Osteoarthritis (OA) is a degenerative disease which serious affects patients. Ligusticum chuanxiong (CX) has been shown to have a certain curative effect on osteoarthritis in traditional Chinese medicine therapy. This study is based on network pharmacol. and mol. docking technol. to explore the potential mechanism of CX. Components of CX to treat osteoarthritis were screened in the TCMSP database and targets were predicted by the PharmMapper database, the osteoarthritis targets were collected from the GeneCards database, and intersection genes were found to be the possible targets of CX anti-OA. The STRING database and Cytoscape software were utilized for protein-protein interaction anal. and further screening of core targets. The Metascape database was used for KEGG and GO enrichment analyses. Then, the top 10 pathways were selected to construct “drug-compound-target-pathway-disease” network anal. Finally, mol. docking was used to analyze the binding affinity of seven compounds with core targets and TNF-α. Seven compounds with 253 non-repetitive targets of CX were screened fromthe TCMSP database and 60 potential intersection targets of CX anti-OA were found. PPI network anal. showed that the core targets were ALB, AKT1, IGF1, CASP3, MAPK1, ANXA5, and MAPK14, while GO and KEGG pathway enrichment analyses showed that the relevant biol. processes involved in the treatment of osteoarthritis by CX might include the MAPK cascade and reactive oxygen species metabolic process. The KEGG pathway anal. result was mainly associated with the MAPK signaling pathway and PI3K-AKT signaling pathway. We further docked seven ingredients with MAPK1 and MAPK14 enriched in the MAPK pathway, and TNF-α as the typical inflammatory cytokine. The results also showed good binding affinity, especially FA, which may be the most important component of CX anti-OA. Our research revealed the potential mechanism of CX in the treatment of OA, and our findings can also pave the way for subsequent basic exptl. verification and a new research direction. Here, a new approach is reported to construct structural colored hydrogel films with excellent UV blocking performance for contact lenses. However, such hydrogel films cannot effectively block UV light. By slightly breaking the structural periodicity (quasi-periodic structure), strong diffuse scattering or pseudoabsorption of light can occur for all of the wavelengths shorter than a structural characteristic length, leading to perfect UV blocking. According to the theor. prediction, a structural colored hydrogel film with nearly periodic polystyrene sphere arrays in poly(hydroxyethyl methacrylate) hydrogel matrix is fabricated; this hydrogel film possesses brilliant colors and perfect UV blocking, and the core particle composition and size have been investigated in detail for the optimized properties of contact lenses. A quasi-solid-state gel polymer electrolyte (GPE) is designed by integrating a free-standing cross-linked film and an injectable polymer electrolyte for applications in acetonitrile-based supercapacitors (SCs) using activated carbon (AC) electrodes. The synthesized GPE exhibits a high ionic conductivity over a wide temperature range (∼1.7 × 10-3 S cm-1 at -20°C to ∼7.9 × 10-3 S cm-1 at 60°C) as well as thermal stability and flexibility. A quasi-solid-state SC equipped with the GPE exhibits ∼13% higher specific capacitance (∼56% smaller Warburg resistance) at room temperature (RT) than that of an SC containing a corresponding liquid electrolyte. The quasi-solid-state SC also exhibits excellent electrochem. characteristics over a temperature range of -20 to 60°C with only ∼23% capacitance loss at -20°C compared to that at RT. Furthermore, the quasi-solid-state SC maintains ∼93% of its initial capacitance (only ∼4% increase in ESR) after 120-h storage at 85°C (above the b.p. of acetonitrile). This capacitance is superior to an SC equipped with a film-only GPE (∼73% capacitance retention and ∼270% increase in ESR). The proposed strategy provides new insights into the development of GPE designs for practical applications to solid-state SCs operating over a wide temperature range. Platelet concentrate products are increasingly used in many medical disciplines due to their regenerative properties. As they contain a variety of chemokines, cytokines, and growth factors, they are used to support the healing of chronic or complicated wounds. Real-time PCR analyses of PRGF-treated fibroblasts and skin explants confirmed the induction of ECM-related genes, in particular transforming growth factor beta-induced protein (TGFBI), fibronectin 1 (FN1), matrix metalloproteinase-9 (MMP-9), transglutaminase 2 (TGM2), fermitin family member 1 (FERMT1), collagen type I alpha 1 (COL1A1), a disintegrin and metalloproteinase 19 (ADAM19), serpin family E member 1 (SERPINE1) and lysyl oxidase-like 3 (LOXL3). The induction of these genes was time-dependent and in part influenced by the epidermal growth factor receptor (EGFR). Moreover, PRGF induced migration and proliferation of the fibroblasts. Taken together, the observed effects of PRGF on human fibroblasts may contribute to the underlying mechanisms that support the beneficial wound-healing effects