The UV filter 4-methylbenzylidene camphor (4-MBC), used in cosmetics, the antioxidant butylated hydroxytoluene (BHT), used inter alia as a food additive and in cosmetics, and the plasticizer tris(2-ethylhexyl) trimellitate (TOTM), used mainly in medical devices as substitute for di-(2-ethylhexyl) phthalate (DEHP), are suspected to have endocrine disrupting effects. Human biomonitoring methods that allow for assessing the internal exposure of the general population to these substances were recently developed in a German cooperation to enhance the use of human biomonitoring. First-morning void urine samples from 3- to 17-yr-old children and adolescents living in Germany were analyzed for metabolites of 4-MBC (N = 447), BHT (N = 2091), and TOTM (N = 431) in the population-representative German Environmental Survey on Children and Adolescents 2014-2017 (GerES V). 4-MBC metabolites were found in quantifiable amounts only in single cases and exposure levels remained well below health-based guidance values. In contrast, ubiquitous exposure to BHT became evident with a geometric mean (GM) urinary concentration of the metabolite BHT acid of 2.346μg/L (1.989μg/gcreatinine) and a maximum concentration of 248μg/L (269μg/gcrea). The highest GM concentration was found in young children aged 3-5 years, yet no specific sources of exposure could be identified. Also, TOTM metabolites were found in quantifiable amounts only in very few samples. None of these findings could be related to previous hospital treatment or exposure via house dust. The presented results will be the basis to derive reference values for exposure of children and adolescents in Germany to BHT and will facilitate to identify changing exposure levels in the general population. There is enormous interest in myo-inositol derivatives as they serve as precursors for the synthesis of several biol. important phosphoinositols, natural products, catalyst, supramol. architectures etc. However the presence of six secondary hydroxy groups of similar reactivity warrants protection of inositol hydroxyl groups for effective synthesis. Acid catalyzed protection of inositol hydroxyl groups as ortho esters or ketals are the most commonly used protecting strategy in inositol chem. Traditionally, homogeneous acid catalysts such as para-toluenesulfonic acid (p-TSA) or camphorsulfonic acid (CSA) are used for these transformations. While the reversible nature of these reactions necessitates the catalyst removal, aqueous work up cannot be employed for their removal as the products are water soluble The authors have circumvented this problem by using H2SO4-silica as the solid supported catalyst, which can be removed by filtration, for these transformations. Treatment of myoinositol with trialkyl ortho esters in presence of H2SO4-silica under normal conditions resulted in esterification at the least reactive hydroxyl group (C2-OH) giving exclusively the corresponding 2-O-acyl-myo-inositol. By doing the reaction in a rotary evaporator under reduced pressure resulted in the formation of the corresponding ortho esters, wherein three hydroxyl groups of inositol are protected simultaneously. The authors could synthesize different myo-inositol ortho esters 6-10 in excellent yields by this method. Ketalization of myo-inositol with one equivalent of 1,1-dimethoxycyclohexane or 2,2-dimethoxypropane in presence of H2SO4-silica under similar conditions resulted in the simultaneous protection of 1-OH and 2-OH giving the 1,2-O-cyclohexylidene-myo-inositol or 1,2-O-isopropylidene-myo-inositol in excellent yields. Also, diketalization of myo-inositol with 2,2-dimethoxypropane gave three diketals namely (±)-1,2:4,5-di-O-isopropylidene-myo-inositol (±)-1,2:5,6-di-O-isopropylidene-myo-inositol and (±)-1,2:3,4-di-O-isopropylidene-myoinositol in considerable yields. Similarly when dimethoxycyclohexane was used, the dicyclohexylidene derivatives (±)-1,2:4,5-di-O-cyclohexylidene-myo-inositol (±)-1,2:5,6-di-O-cyclohexylidene-myo-inositol and (±)-1,2:3,4-di-O-cyclohexylidene-myo-inositol were obtained in good yield. While the yields of 1,2:3,4-di-O-alkylidene-myo-inositols are negligibly small by other known methods, interestingly, our method give these diketals in good yields and hence can be exploited synthetically. The presented study investigated the potential effects of phospholipid-lining as anti-coagulation coating (ACC) on the migration rate of plasticizers from PVC tubing into blood. For the in-vitro study, five different groups of tubing sets in six replicates were perfused with sheep blood (Group A: PVC plasticized with di-(2-ethylhexyl) phthalate (DEHP) without ACC, Group B: DEHP-plasticized PVC with ACC, Group C: PVC plasticized with tri-(2-ethylhexyl) trimellitate (TOTM) without ACC, Group D: TOTM-plasticized PVC with ACC, Group E (control group): polyolefin material with ACC but without plasticizers). Both the levels of the unchanged plasticizers in blood and the concentration levels of their primary degradation products were assessed. For DEHP, the primary metabolite MEHP (mono-(2-ethylhexyl) phthalate) was determined The isomers of MEHTM (mono-(2-ethylhexyl) trimellitate) and DEHTM (di-(2-ethylhexyl) trimellitate), resp., were investigated as primary metabolites of TOTM. The results indicate that phospholipid coating (ACC) rather enhances the migration of plasticizers and of their primary degradation products from PVC tubing into streaming blood. The enhancement effect was found to be slightly greater for TOTM, but as TOTM migrates in significantly lower levels than DEHP in all exptl. settings, TOTM is confirmed to be a recommendable alternative plasticizer to DEHP in medical devices. In preliminary trials using silica gel thin-layer chromatog., the retention behavior of 21 common aromatic compounds was tested in eluents with different solvent strength including EtOAc, MeOH, cyclohexane, and dichloromethane. Based on these results, the SPE sample cleanup with silica gel and cyclohexane as an eluent was suggested as best suitable for the purpose. The SPE cleanup was successfully achieved for all tested potentially interfering aromatic cosmetic ingredients except for butylated hydroxytoluene. The recovery for lipophilic cosmetics is more than 80% based on naphthalene as calculation equivalent The 1H qNMR method is a good complement to the LC-GC-FID method, which is predominantly used for the determination of MOSH and MOAH.
