Epoxy-functional sterically-stabilized diblock copolymer spherical nanoparticles were synthesized via polymerization-induced self-assembly (PISA) in mineral oil. Epoxy groups were located either (i) in the nanoparticle cores or (ii) within the steric stabilizer chains. For the first system, RAFT dispersion polymerization of glycidyl methacrylate (GlyMA) was conducted using a poly(lauryl methacrylate) (PLMA63) precursor. The second system involved statistical copolymerization of GlyMA with lauryl methacrylate to produce a P(LMA50-stat-GlyMA9) precursor followed by chain extension using Me methacrylate (MMA). 1H NMR studies and THF GPC anal. indicated that high monomer conversions (â‰?95%) and narrow mol. weight distributions (Mw/Mn â‰?1.17) were obtained for both formulations. Dynamic light scattering indicated hydrodynamic diameters of 26 nm and 28 nm for P(LMA50-stat-GlyMA9)-PMMA67 and PLMA63-PGlyMA89 spheres, resp. Transmission electron microscopy studies confirmed a well-defined spherical morphol. in each case. Post-polymerization modification of these spherical nanoparticles was examined by reacting the epoxy groups with benzylamine. For the PLMA63-PGlyMA89 spheres, an [amine]/[epoxy] molar ratio of unity was sufficient to react all the epoxy groups. In contrast, the P(LMA50-stat-GlyMA9)-PMMA67 spheres required a fifty-fold excess of benzylamine for complete reaction. Furthermore, epoxy ring-opening reactions were conducted using either a trace amount of water or 50% volume/volume aqueous acetic acid at 110°C.

Flavor is a key attribute defining melon fruit quality and driving consumer preferences. We characterized and compared fruit ripening patterns (ethylene, respiration), physicochem. properties (rind/flesh color, firmness, soluble solids, acidity), aroma volatiles, and flavor-related sensory attributes in seven melon genotypes differing in shelf life capacity. Fruits were evaluated at optimal maturity and after storage for six days at 5°C plus one day at room temperature Total volatile content increased after storage in all genotypes, with esters being dominant. Shorter shelf-life genotypes, displaying a sharper climacteric phase, correlated with fruity/floral/sweet flavor-related descriptors, and with esters, sulfur-containing compounds and a terpenoid. Longer shelf-life types were associated with firmness, green and grassy aroma/flavor and aldehydes. Multivariate regression identified key volatiles that predict flavor sensory perception, which could accelerate breeding of longer shelf-life melons with improved flavor characteristics.

Volatile compounds determine the aroma of fruits, giving their unique flavor characteristics. The aim of many plant breeding projects is to improve the consumers’ flavor experience when eating fresh produce. Large scale breeding trials produce thousands of samples which need volatile profiling amongst other phenotypes. Despite this interest, current methods have limitations: sampling unsuitable for field conditions, high cost and the inherent issue of highly variable data, which can hinder interpretation. We introduced a simple and robust sampling methodol. based on silicone rod extraction, thermal desorption gas chromatog. – mass spectrometry (GC-MS) to address these issues. We used differentiated calibration standards to generate quant. data for metabolites of varying abundance. The method was used to profile 327 melons with high sensitivity (0.05-10 ng/mL, compound dependent), good reproducibility (7%) and differentiate melon varieties based on their volatile profile. The data were then used for line selection for a desired flavor profile.

Fresh-cut fruits are popular due to the convenience provided. However, fresh-cut processes damage fruit tissues and reduce the shelf life of products. Pulsed light (PL) treatment is a decontamination method of foods. PL treatment given repetitively at a certain interval during storage could further extend the shelf life of fresh-cut fruits. Edible coating preserves fresh-cut fruits by providing mech. strength and reducing respiration and water loss. This study was to evaluate the effects of alginate coating combined with repetitive pulsed light (RPL) on sensory quality and flavor of fresh-cut cantaloupes during storage. Cantaloupes were treated with alginate (1.86%, w/v) and RPL (0.9 J/cm2 at every 48 h up to 26 days) alone or in combination. Flavor anal. of fresh-cut cantaloupes was carried out every 12 days during storage at 4 ± 1 °C while sensory anal. was performed on day 32. Alginate coating and/or RPL retained sugar contents (17.92-20.01 g/kg FW for fructose, 18.77-19.98 g/kg FW for glucose and 23.02-29.41 g/kg FW for sucrose) in fresh-cut cantaloupes during storage. Combination of alginate with RPL reduced accumulation of lactic acid although alginate coating was more effective to minimise changes of other organic acids in fresh-cut cantaloupes. The combined treatment was also more effective than individual treatment in retaining total aroma compound concentration of fresh-cut cantaloupes during storage with the highest relative concentration, i.e. 3.174 on day 36. Overall, the combined alginate coating and RPL was effective to maintain the fresh-like sensory quality of fresh-cut cantaloupes with insignificant overall acceptability compared to the control.

