Effect of cooking temperature and time on total phenolic content, total flavonoid content and total in vitro antioxidant activity of garlic

Objective To investigate the effect of cooking temperature and time on the total phenolic content, total flavonoid content and antioxidant activity of aqueous and ethanolic extracts of garlic. Results The mean total phenolic content of fresh garlic were 303.07 +/- 6.58 mg gallic acid equivalent per 100 g (GAE/100 g) and 638.96 +/- 15.30 mg GAE/100 g of plant material for the aqueous and ethanolic extracts respectively. The mean total flavonoid content 109.78 +/- 6.78 mg quercetin equivalent per 100 g (QE/100 g) and 258.47 +/- 12.37 QE/100 g for aqueous and ethanolic extracts respectively. Fourier transform infrared spectral data showed absorptions in the range for carboxylic acids, hydroxyl group, esters, and alcohols, confirming the presence of phenols and flavonoids in the extracts. Cooking temperature had a significant effect on total phenolic content and total flavonoid content while cooking time did not have a significant effect on the phytochemicals and antioxidant activity.

Microplastics in soils: A review of methods, occurrence, fate, transport, ecological and environmental risks

The global prevalence of microplastics (MPs) poses a potential threat and unpredictable risk to the function and health of environmental systems. However, the research progress of soil MPs is restricted by the inherent technical inconformity and difficulties in analyzing particles in complex matrices. Here, we reviewed a selection of papers and then extrapolated a tentative standardized method for such analyses. The multiple sources of soil MPs in soil need to be quantified. Global monitoring data of soil MPs is far from sufficient. The interaction between MPs and different properties and environmental factors controls the migration and retention of MI’s in soil. The migration behavior and key mechanisms of MI’s in real-world environments remain to be determined. The presence of MPs threatens soil microbial-plant-animal ecosystem function and health, and may enter the human body through the food chain, although the extent of these hazards is currently debated. In particular, attention should be paid to the potential transport and ecotoxicological mechanisms of contaminants derived and adsorptive from MPs and of harmful microorganisms (such as pathogens) attached as biofilms. Although there exist preliminary studies on soil MPs, it is urgent to consider the diversity of MPs as a suite of contaminants and to systematically understand the sources, flux and effects of these artificial pollutants in time and space from the perspective of plastic environmental cycle. More comprehensive quantification of their environmental fate is undertaken to identify risks to global human and ecological systems. From the perspective of controlling soil MP pollution, the responsibility assignment of government manage-producer-consumer system and the strategy of remediation should be implemented. This review is helpful for providing an important roadmap and inspiration for the research methods and framework of soil MPs and facilitates the development of waste management and remediation strategies for regional soil MP contamination. Digoxin Combined with Aerobic Interval Training Improved Cardiomyocyte Contractility

Digoxin is a cardiotonic that increases the cardiac output without causing deleterious effects on heart, as well as improves the left ventricular performance during physical exercise. We tested whether the association between chronic digoxin administration and aerobic interval training (AIT) promotes beneficial cardiovascular adaptations by improving the myocardial contractility and calcium (Ca (2+)) handling. Male Wistar rats were randomly assigned to sedentary control (C), interval training (T), sedentary digoxin (DIGO) and T associated to digoxin (TDIGO). AIT was performed on a treadmill (1h/day, 5 days/week) for 60 days, consisting of successive 8-min periods at 80% and 20% of VO (2) max for 2 min. Digoxin was administered by orogastric gavage for 60 days. Left ventricle samples were collected to analysis of Ca (2+) handling proteins; contractility and Ca (2+) handling were performed on isolated cardiomyocytes. TDIGO group had a greater elevation in fractional shortening (44%) than DIGO, suggesting a cardiomyocyte contractile improvement. In addition, T or TDIGO groups showed no change in cardiomyocytes properties after Fura2-acetoxymethyl ester, as well as in sarcoplasmic reticulum Ca (2+-) ATPase (SERCA2a), phospholamban and calcineurin expressions. The main findings indicate that association of digoxin and aerobic interval training improved the cardiomyocyte contractile function, but these effects seem to be unrelated to Ca (2+) handling.

Characterization of N-phenylmaleimide-terminated poly(ethylene glycol)s and their application to a tetra-arm poly(ethylene glycol) gel

Tetra-arm poly(ethylene glycol) (TetraPEG) gels are tough materials whose toughness originates from their uniform network structure. They can be formed by combining the termini of tetra-arm polymers via chemical reactions with high conversion efficiency, such as the Michael addition, condensations using an active ester group, and alkyne-azide cycloadditions. Herein, we report the synthesis of a tetra-PEG gel using a tetra-arm polymer with N-phenylmaleimide moieties at the polymer ends (tetra-N-aryl MA PEG) as a scaffold. Tetra-N-aryl MA PEG can be obtained via a simple maleimidation using the modification agent p-maleimidophenyl isocyanate (PMPI), which directly transforms the hydroxy groups at the polymer ends into reactive N-aryl maleimide groups in a one-pot reaction. The thus-obtained tetra-N-aryl MA PEG was fully characterized using high-performance liquid chromatography (HPLC), matrix-assisted laser desorption ionization time of flight mass spectrometry, and proton nuclear magnetic resonance spectroscopy. HPLC analysis not only demonstrated the high purity of tetra-N-aryl MA PEG and the full conversion of the hydroxy groups, but also provided an effective characterization method for N-aryl maleimide-based PEG using a simple protocol, which enables us quantitative analysis of functionalized polymers with different N-aryl maleimide numbers. Furthermore, we fabricated a TetraPEG gel via Michael addition of the obtained tetra-N-aryl MA and thiol-terminated TetraPEGs. Most proteins in the alpha-macroglobulin (alpha M) superfamily contain reactive thiol esters that are required for their biological function. Here, we have characterized the human alpha 2-macroglobulin (A2M) and complement component C3 mutants A2M Q975C and C3 Q1013C, which replace the CGEQ thiol ester motifs of the original proteins with the disulfide-forming sequence CGEC. Mass spectrometry showed that the intended disulfide was formed in both proteins. The correct folding and native conformation of A2M Q975C were shown by its assembly to a tetramer, an initially slow electrophoretic mobility with a demonstrable conformational collapse induced by proteolysis, functional protease trapping, and conformation-dependent interactions with low-density lipoprotein receptorrelated protein 1. However, A2M Q975C had a decreased capacity to inhibit trypsin and was more susceptible to cleavage by trypsin or thermolysin when compared to wild-type A2M. C3 Q1013C also folded correctly and was initially in a native conformation, as demonstrated by its cation exchange elution profile, electrophoretic mobility, and interaction with complement factor B, although it assumed a conformation that was distinct from native C3, C3b, or C3(H2O) when cleaved by trypsin. These results demonstrate that disulfides can substitute thiol esters and maintain the native conformations of A2M and C3.
