus vulgaris, followed by Staphylococcus aureus and Bacillus subtilis with a diameter of growth inhibition ranging from 13 to 24 mm. While weak activities were recorded for fungi Candida albicans (10 mm). The extract showed mild antiviral activity against human coronavirus 229E with a selective index (SI) of 10.4, but not against human H3N2 (SI of 0.67). The mol. docking study′s energy ratings were in good promise with the experiment documents of antibacterial and antiviral activities. The findings suggest that D. caffra juice extract is a potential candidate for further experiments to assess its use as potential alternative therapeutic agent.

Ischemic heart disease (IHD) has always been the focus of attention of many researchers in cardiovascular disease, and its pathogenesis is also very complicated. Ferroptosis may be involved in the occurrence and development of IHD. First, primary cardiomyocytes were treated with H2O2 to simulate the IHD in vitro model. Luminescent discotic liquid crystals based on tris(N-salicylideneaniline)s [TSANs] bearing trans-stilbene fluorophores have been synthesized. Their mesomorphic and photophys. properties were studied. These TSANs existing in the form of C3h and Cs geometrical isomers of the keto-enamine tautomer self-assemble to form the columnar phase and exhibit fluorescence both in solution and mesophase states. These columnar mesophases also exhibit frozen (glassy) columnar phase, which ensures preserved fluorescence intensity, defect free alignment and simultaneous restriction on the motion of ionic impurities. These features make them promising candidates for the use in organic light emitting diodes, particularly as emissive layers. This study also led to an understanding about the dependence of their mesomorphism (mesophase type, stability and thermal range) and photophys. features on the number and pattern of their peripheral substitution, in comparison to the analogous columnar liquid crystalline TSANs reported earlier.