Two rootstock crosses of sweet melon and watermelon (Cucurbita moschata x Cucurbita maxima hybrids), ‘Kickstart’ and ‘Carnivor’, grafted onto four melon cultivars (cantaloups; ‘Majestic’, ‘Hunter’, ‘Honeygoal’ and ‘Honeyval’) were evaluated for quality, phytonutrient composition and volatile compounds in the 2020 and 2021 growing season at two harvests. Grafted fruits showed higher SSC/TA ratio, and antioxidant scavenging activity. Grafted ‘Majestic/Carnivor’ and ‘Honeyval/ Carnivor’ melons significantly increased the ascorbic acid content than the controls. ‘Majestic/Carnivor’ melons showed the highest β- carotene and lutein than the controls. PLS-DA score plots discriminated the grafted cantaloupes and honeydew melons from controls based on the concentration of volatile compounds Hexyl acetate and hexadecane resp. discriminated the grafted cantaloups and honeydew melons from ungrafted fruits. ‘Carnivor’ rootstocks showed higher expression of volatiles in the grafted cantaloupes and honeydew melons in the heat map. Panellists preferred Majestic/Carnivor’ and ‘Honeyval/ Carnivor’ melons.

Improvement of the efficacy of radiotherapy for lung cancer and glioblastoma is urgently needed. We synthesized several novel DNA methyltransferase inhibitors and evaluated their potentials as possible radiosensitizers. Eleven non-nucleoside compounds were synthesized and evaluated along with one known compound using human lung cancer (A549) and glioblastoma (U373MG) cells. Cytotoxicity and radiosensitizing effects were evaluated using clonogenic assay. Sensitizer enhancement ratios at a survival fraction of 0.5 were calculated, and statistical anal. was performed using the ratio paired t-test. The inhibitory effects of three selected compounds on the activity of DNA methyltransferase 1 (DNMT1) and the pharmacokinetic profiles were analyzed. All twelve compounds demonstrated various levels of cytotoxicity. Of the twelve compounds, eleven and eight compounds radiosensitized A549 and U373MG cells, resp., with at least marginal significance (p<0.10). The sensitizer enhancement ratios in A549 and U373MG ranged 1.166-2.537 and 1.083-1.743 among compounds with radiosensitizing effects, resp. The three selected compounds inhibited DNMT1 activity by 26.5-78.5%. Elimination half-lives ranged from 0.3 to 1.3 h. Novel DNA methyltransferase inhibitors with significant radiosensitizing capacity and improved biostability were synthesized. These materials will serve as a basis for the development of novel radiosensitizers. Essential oils of Sambucus nigra were obtained by hydrodistillation and analyzed using the GC-MS technique, which led to the identification and quantification of 66 components from the leaves and 74 components from the flowers, accounting for 94.0 and 96.4% of the total components, resp. The main class of compounds present in the leaves were fatty acids and their derivatives: Me linoleate, palmitic acid, hydrocarbon tritetracontane, and an aromatic compound benzoic aldehyde. In the flowers there were hydrocarbons: tritetracontane and n-hexatriacontane, fatty acids and their derivatives: palmitic acid and linoleic acid, and alcs.: 2-methyl-3,15-octadecadienol and 2-hexyl-1-octanol. Among the dominant components of the essential oil of S. nigra flowers, there were also monoterpenes such as rose oxide, epoxylinalol, while the essential oil of the leaves contained a few compounds of monoterpenes in small amounts such as β-pinene and α-pinene. Antioxidant activity of the obtained essential oils was determined by the FRAP and DPPH assay techniques.

Studies of the composition of vegetable oils are expedient in order to achieve quality product modeling. The goal of this research was to determine the composition of fatty acids using gas chromatog. (GC) and antioxidant activity using DPPH and ABTS spectrophotometric methods. Analyzed oils were castor seed, sunflower seed, sesame seed, fibrous hemp seed, black radish seed and olive oils. Different classes of fatty acids were determined: polyunsaturated, monounsaturated and saturated; omega-3, omega-6 and omega-9 acids and individual constituents. The study results showed that the dominant acids in all oils were oleic and linoleic fatty acids. The best ratio of omega-6 to omega-3 acids was found in fibrous hemp seed oil -3:1. All analyzed vegetable oils demonstrated a high antioxidant activity.

Pomegranate seed oil is valuable source of bioactive compounds with health-beneficial effects, but it is sensitive to oxidation due to high content of PUFA. In order to improve stability of pomegranate seed oil, the influence of natural antioxidant (0.1% pomegranate peel extract) and a combination of synthetic antioxidant (0.01% butylhydroxytoluene) and pomegranate peel extract (0.05%) was investigated. The oxidation process was followed by monitoring fatty acid composition and measuring the quality and stability parameters of oil (acid value, peroxide value, anisidine value and thiobarbituric acid reactive substances value) during storage for 12 days at 65 0C. Furthermore, total phenolic content and DPPH radical scavenging activity was measured. Pomegranate peel extract as well as a combination of pomegranate peel extract and BHT can have a significant pos. impact (P < 0.05) on improvement of the quality and stability parameters of pomegranate seed oil. Furthermore, such activity was better than BHT used alone. Inflammation and oxidative stress are involved in the pathogenesis and progression of retinopathies, such as diabetic retinopathy (DR) and retinopathy of prematurity (ROP). Since Muller glial cells (MGCs) play a key role in the homeostasis of neural retinal tissue, they are considered an ideal model to study these cellular protective mechanisms. In this sense, quantifying ROS levels with a reproducible and simple method is essential to assess the contribution of pathways or mols. that participate in the antioxidant cell defense mechanism. In this article, we provide a complete description of the procedures required for the measurement of ROS with DCFH-DA probe and flow cytometry in MGCs. Key steps for flow cytometry data processing with the software are provided here, so the readers will be able to measure ROS levels (geometric means of FITC) and analyze fluorescence histograms. These tools are highly helpful to evaluate not only the increase in ROS after a cellular insult but also to study the antioxidant effect of certain mols. that can provide a protective effect on the cells.
