An efficient methodol. for the selective and controlled oxidation of petroleum waste methylarenes/alkylarenes to aromatic carboxylic acids/ketone using easily recoverable and recyclable binaphthyl stabilized Pt nanoparticles (Pt-BNP) as a catalyst has been developed. The greener oxidant aqueous tert-Bu hydroperoxide (TBHP) and green solvent water have been utilized in this oxidation reaction. The methodol. is well tolerated with different functional groups. The less reactive electron deficient toluenes are also oxidized by Pt-BNP and gave a good yield of corresponding carboxylic acids with a high turnover number (TON) 738-1448. The Pt-BNP catalyst was recovered and reused up to five catalytic cycles. The heterogeneous test suggested that the reaction is catalyzed by heterogeneous Pt-BNP catalysts. Based on control experiments and literature reports, a possible reaction mechanism has been proposed.

The synthesis of all-carbon tetrasubstituted olefins under mild reaction conditions is challenging because of the inevitable issues including significant steric hindrance and the uncontrolled Z/E stereoselectivity. In this paper, authors report the synthesis of all-carbon tetrasubstituted alkenes from readily available carboxylic acids and alkenyl triflates with the synergistic catalysis of cyclo-octa-1,5-diene(tetramethyl-1,4-benzoquinone)nickel and visible light under an air atm., thus avoiding the need for a glovebox or a Schlenk line. A wide range of aromatic carboxylic acids and cyclic and acyclic alkenyl triflates underwent the C-C coupling process smoothly, forming structurally diverse alkenes stereospecifically in moderate to good yields. The practicality of the method is further illustrated by the late-stage modification of complex mols., the one pot synthesis and gram-scale applications. This is an important step towards the valuable utilization of carboxylic acids, and it also simplifies the exptl. operation of metallophotoredox catalysis with moisture sensitive nickel(0) catalysis.

Farnesoid X receptor (FXR), a bile acid-activated nuclear receptor, regulates the metabolism of bile acid and lipids as well as maintains the stability of internal environment. FXR was considered as a therapeutic target of liver disorders, such as drug-induced liver injury, fatty liver and cholestasis. The previous reported FXR partial agonist I was a suitable lead compound in terms of its high potent and low mol. size, while the docking study of compound I suggested a large unoccupied hydrophobic pocket, which might be provided more possibility of structure-activity relationship (SAR) study. In this study, we have performed comprehensive SAR and mol. modeling studies based on lead compound I. All of these efforts resulted in the identification of a novel series of FXR partial agonists. In this series, compound II revealed the best activity and strong interaction with binding pocket of FXR. Moreover, compound II protected mice against acetaminophen-induced hepatotoxicity by the regulation of FXR-related gene expression and improving antioxidant capacity. In summary, these results suggest that compound II is a promising FXR partial agonist suitable for further investigation.

The N-Py-amide substrates are readily prepared from carboxylic acids and PyNH2, and the resulting N-Py-1-arylalkanamine reaction products are easily transformed into the corresponding chlorides by substitution of the HN-Py group with HCl. The 1-aryl-1-chloroalkane products allow substitution and cross-coupling reactions. Therefore, a general protocol for the transformation of carboxylic acids into a variety of functionalities is obtained. The Py-NH2 byproduct can be recycled.
