Comparison of five membrane filters to collect bioaerosols for airborne microbiome analysis

Aims Recovering DNA of airborne micro-organisms (AM) from air is a challenging task. We compared five membrane filters for bioaerosol sampling-mixed cellulose ester (MCE), polyethersulfone (PES), polyamide (PA), polytetrafluorethylene (PTFE) and polyvinylidene fluoride (PVDF)-based on their bacterial, fungal and eukaryotic DNA recoveries. Methods and results Bacterial, fungal and eukaryotic populations were quantified using quantitative PCR. With a bacterial consortium, PTFE exhibited the best recovery efficiency (113%), followed by PA (92%), PES (86%), MCE (48%) and PVDF (1%). When filters were compared with air, PA was used as a control to normalize results from the others. The bacterial, fungal and eukaryotic DNA recovery ratios were markedly greater in PES (9 center dot 3, 11 center dot 5 and 10 center dot 3 respectively) than in the remaining. Eukaryotic MiSeq sequencing revealed that PES recovered a more diverse and considerably richer assemblage (richness ratios, 4 center dot 97 vs <= 1 center dot 16 for PES vs the others). Rank abundance distribution analysis showed that distribution tails were longer (>4 times) in PES, but these did not differ between the remaining and PA. Community comparison showed that PES exhibited a lower variation across trials than the PA, while the remaining did not. Conclusions PES filter markedly outperformed the other filters in quantitative and qualitative recovery of AM. Significance and Impact of the Study Our findings demonstrated the importance of filter selection for sampling AM.

Infectious Effect of Organic Small Molecules on Photo-oxidative Aging of Polypropylene

Aging of a polymer is generally affected by adjacent polymers, known as infectious behavior of aging. Organic small molecules generated during aging are considered to be infection agents. However, the way they influence aging process and the difference among various infection agents are still unknown. In this paper, the influence of 18 representative organic small molecules on photo-oxidative aging of polypropylene (PP) was studied. The small molecules included acids, esters, aldehydes, ketones and alcohols, corresponding to typical photo-oxidative degradation products of polymers. When they were placed near PP during aging, the carbonyl index (CI) of PP was higher compared to the case when PP was aged individually. It was demonstrated that photo-oxidative aging of PP was accelerated by all the small molecules. The acceleration effects of acids, aldehydes and ketones were outstanding. Especially for propanoic acid, isobutyric acid, methacrylic acid, formaldehyde, acetaldehyde and butanone, CI was increased by more than 700% when PP was aged with them. Further, infection mechanism of acetone and acetic acid was investigated , since they had been identified as typical infection agents. CI and concentrations of gaseous degradation products of PP with and without acetone or acetic acid were measured with aging time. When PP was aged with acetone, CI was increased from as early as 24 h at an extremely high rate. With acetic acid, CI was increased after an obvious induction period and the increasing rate was lower than that with acetone. Similar phenomenon was observed in gaseous degradation products. The results showed that acetone acted as an initiator, photolyzed to generate CH3 center dot that could initiate the oxidation of PP; while acetic acid acted as a catalyzer, facilitating the decomposition of hydroperoxides and the accumulation of oxidation products.

In vitro kinetic release study, antimicrobial activity and in vivo toxicity profile of a kojic acid ester-based nanoemulsion for topical application

Nanoemulsions have emerged as novel vehicles for drug delivery that allow sustained or controlled release for topical application. In this study, kojic acid ester-based nanoemulsion (KAE-NA) was analyzed for in vitro permeation evaluation, kinetic release study, in vitro antimicrobial activity and in vivo toxicity profile on embryonic zebrafish (Danio rerio). Based on KAE-NA in vitro permeation evaluation, the percentage of permeation was significantly improved from 4.94% at 1 h to 59.64% at 8 h of application. The permeation rate of KAE-NA at 8 h was 4659.50 mu g cm(-2) h(-1) (initial concentration, C-0 = 2000 mu g mL(-1)) with a permeability coefficient (K-p) value of 0.48 cm h(-1). The kinetic release analysis showed the Korsmeyer-Peppas model was the best fitted kinetic model with high linearity [R-2 = 0.9964]. Antimicrobial activity of KAE-NA was studied against the skin pathogen bacteria Staphylococcus aureus ATCC 43300. The results indicated that the inhibition zone size of the KAE-NA (8.00 +/- 0.0 mm) was slightly bigger than that of its active ingredient, kojic acid ester (6.5 +/- 0.0 mm). The toxicity profile of KAE-NA on embryonic zebrafish revealed less toxicity with LC50 (50% lethal concentration) more than 500 mu g mL(-1). The survival rate of the embryonic zebrafish was more than 80% when treated at doses ranging from 7.81-250 mu g mL(-1) and showed normal development throughout the experiment without any observed deformation. Hence, KAE-NA proved to be less toxic on the embryonic zebrafish.

