In this work, we employ mol. dynamics simulations to model polyester-based urethane thermosets that are specifically targeted for coating applications. Parameterizing force field interactions with a statistical associating fluid theory (SAFT)-γ Mie approach in conjunction with corresponding state correlations; permits the facile development of effective models for our thermosetting system. We have devised a theor. model to fit exptl. kinetic data and implement a crosslinking algorithm that replicates the theor. kinetics. Our mol. simulations capture the cure kinetics regarding the reactions of the isocyanate group with the primary and secondary hydroxyl termini. Anal. of mol.-level connections that arise during crosslinking affords new information about network structure development. Predicted glass-transition temperatures and thermomech. properties agree well with exptl. data.

Little is known about the effects of inoculation with beneficial microorganisms under diversified crop rotations on agronomic performance of the high-value vegetable crop tomato (Solanum lycopersicum L.). A greenhouse experiment was conducted to evaluate the interactive effect of crop precession with two native species, faba bean (Leguminosae) and barley (Graminaceae), and inoculations with com. arbuscular mycorrhizal fungi (AMF), and phosphate solubilizing Streptomyces roseocinereus MS1B15 on tomato plant growth, yield, fruit quality and the contents of various mineral nutrients. The overall results revealed that crop inoculation with S. roseocinereus MS1B15 significantly increased tomato growth and yield while crop precession showed no differences among tested treatments. Plant nutrient assimilation was significantly increased by both inoculations and was differentially modulated under faba bean and barley rotations. Furthermore, fruit quality demonstrated significant variations between treatments: whereas the inoculations significantly increased tomato vitamin C, crop rotations increased antioxidant compounds and organic acids. Overall, faba bean rotation combined with S. roseocinereus MS1B15 inoculation showed the best benefits for tomato plants, increasing most of the biometric variables evaluated in this study. Such combinations could be recommended as a sustainable and feasible model for tomato production

A completely randomized design with e planting altitudes, five fermentation processes, and five repetitions was performed. Lipids, trigonelline, citrate, and malate were the compounds that most contribute to the chem. discrimination of coffee in the altitudes below 969 m. While, in the high altitudes (> 1000 m), this discrimination was due to the HMF, quinic acid, caffeine, and formic acid, and the global notes of coffee beverages were higher than 80 points. In fermented coffee, the LDA of the chem. data indicates the formation of five clusters, showing how the compounds can suffer changes depending on the form of processing used in coffee. The best score of beverage was observed in samples of 1078.08 m under dry fermentation and only in samples of 969 m was observed a significant difference in the sensory score between spontaneous fermentation and induced fermentation Thus, coffee sensory scores were dependent on planting and fermentation methods and NMR and LDA techniques proved important in chem. and sensory discrimination of coffees.

Fluorescein is a fluorescent dye used as a diagnostic tool in various fields of medicine. Although fluorescein itself possesses low toxicity, after photoactivation, it releases potentially toxic mols., such as singlet oxygen (1O2) and, as we demonstrate in this work, also carbon monoxide (CO). As both of these mols. can affect physiol. processes, the main aim of this study was to explore the potential biol. impacts of fluorescein photochem. In our in vitro study in a human hepatoblastoma HepG2 cell line, we explored the possible effects on cell viability, cellular energy metabolism, and the cell cycle. We observed markedly lowered cell viability (&30%, 75-2400μM) upon irradiation of intracellular fluorescein and proved that this decrease in viability was dependent on the cellular oxygen concentration We also detected a significantly decreased concentration of Krebs cycle metabolites (lactate and citrate < 30%; 2-hydroxyglutarate and 2-oxoglutarate < 10%) as well as cell cycle arrest (decrease in the G2 phase of 18%). These observations suggest that this photochem. reaction could have important biol. consequences and may account for some adverse reactions observed in fluorescein-treated patients. A number of (E)-2-styrylthiophenes, exhibiting a range of electronic effects, and (E)-2-styrylpyrrole were synthesized and their electrochem. behavior investigated. The films, produced by anodic electropolymerization, were redox inactive and exhibited conductivities of the order of 10-6 S cm-1. The nature of the films was investigated by electrochem., spectroscopic and microscopic techniques. The low conductivities exhibited by these films may be attributed to oxidation of the alkene spacer linkage resulting in a crosslinked polymeric matrix. Solid-state NMR, FTIR and photoacoustic-IR confirmed the presence of saturated carbon atoms which would inevitably lead to a decrease in conjugation length. However, in all cases, stable coherent films were obtained from the electropolymerization reaction and an exceptionally smooth film of 20 μm in thickness could be grown from (E)-2-styrylpyrrole on ITO glass.

BACKGROUND: The incidence of osteoporosis and osteoporotic fractures is increasing every year. Traditional Chinese Medicine (TCM) can shed new light on the treatment of osteoporosis. This study aimed to explore the role and mechanism of paeoniflorin in promoting osteogenic differentiation of an osteoblast precursor cell line (MC3T3-E1). METHODS: MC3T3-E1 cells were cultured in osteogenic induction medium (OIM) and OIM combined with different concentrations of paeoniflorin. The optimal dose of paeoniflorin was assessed by a cell counting kit-8 (CCK-8) assay. Then, alkaline phosphatase (ALP) and Alizarin Red S (ARS) staining were performed to assess the osteogenic capacity of paeoniflorin. The transcription of osteogenic genes and the expression of osteogenic proteins were assessed by RT-PCR and Western blotting, respectively. The transcription of Wnt/β-catenin signaling pathway genes and proteins was assessed by RT-PCR and Western blotting, respectively. Finally, Dickkopf-1 (DKK-1), a Wnt/β-catenin signaling pathway inhibitor, was used to identify whether the Wnt/β-catenin signaling pathway was involved in the osteogenic differentiation of paeoniflorin. Osteoclastogenesis in RAW264.7 cells was identified by tartrate-resistant acid phosphatase (TRAP) staining. RESULTS: At concentrations ranging from 0.1 to 100 μM, paeoniflorin was not cytotoxic to MC3T3-E1 cells. Paeoniflorin significantly increased the osteogenic differentiation of MC3T3-E1 cells in a dose-dependent manner. Moreover, paeoniflorin significantly increased osteogenic differentiation gene and protein expression. Through bioinformatic analysis, paeoniflorin-affected genes were found to be involved in different signaling pathways, such as the Wnt/β-catenin signaling pathway. Paeoniflorin enhanced β-catenin and CyclinD1 expression compared with that of the control groups. DKK-1 partially reversed the promoting effects of paeoniflorin in promoting osteogenic differentiation of MC3T3-E1 cells. Moreover, paeoniflorin inhibited the osteoclastogenesis of RAW264.7 cells. CONCLUSION: Paeoniflorin promotes osteogenic differentiation in MC3T3-E1 cells by regulating the Wnt/β-catenin pathway. Paeoniflorin is a potential therapeutic agent for the treatment of osteoporosis.
