In the in vitro cell-based assays, 68, 38, 20, 17 and 17 compounds were identified as positives for PXR, PPARα, LXRα, FXR and RXRα, resp. The association analyses demonstrated that the PXR-pos. compounds showed high frequency of endpoints related to liver hypertrophy, such as centrilobular hepatocellular hypertrophy, suggesting that PXR activation is involved in chem.-induced liver hypertrophy in rats. It is intriguing to note that the PXR-pos. compounds also showed statistically significant associations with both prolonged activated partial thromboplastin time and prolonged prothrombin time, suggesting a possible involvement of PXR in the regulation of blood clotting factors. Furthermore, the identification of unknown mols. requiring the implementation of addnl. confirmatory tools in the absence of reference standards, such as tandem mass spectrometry spectra comparisons and in silico prediction of mass spectra, is a major bottleneck. Methods : We applied a combination of four chromatog./ionization techniques (reversed-phase (RP) – heated electrospray ionization (HESI) in both pos. (+) and neg. (-) modes, RP – atm. pressure chem. ionization (APCI) in pos. mode, and hydrophilic interaction liquid chromatog. (HILIC) – HESI pos.) using a Thermo Q Exactive® liquid chromatog./high-resolution accurate mass spectrometry (LC/HRAM-MS) platform for the anal. of 3R4F-derived smoke. Compound identification was performed by using mass spectral libraries and in silico predicted fragments from multiple integrated databases. Results : A total of 331 compounds with semi-quant. estimates ≥100 ng per cigarette were identified, which were distributed within the known chem. space of tobacco smoke. The integration of multiple LC/HRAM-MS-based chromatog./ionization approaches combined with complementary compound identification strategies was key for maximizing the number of amenable compounds and for strengthening the level of identification confidence. A total of 50 novel compounds were identified as being present in tobacco smoke. In the absence of reference MS2 spectra, in silico MS2 spectra prediction gave a good indication for compound class and was used as an addnl. confirmatory tool for our integrated non-targeted screening (NTS) approach. The fruit fly Drosophila melanogaster is a powerful model system for the study of innate immunity in vector insects as well as mammals. For vector insects, it is particularly important to understand all aspects of their antiviral immune defenses, which could eventually be harnessed to control the transmission of human pathogenic viruses. The immune responses controlling RNA viruses in insects have been extensively studied, but the response to DNA virus infections is poorly characterized. Here, we report that infection of Drosophila with the DNA virus Invertebrate iridescent Virus 6 (IIV-6) triggers JAK-STAT signaling and the robust expression of the Turandots, a gene family encoding small secreted proteins. To drive JAK-STAT signaling, IIV-6 infection more immediately induced expression of the unpaireds, a family of IL-6-related cytokine genes, via a pathway that required one of the three Drosophila p38 homologs, p38b. In fact, both Stat92E and p38b were required for the survival of IIV-6 infected flies. In addition, in vitro induction of the unpaireds required an NADPH-oxidase, and in vivo studies demonstrated Nox was required for induction of TotA. These results argue that ROS production, triggered by IIV-6 infection, leads to p38b activation and unpaired expression, and subsequent JAK-STAT signaling, which ultimately protects the fly from IIV-6 infection. Deep eutectic solvent based on Ph salicylate (salol) and DL-menthol (1:1) was used as the extraction solvent. The control of temperature enhanced the dispersion of deep eutectic solvent and a homogeneous solution was made. Upon cooling the solution in an ice bath, a cloudy solution was formed. Afterwards, the mixture was centrifuged and the sediment phase was analyzed by Electrothermal Atomic Absorption Spectroscopy (ETAAS). To improve the extraction efficiency, the main parameters were optimized. Under the optimized extraction conditions, the limit of detection (LOD) and limit of quantification (LOQ) were 0.03μg L-1 and 0.10μg L-1 resp. Preconcentration factor was obtained 191. The precision, expressed as the relative standard deviation (RSD, n = 10), found to be 4.6%. Finally, the method was an effective procedure for the determination of palladium in water, catalytic converter and road dust samples with acceptable extraction recoveries of 95.1% to 100.4%. The obtained results confirmed the ability of the developed method in the different samples. After centrifugation of the cloudy solution at 20 °C, the settled solidified extractant was isolated from the aqueous solution simply by decanting the tube. Predominant exptl. variables affecting the extraction efficiency of explosives were evaluated. This method was coupled with high performance liquid chromatog.-ultra violet detection for the anal. of nitroarom. compounds Under the optimized conditions, the limits of detection achieved were in the range of 0.03-0.05 μg L-1. The enrichment factors were between 576 and 702 and the relative standard deviations were <10%.