Partially acetylated cellulose nanofibrils were obtained from the major residual stream associated with tequila production. Background Insulin resistance could be associated with the development of Alzheimer disease (AD). The neuropathological hallmarks of AD are beta amyloid (A beta) produced from sequential cleavage initiated by beta-secretase and degraded by insulin degradation enzyme (IDE), as well as hyperphosphorylation of tau (p-tau). Insulin action involves the cascades of insulin receptor substrates (IRS) and phosphatidylinositol 3-kinase (PI3K), while phosphorylation of IRS-1 at ser307 (p-(IRS)-I-ser307-1) hinders the response. Our previous report suggested dipeptidyl peptidase-4 (DPP-4) is crucial to insulin resistance, and the subfractions of Abelmoschus esculentus (AE), F1 and F2, attenuate the signaling. Here we aim to investigate whether AE works to reduce A beta generation via regulating DPP4 and insulin resistance. Methods The subfractions F1 and F2 were prepared according to a succession of procedures. F1 was composed by quercetin glycosides and triterpene ester, and F2 contained a large amount of polysaccharides. The in vitro insulin resistance model was established by SK-N-MC cell line treated with palmitate. MTT was used to define the dose range, and thereby Western blot, ELISA, and the activity assay were used to detect the putative markers. One-way ANOVA was performed for the statistical analysis. Results Treatment of palmitate induced the level of p-(IRS)-I-ser307-1. Both F1 and F2 effectively decrease p-(IRS)-I-ser307-1, and recover the expression of p-PI3K. However, the expression of total IRS plunged with 25 mu g/mL of F1, while descended steadily with 5 mu g/mL of F2. As palmitate increased the levels of A beta 40 and A beta 42, both AE subfractions were effective to reduce A beta generation of and beta-secretase activity, but IDE was not altered in any treatment conditions. The expression of DPP4 was also accompanied with insulin resistance signals. Inhibition of DPP4 attenuated the activity of beta-secretase and production of A beta. Moreover, the present data revealed that both AE subfractions significantly decrease the level of p-Tau. Conclusions In conclusion, we demonstrated that AE would be a potential adjuvant to prevent insulin resistance and the associated pathogenesis of AD, and F2 seems more feasible to be developed.

Response of fatty acids and lipid metabolism enzymes during accumulation, depuration and esterification of diarrhetic shellfish toxins in mussels (Mytilus galloprovincialis)

Bivalve mollusks accumulate diarrhetic shellfish toxins (DSTs) from toxigenic microalgae, thus posing a threat to human health by acting as a vector of toxins to consumers. In bivalves, free forms of DSTs can be esterified with fatty acids at the C-7 site to form acyl esters (DTX3), presumably a detoxification mechanism for bivalves. However, the effects of esterification of DSTs on fatty acid metabolism in mollusks remain poorly understood. In this study, mussels (Mytilus galloprovincialis) were fed the DST-producing dinoflagellate Prorocentrum lima for 10 days followed by an additional 10-days depuration in filtered seawater to track the variation in quantity and composition of DST acyl esters and fatty acids. A variety of esters of okadaic acid (OA) and dinophysistoxin-1 (DTX1) were mainly formed in the digestive gland (DG), although trace amounts of esters also appeared in muscle tissue. A large relative amount of OA (60%-84%) and DTX1 (80%-92%) was esterified to DTX3 in the visceral mass (referred to as digestive gland, DG), and the major ester acyl chains were C16:0, C16:1, C18:0, C18:1, C20:1 and C20:2. The DG and muscle tissues showed pronounced differences in fatty acid content and composition during both feeding and depuration periods. In the DG, fatty acid content gradually decreased in parallel with increasing accumulation and esterification of DSTs. The decline in fatty acids was accelerated during depuration without food. This reduction in the content of important polyunsaturated fatty acids, especially docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA), would lead to a reduction in the nutritional value of mussels. Enzymes involved in lipid metabolism, including acetyl-coenzyme A carboxylase (ACC), fatty acid synthase (FAS), lipoprotein lipase (LPL) and hepatic lipase (HL), were actively involved in the metabolism of fatty acids in the DG, whereas their activities were weak in muscle tissue during the feeding period. This study helps to improve the understanding of interactions between the esterification of DSTs and fatty acid dynamics in bivalve mollusks.
