Construction of an efficient electrocatalyst for the oxygen reduction reaction (ORR) and oxygen evolution reaction (OER) with low overpotential and cycling stability for lithium-oxygen batteries still remains a puzzling challenge. Herein, we propose a scalable approach to integrate ZIF derivatives into cavity porous carbon nanofibers (CPCNFs) via an electrospinning technique and thermal treatment (Zn/CoNC@CPCNFs). The ultralong interconnected nanofiber matrix is beneficial, and the developed Zn/CoNC@CPCNFs catalyst with excellent flexibility can be utilized as a free-standing electrode based on an air-cathode. Moreover, this confinement strategy ensures the dispersion of Co-based species and abundant porosity structure, which contributes to the transport and adsorption of oxygen and exposes more Co-N coordination catalytic centers, as a result of a drastically ultralow voltage gap. Consequently, a cell based on a Zn/CoNC@CPCNF electrode presents remarkably decreased charge-discharge polarization (0.36 V), a high initial discharge capacity with an ultra-low overpotential of 0.59 V, and long-term cyclability with a cut-off capacity of 0.2 mA h cm-2 at 0.02 mA cm-2. We hope that our protocol will offer instruction for the design and application of oxygen electrocatalysts for energy conversion and storage.

The total syntheses of the marine natural products neohalicholactone and halicholactone, in enantiomerically pure form, were reported. Key steps in the syntheses of each compound included a cis-selective Wittig reaction, stereoselective cyclopropanation, nine-membered lactone formation using the Yamaguchi method and late-stage stereoselective Cr(II)/Ni(II) mediated coupling of vinyl iodides (R,E)-Me(CH2)4CH(OSiPh2CMe3)CH:CHI and (1E,3R,5Z)-MeCH2CH:CHCH2CH(OSiMe2CMe3)CH:CHI with aldehyde I. In the case of the neohalicholactone synthesis the two major components, which were coupled in this convergent synthesis, were each derived from the enantiomers of com. available malic acid. The synthesis served to confirm the original assignment of absolute configuration which was made by Yamada and Clardy. It was also demonstrated, through the preparation of diastereoisomers, that another reported compound closely related to neohalicholactone was likely to be the 15-epineohalicholactone.

The synthesis of ruthenium(II)-terpyridine complexes-containing glyconanoparticles (Ru-GNPs) for one- and two-photon excited fluorescence imaging was described. Ru-GNPs were built from diblock copolymers of pentafluorophenyl acrylate (PFA) and 2-(2′,3′,4′,6′-tetra-O-acetyl-尾-D-glucosyloxy)ethyl methacryalte (AcGlcEMA), prepared by reversible addition-fragmentation chain transfer (RAFT) polymerization The pendant reactive pentafluorophenyl ester groups were substituted and the copolymer cross-linked by the two-photon absorption (TPA) chromophore, bis[4′-(4-aminophenyl)-2,2′:6′,2”-terpyridyl]ruthenium(II) hexafluorophosphate. After deacetylation of the acetyl-protecting groups in the AcGlcEMA block, TPA nanoparticles with cross-linked ruthenium(II)-terpyridine complexes as the core and glycopolymers as the corona were obtained. The resulting Ru-GNPs exhibit good water dispersity, one-photon absorption and emission and low cytotoxicity towards KB cell line. Ru-GNPs also undergo two-photon excitation when excited in the near-IR (NIR, 720-880 nm) in water, with TPA cross-sections in the range of 0.2-22 G枚ppert-Mayer (GM, 1 GM = 10-50 cm4 s1 photon-1). Ru-GNPs are thus potentially useful as effective cellular labeling agents for one- and two-photon excited fluorescence imaging.

HIV-associated sensory neuropathy (HIV-SN) is a disabling complication of HIV disease and antiretroviral therapies (ART). Since stavudine was removed from recommended treatment schedules, the prevalence of HIV-SN has declined and associated risk factors have changed. With stavudine, rs1799964*C (TNF-1031) associated with HIV-SN in Caucasians and Indonesians but not in South Africans. Here, we investigate associations between HIV-SN and rs1799964*C and 12 other polymorphisms spanning TNF and seven neighboring genes (the TNF-block) in Indonesians (n = 202; 34/168 cases) and South Africans (n = 75; 29/75 cases) treated without stavudine. Haplotypes were derived using fastPHASE and haplotype networks built with PopART. There were no associations with rs1799964*C in either population. However, rs9281523*C in intron 10 of BAT1 (alternatively DDX39B) independently associated with HIV-SN in Indonesians after correcting for lower CD4 T-cell counts and >500 copies of HIV RNA/mL (model p = 0.0011, Pseudo R2 = 0.09). rs4947324*T (between NFKBIL1 and LTA) independently associated with reduced risk of HIV-SN and shared haplotype 1 (containing no minor alleles) associated with increased risk of HIV-SN after correcting for greater body weight, a history of tuberculosis and nadir CD4 T-cell counts (model: p = 0.0003, Pseudo R2 = 0.22). These results confirm TNF-block genotypes influence susceptibility of HIV-SN. However, critical genotypes differ between ethnicities and with stavudine use.

Cryptosporidiosis is a human gastrointestinal disease caused by protozoans of the genus Cryptosporidium, which can be fatal in immunocompromised individuals. The essential enzyme, thymidylate synthase (TS), is responsible for de novo synthesis of deoxythymidine monophosphate. The TS active site is relatively conserved between Cryptosporidium and human enzymes. In previous work, we identified compound 1, (2-amino-4-oxo-4,7-dihydro-pyrrolo[2,3-d]pyrimidin-methyl-phenyl-L-glutamic acid), as a promising selective Cryptosporidium hominis TS (ChTS) inhibitor. In the present study, we explore the structure-activity relationship around 1 glutamate moiety by synthesizing and biochem. evaluating the inhibitory activity of analogs against ChTS and human TS (hTS). X-Ray crystal structures were obtained for compounds bound to both ChTS and hTS. We establish the importance of the 2-phenylacetic acid moiety methylene linker in optimally positioning compounds 23, 24, and 25 within the active site. Moreover, through the comparison of structural data for 5, 14, 15, and 23 bound in both ChTS and hTS identified that active site rigidity is a driving force in determining inhibitor selectivity.
