As a first step, risk assessment was performed to select the critical method parameters (CMPs) which were decided as organic mobile phase ratio and column oven temperature In order to evaluate the effect of the CMPs on critical method attributes (CMAs) of peak resolution and tailing, central composite design (CCD) was employed. A wide range of natural purine analogs was used as probe to assess the mechanism of recognition by the wild-type (WT) E. coli purine nucleoside phosphorylase (PNP) vs. its Ser90Ala mutant. The results were analyzed from viewpoint of the role of the Ser90 residue and the structural features of the bases. It was found that the Ser90 residue of the PNP (1) plays an important role in the binding and activation of 8-aza-7-deazapurines in the synthesis of their nucleosides, (2) participates in the binding of α-D-pentofuranose-1-phosphates at the catalytic site of the PNP, and (3) catalyzes the dephosphorylation of intermediary formed 2-deoxy-α-D-ribofuranose-1-phosphate in the trans-2-deoxyribosylation reaction. 5-Aza-7-deazaguanine manifested excellent substrate activity for both enzymes, 8-amino-7-thiaguanine and 2-aminobenzothiazole showed no substrate activity for both enzymes. On the contrary, the 2-amino derivatives of benzimidazole and benzoxazole are substrates and are converted into the N1- and unusual N2-glycosides, resp. 9-Deaza-5-iodoxanthine showed moderate inhibitory activity of the WT E. coli PNP, whereas 9-deazaxanthine and its 2′-deoxyriboside are weak inhibitors.

An innovative alternative to the synthesis of non-isocyanate polyurethanes (NIPUs) thermoset materials was thoroughly investigated. This pathway, involving the reaction between primary carbamate functions and aldehyde functions, was studied using model reactions between Et carbamate and different aldehydes that lead to the highlights of the key parameters as well as the first reported proposed mechanism for this reaction. In a second time, polycarbamates synthesized from two different polyols were synthesized via transcarbamoylation before participating in a polycondensation reaction with different monoaldehydes. First a linear NIPU oligomer was synthesized using a dicarbamate derived from ethylene glycol and benzaldehyde. This product was analyzed by NMR spectroscopy and MALDI-TOF mass spectrometry. This oligomer exhibited low molar masses attributed to many secondary termination reactions, mainly due to the presence of acetic anhydride used as solvent in the reaction. Crosslinked NIPU materials were then synthesized without solvent, from the reaction between a tricarbamate derived from trimethylol propane and vanillin, a biobased monoaldehyde, for the first time. The process parameters for the synthesis of these materials were optimized to obtain biobased NIPU thermosetting networks with gel fraction (GF) in dimethylsulfoxide (DMSO) up to 82%, with Tg values ranging between 39°C and 65°C depending on the GF value. This study brings a much needed highlight on the little-known carbamate/aldehyde reaction and demonstrates its potential for the synthesis of novel biobased NIPU thermosetting coatings using a simple monoaldehyde such as vanillin.

This study aims to investigate the thermal conductivity, viscosity and thermal degradation of naphthenic-based mineral oil, palm oil Me ester (POME) and its blend. Mineral oil and POME were mixed at various mass ratios (70:30, 80:20 and 90:10) via homogenisation for 10 min at 25,000 RPM. The thermo-phys. properties of oil studied were thermal conductivity, viscosity, d., flash point and thermal degradation Results indicated enhancement in thermal conductivity with an increase in temperature The highest enhancement (0.185 W mK-1) was observed for the 70:30 blend ratio at 80°C. The viscosity decreased as temperature increased, whereby POME resulted in lower values (2.3 mPa.s) than mineral oil (4.3 mPa.s) at 80°C. D. also showed a decreasing trend with temperature with no significant difference between POME and mineral oil. However, POME (175.5°C) resulted in a higher flash point compared to mineral oil (146.5°C), resulting in the enhanced flashpoint of blend oils. Blend oil with a 70:30 ratio resulted in the highest thermal degradation of 274.56°C in the air environment. Oxidation induction time (OIT) decreased with temperature from 160-180°C where mineral oil resulted in higher induction time than POME. The results proved improvement in properties of mineral oil blend with POME, which can enhance the system performance and faster heat dissipation.