of thrombocyte concentrate products. In recent years, shape-memory polymers (SMPs) have received extensive attention to be used as actuators in a broad range of applications such as medical and robotic devices. Their ability to recover large deformations and their capability to be stimulated remotely have made SMPs a superior choice among different smart materials in various applications. In this study, a ductile SMP composite with enhanced shape recovery ability is synthesized and characterized. This SMP composite is made by a mixture of acrylate-based crosslinkers and monomers, as well as polystyrene (PS) with UV curing. The composite can achieve almost 100% shape recovery in 2 s by hot water or hot air. This shape recovery speed is much faster than typical acrylate-based SMPs. In addition, the composite shows excellent ductility and viscoelasticity with reduced hardness. Mol. dynamics (MD) simulations are performed for understanding the curing mechanism of this composite. This bioinformatics study aimed to reveal potential cross-talk genes, related pathways, and transcription factors between periimplantitis and rheumatoid arthritis (RA). The datasets GSE33774 (seven periimplantitis and eight control samples) and GSE106090 (six periimplantitis and six control samples) were included from the National Center for Biotechnol. Based on this, a protein-protein interaction (PPI) network was constructed by Cytoscape. RA-related genes were extracted from DisGeNET database, and an overlap between periimplantitis-related genes and these RA-related genes was examined to identify potential cross-talk genes. Gene expression was merged between two datasets, and feature selection was performed by Recursive Feature Elimination (RFE) algorithm. For the feature selection cross-talk genes, support vector machine (SVM) models were constructed. The expression of these feature genes was determined from GSE93272 for RA. Finally, a network including cross-talk genes, related pathways, and transcription factors was constructed. Periimplantitis datasets included 138 common differentially expressed genes (DEGs) including 101 up- and 37 downregulated DEGs. The PPI interwork of periimplantitis comprised 1,818 nodes and 2,517 edges. The RFE method selected six features, i.e., MERTK, CD14, MAPT, CCR1, C3AR1, and FCGR2B, which had the highest prediction. Out of these feature genes, CD14 and FCGR2B were most highly expressed in periimplantitis and RA. The final activated pathway-gene network contained 181 nodes and 360 edges. Nuclear factor (NF) kappa B signaling pathway and osteoclast differentiation were identified as potentially relevant pathways. This current study revealed FCGR2B and CD14 as the most relevant potential cross-talk genes between RA and periimplantitis, which suggests a similarity between RA and periimplantitis and can serve as a theor. basis for future research. The role of the epithelial-mesenchymal transition (EMT) in lung epithelial cells is increasingly being recognized as a key stage in the development of COPD, fibrosis, and lung cancers, which are all highly associated with cigarette smoking and with exposure to second-hand smoke. Using the exposure of human lung cancer epithelial A549 cells and non-cancerous Beas-2B cells to sidestream cigarette smoke extract (CSE) as a model, we studied the protective effects of adipose-derived stem cell-conditioned medium (ADSC-CM) against CSE-induced cell death and EMT. CSE dose-dependently induced cell death, decreased epithelial markers, and increased the expression of mesenchymal markers. Upstream regulator anal. of differentially expressed genes after CSE exposure revealed similar pathways as those observed in typical EMT induced by TGF-β1. CSE-induced cell death was clearly attenuated by ADSC-CM but not by other control media, such as a pass-through fraction of ADSC-CM or A549-CM. ADSC-CM effectively inhibited CSE-induced EMT and was able to reverse the gradual loss of epithelial marker expression associated with TGF-β1 treatment. CSE or TGF-β1 enhanced the speed of A549 migration by 2- to 3-fold, and ADSC-CM was effective in blocking the cell migration induced by either agent. Future work will build on the results of this in vitro study by defining the mol. mechanisms through which ADSC-CM protects lung epithelial cells from EMT induced by toxicants in second-hand smoke. The authors report here the preparation and use of perylene diimide derivative (PDI) with NIR absorbance (around 700 nm) as nanoprobes for tracking mesenchymal stromal cells (MSCs) in mice. Employing an inhouse synthesized star hyperbranched polymer as a stabilizer is the key to the formation of stable PDI nanoparticles with low toxicity and high uptake by the MSCs. The PDI nanoparticles remain within the MSCs as demonstrated by in vitro and in vivo assessments. The PDI-labeled MSCs injected s.c. on the flanks of the mice are clearly visualized with PAI up to 11 days postadministration. Furthermore, bioluminescence imaging of PDI-labeled luciferase-expressing MSCs confirms that the administered cells remain viable for the duration of the experiment These PDI nanoprobes thus have good potential for tracking administered cells in vivo using PAI.
