Atopic dermatitis (AD) is a skin inflammatory disease characterized by erythema, eruption, lichenification and pruritus. Shi Zhen Formula (SZF), an empirical Chinese herbal preparation, has clin. efficacy in relieving the symptoms of AD patients. However, the underlying mol. mechanisms of SZF remained unclear. We aimed to investigate the anti-AD effects of SZF and elucidate its underlying mol. mechanisms using in vitro and in vivo models of AD. High-performance liquid chromatog. anal. was performed for quality control of SZF extract The anti-inflammatory effect of SZF was investigated through evaluating the levels of nitric oxide (NO), chemokines and pro-inflammatory cytokines in the lipopolysaccharide (LPS) stimulated RAW264.7 cells. AD-like skin lesions in female BALB/c mice were induced by 2,4-dinitrochlorobenzene (DNCB). SZF (3.15, 6.30 and 9.45 g/kg) and dexamethasone (5 mg/kg) were administered by gavage daily for 15 consecutive days. The body weight, skin thickness, skin dermatitis severity and scratching behaviors were recorded throughout the study. Histol. anal., reverse transcription-quant. polymerase chain reaction (RT-PCR), western blot (WB) and ELISA anal. were used to illuminate the mol. targets associated with the anti-AD effects of SZF. SZF markedly decreased the epidermal thickening and infiltration of mast cells in the ears and dorsal skin of the 2,4-dinitrochlorobenzene (DNCB)-treated mice. SZF not only suppressed the levels of IgE (IgE), histamine, thymic stromal lymphopoietin (TSLP) and IL-4 in the serum but also suppressed the over-production of IL-4 and IL-6 and gene expressions of IL-4, IL-13, IL-31 and TSLP in the dorsal skin. Moreover, SZF improved epidermal barrier by increasing the protein expressions of filaggrin, involucrin and loricrin and inhibited the activation of NF-κB p65 pathway in the dorsal skin of the DNCB-treated mice. SZF alleviates DNCB induced AD-like skin lesions in mice through regulating Th1/Th2 balance, improving epidermal barrier and inhibiting skin inflammation. Our research findings provide scientific footing on the use of this Chinese herbal formula for the treatment of AD.

Oxygen reduction reactions (ORRs) with one- or two-electron-transfer pathways are the essential process for aprotic metal-oxygen batteries, in which the stability of superoxide intermediates/products (O2-, LiO2, NaO2, etc.) mainly dominates the ORR activity/stability and battery performance. However, little success in regulating the stability of the superoxides has been achieved due to their highly reactive characteristics. Herein, we identified and modulated the stability of superoxides by introducing anthraquinone derivatives as cocatalysts which functioned as superoxide trapper adsorbing the superoxides generated via surface-mediated ORR and then transferring them from the solid catalyst surface into electrolyte. Among the studied trappers, 1,4-difluoroanthraquinone (DFAQ) with electron-withdrawing groups showed the highest adsorption towards superoxides and could efficiently stabilize LiO2 in electrolyte, which greatly promoted the surface-mediated ORR rate and stability. This highlighted the magnitude of adsorption between the trapper and LiO2 on the ORR activity/stability. Using an aprotic Li-O2 battery as a model metal-O2 battery, the overall performance of the cell with DFAQ was substantially improved in terms of cell capacity, rate capability and cyclic stability. These results represent a significant advance in the understanding of ORR mechanisms and promoting the performance of metal-O2 batteries.

Potassium-ion batteries (PIBs) are attracting great interest for large-scale energy storage owing to the abundant resources and low redox potential of K+/K. However, the large volume changes and slow kinetics caused by the larger ionic radius of K+ for cathode materials remain a critical challenge for PIBs. Herein, we construct few-layered covalent organic frameworks integrated with carboxylated carbon nanotubes (DAAQ-COF@CNT) as cathode materials for PIBs. The synthesized DAAQ-COF@CNT features numerous active sites, a stable conductive framework, and an appropriate surface area with nanopores, which can render high elec. conductivity, shorten the ion/electron diffusion distance, and accelerate K+ diffusion. In consequence, the DAAQ-COF@CNT delivers a high reversible capacity of 157.7 mAh g-1 at 0.1 A g-1, an excellent rate capability of 111.2 mAh g-1 at 1 A g-1, and a long cycling stability of 77.6% capacity retention after 500 cycles at 0.5 A g-1. The integrated characterization of ex situ XPS, Fourier transform IR spectroscopy, and theor. simulation discloses that the storage mechanism of DAAQ-COF@CNT is based on the reversible reaction between electroactive C=O groups and K+ during two successive steps. This work provides a promising high-performance cathode material for PIBs and encourages the development of new types of covalent organic frameworks for PIBs.