Rodent tuber (Typhonium flagelliforme) as an herbal medicine has a play role to against cancer disease. This study aims to identify the bioactive compounds and to evaluate the anticancer activity on MCF-7 cancer cells of rodent tuber mutant plant (KB 6-1-2) extract and nanoemulsion formula in vitro. Nanoemulsion of rodent tuber mutant plant was developed with a 750 rpm homogenization technique for 30 min. The nanoemulsion were formulated with various surfactants and concentrations i.e. tween 80 (0.1%, 0.5%) and glycerol (0%, 2.5%, 5%, 10%). The best characteristic formula result was shown as F22, which has a particle size of 30.12 nm, polydispersity index of 0.51, and zeta potential of -16.20 mV. The results of the anticancer activity test have an IC50 value of rodent tuber mutant plant extract was 2.35μg mL-1, and the F22 has an IC50 value of 1.89μg mL-1 against MCF-7 breast cancer cells. The GC-MS anal. was showing that the extract and F22 have an anticancer compound as hexadecanoic acid (22.8%) and n-hexadecanoic acid (1.68%), resp. The results revealed that nanoemulsion F22 is more effective at inhibiting the growth of MCF-7 cancer cells than the extract only. F22 can be used as a candidate formula for application to functional drinks.

In this study, a methodol. is proposed: it combines dynamic simulation with ASPEN PLUS DYNAMICS 9.0 software tool, lessons learnt from accidents and risk anal. The primary focus of this paper is on risks occurring in distillation columns, unit operation involved in the case study. The case study concerns a continuous process of propylene glycol (C3H8O2) production The synthesis unit involves three consecutive competitive reactions leading to the formation of byproducts: di and tri glycols (C6H14O3 and C9H20O4 resp.). The process is composed of a CSTR reactor and two distillation columns in series (C1 and C2). Two deviation scenarios on C1 reboiler, chosen according lessons learnt from previous accidents, are simulated and the consequences on C1 and C2 operation analyzed. Overpressure, flooding and weeping occurrence are put in evidence and quantified. This work provides a new perspective on risk quantification through the anal. of deviation propagation along a process involving several unit operations.

It is well-established that Extended-spectrum beta-lactamase-producing (ESBL-) Escherichia coli challenge reliable detection of campylobacters during enrichment in Bolton broth (BB) following ISO 10272-1:2017. The overgrowth of Campylobacter by ESBL-E. coli in the enrichment medium BB can lead to false-neg. detection outcomes, but the cause for the growth suppression is yet unknown. A plausible reason could be the competition-induced lack of certain growth substrates. Therefore, this study aimed to investigate whether campylobacters and ESBL-E. coli compete for the same medium components and whether this is the cause for the observed growth repression. The availability of possible growth substrates in BB was determined and changes in their extracellular concentration were measured over time during mono-culture enrichment of C. jejuni, C. coli or ESBL-E. coli as well as in co-culture enrichments of campylobacters and ESBL-E. coli. Comparative anal. showed lactate and fumarate utilization by C. jejuni and C. coli exclusively, whereas ESBL-E. coli rapidly consumed asparagine, glutamine/arginine, lysine, threonine, tryptophan, pyruvate, glycerol, cellobiose, and glucose. Both campylobacters and ESBL-E. coli utilized aspartate, serine, formate, a-ketoglutarate and malate. Trends in compound utilization were similar for C. jejuni and C. coli and trends in compound utilization were rather comparable during enrichment of reference and freeze-stressed campylobacters. Since final cell densities of C. jejuni and C. coli in co-cultures were not enhanced by the addition of surplus L-serine and final cell densities were similar in fresh and spent medium, growth suppression seems not to be caused by a lack of substrates or production of inhibitory compounds We hypothesized that oxygen availability was limiting growth in co-cultures. Higher oxygen availability increased the competitive fitness of C. jejuni 81-176 in co-culture with ESBL-E. coli in duplicate experiments, as cell concentrations in stationary phase were similar to those without competition. This could indicate the critical role of oxygen availability during the growth of Campylobacter and offers potential for further improvement of Campylobacter spp. enrichment efficacy.
