Girardinia diversifolia, also known as Himalayan nettle, is a perennial herb used in Nepal to make fiber as well as in traditional medicine for the treatment of several diseases. To date, phytochem. studies and biol. assays on this plant are scarce. Thus, in the present work, the G. diversifolia extracts have been evaluated for their potential pharmaceutical, cosmetic and nutraceutical uses. For this purpose, detailed phytochem. analyses were performed, evidencing the presence of phytosterols, fatty acids, carotenoids, polyphenols and saponins. The most abundant secondary metabolites were β- and γ-sitosterol (11 and 9% dw, resp.), and trans syringin (0.5 mg/g) was the most abundant phenolic. Fatty acids with an abundant portion of unsaturated derivatives (linoleic and linolenic acid at 22.0 and 9.7 mg/g resp.), vitamin C (2.9 mg/g) and vitamin B2 (0.12 mg/g) were also present. The antioxidant activity was moderate while a significant ability to inhibit acetylcholinesterase (AChE), butyrilcholinesterase (BuChE), tyrosinase, α-amylase and α-glucosidase was observed A cytotoxic effect was observed on human ovarian, pancreatic and hepatic cancer cell lines. The effect in hepatocarcinoma cells was associated to a downregulation of the low-d. lipoprotein receptor (LDLR), a pivotal regulator of cellular cholesterol homeostasis. These data show the potential usefulness of this species for possible applications in pharmaceuticals, nutraceuticals and cosmetics.

Natural organic matter (NOM) from Sphagnum peat soil is extracted in water and subjected to several investigations to obtain structural and conformational information. Data show that the extracted NOM is self-organized in colloidal aggregates of variable sizes (from nano to micro scales, depending on the solvent composition, i.e., ultrapure water, solutions with denaturing agents, acetone, ethanol). Aggregates are formed by highly heterogeneous classes of organic compounds According to the results of NMR and fluorescence measurements, the three-dimensional structure of aggregates, revealed by scanning electron microscope imaging, is supposed to be stabilized by the exposition of polar functional groups to the solvent, with consequent formation of hydrogen bonds, dipole-interactions and cation bridging. In contrast, the inner part of the aggregates displays hydrophobic features and is hypothesized to be further reinforced by the establishment of π-stacking interactions. The structure is assumed to be a supramol. aggregation of small-medium oligomeric fragments (Max 750 Da) in which priority pollutants are entrapped by dispersive forces. The structures are shown to be nanosized spheroidal particles further aggregated to form higher dimension supra-structures. Carbohydrates play primary role, stabilizing the structure and giving marked hydrophilic properties to the aggregates.

Gas chromatog.-mass spectrometry (GC-MS) is one of the major platforms for analyzing volatile compounds in complex samples. However, automatic and accurate extraction of qual. and quant. information is still challenging when analyzing complex GC-MS data, especially for the components incompletely separated by chromatog. Deep-Learning-Assisted Multivariate Curve Resolution (DeepResoln.) was proposed in this study. It essentially consists of convolutional neural networks (CNN) models to determine the number of components of each overlapped peak and the elution region of each compound With the assistance of the predicted elution regions, the informative regions (such as selective region and zero-concentration region) of each compound can be located precisely. Then, full rank resolution (FRR), multivariate curve resolution-alternating least squares (MCR-ALS) or iterative target transformation factor anal. (ITTFA) can be chosen adaptively to resolve the overlapped components without manual intervention. The results showed that DeepResoln. has superior compound identification capability and better quant. performances when comparing with MS-DIAL, ADAP-GC and AMDIS. It was also found that baseline levels, interferents, component concentrations and peak tailing have little influences on resolution result. Besides, DeepResoln. can be extended easily when encountering unknown component(s), due to the independence of each CNN model. All procedures of DeepResoln. can be performed automatically, and adaptive selection of resolution methods ensures the balance between resolution power and consumed time. It is implemented in Python and available at https://github.com/XiaqiongFan/DeepResoln.

