Species captured included representatives of the following families: Gracillariidae (3 species), Yponomeutidae (1), Oecophoridae (1), Elachistidae (1), Coleophoridae (1), Blastobasidae (1), Gelechiidae (4), Tortricidae (38), Pyralidae (1), Zygaenidae (2), Geometridae (1), Lymantriidae (2), Noctuidae (5). New sex attractants were found for some of the species recorded, e.g. Calybites aurogutella, Phyllonorycter mespilellus, Coleophora palliatella, Teleoides decorella, Oegoconia quadripuncta, Cydia gemmiferana, C. succedana, Eana derivana, Dichrorampha incursana, Spatalistis bifasciana, Zeiraphera isertana, Zygaena scabiosae. Chronic orchard pests such as Cydia pomonella, C. funebrana and Anarsia lineatella were abundant in pheromone traps located on uncultivated habitats. Catches of Apotomis infida, Cnephasia oxyacanthana, Cydia gemmiferana, Eana derivana and Pammene albugiana are remarkable, showing that pheromone traps can be used successfully as a complementary method in faunistics.

(Z)-9-Dodecen-1-yl acetate (I) and (Z)-11-tetradecen-1-yl acetate (II), primary components of grape berry moth pheromone, were applied in vineyards via tie-on dispensers for the entire growing season. Mature grapes (100 g) were blended and extracted with acetone; following the addition of water, the analytes were extracted into hexane which, after evaporation, was adsorbed onto a Florisil Sep-pak. Elution was with 10% acetone in hexane. Chromatog. was on a Supelcowax 10 capillary column with temperature programming from 80 to 130° at 5°/min then 130 to 200 at 4°/min with a hold 9 min. Detection by HP-MSD Model #5970B was in the selective ion mode at 166 m/e and 194 m/e for I and II, resp. Corresponding retention times were 17.7 and 22.7 min. The sensitivity was <5 ppb for both materials at 80% recovery. Insect control by this biotech. approach was good, and it eliminated the use of traditional chem. pesticides against grape berry moth. No residues were detected in grape berries.

Many moth pheromones are composed of mixtures of acetates of long-chain (≥10 carbon) fatty alcs. Moth pheromone precursors such as fatty acids and fatty alcs. can be produced in yeast by the heterologous expression of genes involved in insect pheromone production Acetyltransferases that subsequently catalyze the formation of acetates by transfer of the acetate unit from acetyl-CoA to a fatty alc. have been postulated in pheromone biosynthesis. However, so far no fatty alc. acetyltransferases responsible for the production of straight chain alkyl acetate pheromone components in insects have been identified. In search for a non-insect acetyltransferase alternative, we expressed a plant-derived diacylglycerol acetyltransferase (EaDAcT) (EC 2.3.1.20) cloned from the seed of the burning bush (Euonymus alatus) in a yeast system. EaDAcT transformed various fatty alc. insect pheromone precursors into acetates but we also found high background acetylation activities. Only one enzyme in yeast was shown to be responsible for the majority of that background activity, the acetyltransferase ATF1 (EC 2.3.1.84). We further investigated the usefulness of ATF1 for the conversion of moth pheromone alcs. into acetates in comparison with EaDAcT. Overexpression of ATF1 revealed that it was capable of acetylating these fatty alcs. with chain lengths from 10 to 18 carbons with up to 27- and 10-fold higher in vivo and in vitro efficiency, resp., compared to EaDAcT. The ATF1 enzyme thus has the potential to serve as the missing enzyme in the reconstruction of the biosynthetic pathway of insect acetate pheromones from precursor fatty acids in yeast.

This multidimensional anal. revealed that the prevalent chem. frame of noctuid moth pheromones was a monosatd. acetate with Z stereochem. and a double bond on the 5th carbon closest to the nonfunctional branch of the mol. Possible phylogenetic relationships within Noctuidae and between Noctuidae and other families are discussed in light of the sex pheromone biochem. Female sex pheromones appeared to be an addnl. character to be considered in the classification of noctuid moths.