As part of an effort to identify 5-HT1A antagonists that did not possess typical arylalkylamine or keto/amido-alkyl aryl piperazine scaffolds, prototype compound I was identified from earlier work in a combined 5-HT1A antagonist/SSRI program. This quinolyl-piperazinyl piperidine analog displayed potent, selective 5-HT1A antagonism but suffered from poor oxidative metabolic stability, resulting in low exposure following oral administration. SAR studies, driven primarily by in vitro liver microsomal stability assessment, identified compound II, which displayed improved oral bioavailability and lower intrinsic clearance. Further changes to the scaffold resulted in a loss in potency. Compound II displayed cognitive enhancing effects in a number of animal models of learning and memory, enhanced the antidepressant-like effects of the SSRI fluoxetine, and reversed the sexual dysfunction induced by chronic fluoxetine treatment.

Synergistic stabilization of Pickering emulsions by a mixture of surfactants and colloidal particles has received increasing interest in recent years but only a few of them can produce high internal phase double emulsions (HIPDEs) with good stability. In this study, we present a feasible and common method of preparing Pickering high internal phase emulsions (HIPEs) with tunable inner morphol. costabilized by a biosurfactant lecithin and silica nanoparticles. We investigate the influence of the pH value on the interfacial behavior of lecithin and elucidate the synergistic mechanism between lecithin and silica nanoparticles in different conditions in the stability of as-prepared emulsions. Specifically, water-in-oil (W/O) Pickering HIPEs can be successfully stabilized by lecithin and hydrophobic silica nanoparticles in a wide pH range (pH 1-10), while catastrophic phase inversion occurred at high pH values (pH ≥ 11). Interestingly, stable water-in-oil-in-water (W/O/W) high internal phase double emulsions (HIPDEs) can also be prepared via a two-step method by the cooperation of lecithin and silica nanoparticles. Moreover, functional interconnected porous monoliths and microspheres are facilely fabricated by emulsion templates and their potential applications are explored.

Abstract: Purpose: Temozolomide (TMZ), a cytotoxic DNA alkylating agent, is the main chemotherapy used for the treatment of high grade astrocytomas. The active alkylator, methylhydrazine, is not recovered in urine and thus renal function is not expected to affect clearance. Prescribing information for TMZ states pharmacokinetics have not been studied in adults with poor renal function, eGFR < 36 mL/min/1.73 m2. We reviewed our clin. experience with TMZ in patients with impaired renal function to evaluate safety of administering full dose TMZ. Methods: The primary endpoint was to characterize the incidence and severity of thrombocytopenia in patients with eGFR < 60 mL/min/1.73 m2 who received TMZ for treatment of high grade gliomas (HGG) or primary CNS lymphoma (PCNSL). Secondary endpoints included incidence and severity of neutropenia, lymphopenia hepatotoxicity, and number of TMZ cycles administered. Medical records of patients with HGG or PCNSL treated with TMZ from Oct. 1, 2016-Sept. 30, 2019 were accessed to identify cases for this study. Results: Thirty-two patients were eligible for this study. Of the seven patients with eGFR < 36 mL/min/1.73m2, 38/39 cycles (97%) were completed without grade 3-4 thrombocytopenia. No patients experienced grade 3-4 neutropenia, and grade 3-4 lymphopenia occurred in 5 cycles (15%). One patient discontinued TMZ 7 days prior to completion of radiation due to thrombocytopenia. Conclusion: Hematol. toxicity in patients with severe renal dysfunction, eGFR < 36 mL/min/1.73m2, is similar to that of patients with normal renal function. Severe renal impairment does not preclude use of temozolomide, but cautious monitoring of blood counts is warranted. Novel molecularly imprinted polymers (MIPs) based on multiwalled carbon nanotubes (MWNTs) were synthesized using carbofuran as template, methacrylic acid as functional monomer, and trimethylolpropane trimethacrylate as cross-liking agent, resp. Characterization results showed that the carbofuran MIPs have been successfully grafted onto the surface of MWNTs as a thin layer with high stability. The results of adsorption dynamics indicated that the synthesized MWNT-MIPs displayed a biphase adsorption profile and good selective recognition to carbofuran with equilibrium adsorption of 106.2 mg/g. The MWNTs-MIPs synthesized were further applied as the adsorbent material of solid-phase extraction (SPE) for the pretreatment of carbofuran in human serum, analyzed using high performance liquid chromatograph (HPLC). The recoveries obtained ranged from 89.0±4.8 to 93.6±3.2, showing that the MWNTs-MIPs-SPE system developed have specific recognition toward carbofuran. Results above indicated that the proposed system filled with synthesized MWNTs-MIPs provided a fast and selective extraction of carbofuran in serum.