Salsa is a liquid-solid food containing jalapenõ and serrano peppers, which result in multistate outbreak in 2008. Storage temperature and sodium chloride (NaCl) concentration of salsa vary depending on the climate, season, and type of product. In this regard, effect of growth conditions, namely, low temperature (15 °C) or NaCl concentration (4.5%) on the resistance of Escherichia coli O157:H7 ATCC 35150 to ohmic heating was identified in this study. Cells of E. coli O157:H7 ATCC 35150 grown under different growth conditions was inoculated into prepared salsa sample, and then subjected to ohmic heating. Mechanisms of resistance acquisition were identified by transcriptional responses, membrane fatty acid changes and confirmed with propidium iodide (PI) uptake values. Resistance of the pathogen to ohmic heating decreased when growth temperature decreased from 37 °C to 15 °C while increased resistance was observed for this pathogen when grown with 4.5% NaCl. Several heat stress related genes such as dnaK, rpoH, grpE, groES, htpG, and htpX were up-regulated (â‰? fold change) as growth temperature decreasedwhile groEL, dnaK, rpoH were up-regulated when grown with high NaCl concentration in the present study. The ratio of unsaturated fatty acids (USFA) to saturated fatty acids (SFA) of pathogen increased slightly (+0.16) or significantly (+0.79) with increasing NaCl concentration or decreasing temperature, resp. These results indicate that the cell membrane of the pathogen grown at low temperature was more susceptible to heat than when grown under optimal conditions or high NaCl concentration Cell membrane damage measured by PI uptake values of the pathogen grown with high NaCl concentration were not significantly different from those of the control (p > 0.05), while the values were significantly higher for the pathogen grown at low temperature and subjected to ohmic heating (p < 0.05). Based on these results we suggest that resistance of the pathogen grown at low temperature to ohmic heating decreased because of dominant cell membrane damage compared to induced heat stress related genes. The cell membrane damage was dominant by means of an increased ratio of USFA to SFA. On the other hand, pathogen resistance increased when grown in medium of high NaCl concentration because of induced heat stress related genes. Apricot and peach oils are often found in pharmaceutical dosage forms. They can be purchased from different manufacturers, which use various approaches for their standardization. The main determinant of any quality oil is represented by fatty acids (FA). Therefore, determination and comparison of FA composition of these oils and assessment of their storage period influence on it were performed. FA composition was determined by gas chromatog. using specific Rt-2560 column and Supelco 37 Component FAME Mix. 26 FA in apricot and 28 FA in peach oil samples were identified. Oleic (67.21% in apricot oil; 63.29% in peach oil) and linoleic (22.13% in apricot oil; 25.40% in peach oil) acids were characterized by the highest average concentration in both oils. Among saturated FA, palmitic acid was the first (5.87% in apricot oil; 5.95% in peach oil) and stearic acid was the second (2.08% in apricot oil; 2.92% in peach oil). Total saturated fatty acids (TSFA) and total unsaturated fatty acids (TUFA) values also didn’t have significant differences. The oils have close composition, despite various expiration dates and manufacturers.

This work focused on the application of calcium (0.1-1% w/v) to overcome the inhibition caused by the high loadings (2% volume/volume) of fat, oil, and grease (FOG) in the context of biomethane production, organic removal, and microbial community shift. Addition of 0.5% calcium showed maximum biomethane production (6-fold increase); biomethane production decreased following the addition of calcium (>0.5%). The highest organic removal rates were 83 and 89% upon the addition of 0.3 and 0.5% calcium, resp. Addition of calcium facilitated the growth of bacteria of phylum Firmicutes from the Clostridium, Syntrophomonas, and Sedimentibacter genera. The population of members from the genus Methanosaeta increased after the addition of 0.5% calcium, which is one of the factors responsible for high biomethane production This study demonstrated that addition of calcium is an attractive strategy to avoid the inhibition of the growth of anaerobic microflora due to the presence of high FOG concentrations

In this study, we examined the suppressive effect of a methanolic extract (ME) of WBM leaf and fractionated components thereof on live C. acnes-induced in vitro and in vivo inflammation. Using a bioassay-monitored isolation approach and a combination of liquid-liquid extraction and column chromatog., the ME was then separated into n-hexane, Et acetate, n-butanol and water-soluble fractions. The hexane fraction exerted the most potent anti-inflammatory effect, suppressing C. acnes-induced interleukin-8 (IL-8) production by 36%. The ethanol-soluble fraction (ESF), which was separated from the n-hexane fraction, significantly inhibited C. acnes-induced activation of mitogen-activated protein kinase (MAPK)-mediated cellular IL-8 production Similarly, the ESF protected against C. acnes-stimulated mouse ear swelling, as measured by ear thickness (20%) and biopsy weight (23%). Twenty-four compounds in the ESF were identified using gas chromatograph-mass spectrum (GC/MS) anal. Using co-cultures of C. acnes and THP-1 cells, β-ionone, a compound of the ESF, reduced the production of IL-1β and IL-8 up to 40% and 18%, resp. β-ionone also reduced epidermal microabscess, neutrophilic infiltration and IL-1β expression in mouse ear. We also found evidence of the presence of anti-inflammatory substances in an unfractionated phenolic extract of WBM leaf, and demonstrated that the ESF is a potential anti-inflammatory agent for modulating in vitro and in vivo C. acnes-induced inflammatory responses.

This study scientifically validated phytochems. in M. uniflorum for its functional potential in management of Metabolic Syndrome (MS). This study help nutraceutical industries to develop functional foods using M. uniflorum seeds to make porridges and soups or nutraceutical supplements with bioflavonoids isolated from M. uniflorum for management of metabolic disorders by mitigating hyperlipidemia, oxidative stress, and inflammation.

This work focuses on production of biodiesel from natural and refined castor seed oil. Oil was extracted with normal-hexane solvent at 60°C using Soxhlet apparatus and pretreated by alk. refining process before transesterification with methanolic potassium hydroxide solution as catalyst. The effect of alk. refining on the oil characteristics and biodiesel quality parameters was investigated using standard test methods and fatty acid Me ester was characterized using gas chromatog. Results revealed that alk. refining had significant improvement on the oil and biodiesel characteristics. Yield of 43.9 and 46.2% were obtained for refined and unrefined castor oil resp. while biodiesel yield from refined and unrefined castor oil was 43.5 and 41.7% resp. Ricinoleic acid (84.9%) was prevalent in the oil with lignoceric (0.03%) as the smallest. The conformity of refined castor oil biodiesel with EN 14214, ASTM and India biodiesel standards makes it viable economically in Nigeria.
