Nanosized structural defects in metal-organic frameworks (MOFs) attract growing attention and often remarkably enhance functional properties of these materials for various applications. In this work, a series of MOFs [Cu-2(TPTA)(1-x)(BDPBTR)(x)] (H(4)TPTA, [1,1′:3′,1 ”-terphenyl]-3,3 ”,5,5 ”-tetracarboxylic acid; H4BDPBTR, 1,3-bis(3,5,dicarboxyphenyl)-1,2,4-benzotriazin-4-yl radical)) with a new stable radical linker doped into the structure has been synthesized and investigated using Electron Paramagnetic Resonance (EPR). Mixed linkers H(4)TPTA and H4BDPBTR were used to bridge copper(II) paddle-wheel units into a porous framework, where H4BDPBTR is the close structural analogue of H(4)TPTA. MOFs with various x = 0-0.4 were investigated. EPR studies indicated that the radical linker binds to the copper(II) units differently compared to diamagnetic linker, resulting in the formation of nanosized structural defects. Moreover, remarkable kinetic phenomena were observed upon cooling of this MOF, where slow structural rearrangements and concomitant changes of magnetic interactions were induced. Thus, our findings demonstrate that doping of structurally mimicking radical linkers into MOFs represents an efficient approach for designing target nanosized defects and introducing new magnetostructural functionalities for a variety of applications.

Guillermo Calleja and co-workers from @urjc describe URJC-1-MOF as a new heterogeneous recyclable catalyst for c-heteroatom coupling reactions The capacity of copper-based URJC-1-MOF as a MOF catalyst in cross-coupling reactions has been evaluated, focusing on the Chan-Lam-Evans arylation-type reactions on amines and alcohols without extra additives or ligands. The extraordinary chemical and structural stability of URJC-1-MOF and its good specific surface, make this material a promising alternative to homogeneous Cu (II) catalysts for cross-coupling reactions. URJC-1-MOF showed a remarkable catalytic activity for cross-coupling C-N and C-O reactions, higher than other heterogeneous and homogeneous copper-based catalyst, such as CuO, HKUST-1, Cu-MOF-74, Cu(OAc)(2) and CuSO4.5H(2)O. Moreover, its easy recovery by simple filtration and reusability in successive runs without any loss of activity and stability, demonstrates the potential of URJC-1-MOF as an alternative catalyst for this kind of reactions in different chemical media of industrial interest.

A key event in the directed evolution of enzymes is the systematic use of mutagenesis and selection, a process that can give rise to mutant libraries containing millions of protein variants. To this day, the functional analysis and identification of active variants among such high numbers of mutational possibilities is not a trivial task. Here, we describe a combinatorial semi-rational approach to partly overcome this challenge and help design smaller and smarter mutant libraries. By adapting a liquid medium transesterification assay in organic solvent conditions with a combination of virtual docking, iterative saturation mutagenesis, and residue interaction network (RIN) analysis, we engineered lipase B from P. antarctica (CalB) to improve enzyme recognition and activity against the bulky aromatic substrates and flavoring agents methyl cinnamate and methyl salicylate. Substrate-imprinted docking was used to target active-site positions involved in enzyme-substrate and enzyme-product complexes, in addition to identifying ‘hot spots’ most likely to yield active variants. This iterative semi-rational design strategy allowed selection of CalB variants exhibiting increased activity in just two rounds of site-saturation mutagenesis. Beneficial replacements were observed by screening only 0.308% of the theoretical library size, illustrating how semi-rational approaches with targeted diversity can quickly facilitate the discovery of improved activity variants relevant to a number of biotechnological applications.

The genus Ocimum displays excellent variability among the species and sub-species due to the highly crosspollinated nature and interspecific hybridization. The present investigation was carried out to identify the commercially promising growth stage(s) in different Ocimum for exatraction of oil and bioactive compounds at heighest. A set of three cultivated species were evaluated to study the variation in morpho-physiological traits and essential oil composition harvested at three stages namely vegetative, flowering and seed setting stage. The experiment was conducted for three consecutive years in factorial randomized complete block design with three replicates. The analysis of variance exhibited that the individual effect of Ocimum species and growth stage along with their interaction effect was significant in all the morpho-physiological parameters and bioactive constituent of oil in a pooled analysis. Highest plant height (103.95 cm) was observed in O. gratissimum L. harvested at seed setting stage while harvesting at flowering stage exhibited maximum leaf area (26.80 cm(2)), oil (0.52 %) and eugenol content (85.82 %). Sweet basil harvested at seed setting stage disclosed maximum branches per plant (21.53) and leaves per plant (4949) although linalool content (47.99 %) was found highest in harvested at flowering stage. O. basilicum L. was detected a great source of methyl chavicol (7.48 %) though this species was weaker source for methyl cinnamate. Among the studied species, O. gratissimum showed a negligible amount of methyl eugenol content in oil. A positive correlation between the morpho-physiological traits, oil content and its constituents with the various growth stages were recorded. None of bioactive compounds exhibited any correlation with oil content. Principle component analysis revealed that of the total variation, O. gratissimum contributed maximum (61.58 %) variation followed by O. basilicum (35.36 %). The results of current study suggested that the different Ocimum species can be harvest at various growth stages to get the maximum yield and target chemical compounds based on the needy industrial value.

This report describes the synthesis of five new colorful 6-aryldibenzo[b,d]pyrylium cation salts, a largely unexplored structural unit. These rare compounds are benzannulated structural derivatives of the well-known flavylium cations found widespread in natural pigments. These new dyes are directly compared to three previously synthesized 6-aryldibenzo[b,d]pyrylium cation salts as well as eight colorful isomeric 9-aryldibenzo[b,d]pyrylium cation, or 9-arylxanthylium, salts. The 9-arylxanthylium unit is commonly found in the biologically important rhodamine and rosamine dyes, yet six of the analogs presented in this study were either previously unreported or not isolated. The visual and spectroscopic properties of all 16 compounds were analyzed as a function of the structural differences between the compounds. All compounds displayed reversible halochromism in organic solution, displaying bright colors under acidic conditions and becoming colorless under basic conditions. (C) 2020 Elsevier Ltd. In this study, the unique capacity of bifunctional BrOnsted bases to generate alpha-branched ketone dienolates and control both site- and stereoselectivity of their addition reactions to representative classes of carbon electrophiles (i.e., vinyl sulfones, nitroolefins, formaldehyde) is documented. We demonstrate that by using selected chiral tertiary amine/squaramide catalysts, the reactions of beta,gamma-unsaturated cycloalkanones proceed through the dienolate C alpha almost exclusively and provide all-carbon quaternary cyclic ketone adducts in good yields with very high enantioselectivities. A minor amount (<5%) of gamma-addition is observed when nitroolefins are used as electrophiles. The parent acyclic ketone dienolates proved to be less reactive under these conditions, and thus still constitute a challenging class of substrates. Quantum chemical calculations correctly predict these differences in reactivity and explain the observed site-specificity and enantioselectivity.
